Beyond traditional screening: Unveiling antibiotic potentials of actinomycetes in extreme environments

Extreme ecosystems are a rich source of specialized metabolites that can overcome multidrug resistance. However, the low efficiency of traditional exploratory research in discovering new antibiotics remains a major limitation. We hypothesized that actinomycetes may have the ability to produce antibiotics in the extremes of a changing natural environment. This study introduces a novel approach to screening natural antibiotic producers from extreme habitats based on the relationship between organisms' adaptive traits and their metabolic activities. The antibacterial and antifungal properties of 667 actinomycete isolates, obtained from 160 samples of Kazakhstan's diverse extreme habitats, were studied under neutral, saline, and alkaline conditions against MRSA, E. coli, C. albicans, and A. niger. Among these isolates, 113 exhibited antibacterial properties, and 109 demonstrated antifungal properties. Notably, one-fifth of the antagonist isolates could produce active substances solely under extreme growth conditions. Fifty-three antagonistic actinomycetes, possessing these characteristics, have been categorized into groups and warrant further investigation as potential producers of new natural antibiotics. Molecular genetic analysis of the selected isolates revealed a high prevalence of Streptomyces and Nocardiopsis strains. Furthermore, 83.4 % of obtained isolates demonstrated the ability to thrive in all studied habitats—neutral, saline, and alkaline. 96.3 % of actinomycetes isolated from extreme environments exhibited adaptation to neutral conditions, highlighting their inherent versatility. Our findings underscore the nearly complete potential (99.7 %) of isolates to overcome the salinity barrier of 3.5 % NaCl, indicating their capacity to inhabit oceanic environments. We assert that actinomycetes should be perceived as a cohesive, globally adaptive group, capable of migrating between changing conditions or remaining stable within them. Discovering an aid policy to minimize student evasion using offline reinforcement learning

High dropout rates in tertiary education expose a lack of efficiency that causes frustration of expectations and financial waste. Predicting students at risk is not enough to avoid student dropout. Usually, an appropriate aid action must be discovered and applied in the proper time for each student. To tackle this sequential decision-making problem, we propose a decision support method to the selection of aid actions for students using offline reinforcement learning to support decision-makers effectively avoid student dropout. Additionally, a discretization of student's state space applying two different clustering methods is evaluated. Our experiments using logged data of real students shows, through off-policy evaluation, that the method should achieve roughly 1.0 to 1.5 times as much cumulative reward as the logged policy. So, it is feasible to help decision-makers apply appropriate aid actions and, possibly, reduce student dropout. © 2021, CC BY.

Enhanced angiotensin-converting enzyme inhibition and antioxidant activities through synergistic green tea and black rice extracts

The study investigated the synergistic effects of green tea extract (GTE) and black rice extract (BRE) on total phenolic content, flavonoid content, antioxidant activities and angiotensin-converting enzyme (ACE) inhibition. The influence of GTE concentrations (14.64–85.36 % w/w) and pH levels (3.59–6.41) was analyzed by regression and response surface methodology. Optimized conditions of synergistic effect (85.36 % w/w GTE and 14.64 % of BRE at pH 6.0) yielded the highest antioxidant activities (DPPH IC₅₀ = 0.112±0.015 mg/mL, ABTS = 89.14±1.58 mmol TE/100 g, FRAP = 373.66±3.42 mmol Fe²⁺/100 g), the highest total phenolic (120.98±1.60 mmol GAE/100 g), flavonoid contents (15.28±1.07 mmol QE/100 g) and ACE inhibitory activity (IC₅₀ = 6.125±0.106 μg/mL). Therefore, this condition was selected to produce the combined extract for determining the combination index (CI). The CI was 0.349±0.075 for DPPH and 0.505±0.019 for ACE inhibition, indicating confirmation of a synergistic effect. These findings suggest that the combined extract could serve as a valuable functional ingredient, enhancing access to bioactive-rich resources for nutritionally supportive and sustainable diets. With potential applications in food systems and healthcare, this agro-ingredient may contribute to improved access to resources for blood pressure management and non-communicable disease (NCDs) prevention, aligning with broader goals in nutrition and health equity. © 2025

Samosir Island is one of tourism destination in Indonesia and there was an increasing of coffee shop number. This research tries to investigate how much financial profit received by coffee shop if it use biogas as a new energy source. This research had been conducted from April to June 2018. Research parameter were production costs, income, revenue, Benefit Cost Ratio, Revenue Cost Ratio, Return On Investment, Break Even Point, and identify opportunities, threats, strengths and weaknesses by SWOT analysis to identify whether the community of Samosir is willing to use biogas. The results of financial analysis namely production cost IDR 1,964,500;income of IDR.4,360,000;revenue of IDR. 2,303,000;B/C (%) was 79.7;R/C (%) was 151;ROI (year) of 1.6;BEP Price was IDR. 5,302;BEP production of 361 glass. By SWOT Analysis it was known that development strategies are taken place include socialize biogas technology, benefits of biogas on energy, fertilizer and soil quality to the community, benefits of biogas in saving fuel purchases and providing biogas stock devices on biogas group. The result showed that biogas was worthy of being used as a new energy source on the coffee shop business on Samosir Island. © Published under licence by IOP Publishing Ltd.

Hoshin Kanri and A3: a proposal for integrating variability into the policy deployment process

Purpose: The purpose of this paper is to propose a methodology to support the Hoshin Kanri (HK) process taking into account the variance of senior managers’ perspective regarding the relationships among objectives, key performance indicators and continuous improvement (CI) projects. Design/methodology/approach: A four-step methodology incorporates the A3 report into the HK, whose deployment was weighted by the variability among managers’ perception about the importance of each decision. This methodology was applied in a market-leader Mexican auto parts manufacturing company that is undergoing a Lean Manufacturing (LM) implementation. Findings: The results indicate that the proposed methodology provides an easier approach to consolidate different perspectives and weight the importance of CI projects. Moreover, it minimizes the possibility of managerial conflicts or the influence of a determined senior manager on others’ opinions during the deployment and prioritization. Practical implications: This work may be of great interest to managers, consultants and professionals who deal with the implementation of LM and wish to prioritize the importance of CI projects within the perspective of HK. Originality/value: HK provides a systematic approach to integrate strategic management with daily routine management through an adequate deployment of organizational policies to all levels. However, companies usually struggle with several issues such as the style of senior managers and consensus establishment among different opinions, which emphasizes the relevance of the proposed methodology in this study. © 2018, Emerald Publishing Limited.

In the last decade, evidence-based medical practice has been supported on a large scale by computerized decision support tools, aiming to reduce diagnostic and therapeutic uncertainty, complementing the actions of the health professional. With technological developments, it is now possible to consider these systems as part of clinical intervention, both for the diagnosis and treatment of diseases. The literature has described the implementation of e-health tools, that is, technological innovations in the health area such as software, applications, serious games, among others, as a strategy to improve the process and adherence to treatment. However, there is still no standardized instrument in Brazil that can be used to guide the development, from the research phase, and the implementation of these tools as a health intervention, also impacting patient outcomes. With the objective of investigating a new therapeutic and preventive form, based on intervention with a computerized system, this work proposes the creation of guidelines for the registration and implementation of e-health tools as a clinical intervention. The proposal aims to be able to assist in the reporting standardization from the development stage to the application of the e-health tool helping in the treatment of diseases, registering all the experience lived in the research and applying it in different contexts of health.

Manufacturing systems’ efficiency depends on the proper assignment of orders to resources. Due to existing interdependencies, the integrated consideration of production, inventory and delivery processes can improve the overall manufacturing performance. However, the integration can result in high complexity and stochasticity. Thus, the three areas are rarely addressed together. Thereof, this paper proposes an integrated simulation-based optimization method to cope with uncertainty and complexity. The proposed approach was compared to a benchmark approach and the obtained results show that the first is able to handle the complexity and stochasticity of real-world manufacturing systems, surpassing the performance of the latter.

The studies of bioethanol production are being intensively explored over the past years in the provision of biofuel. This has been long recognized as the best alternative for a renewable energy resource to overcome the problem of the depletion of fossil resources. Indonesia's main agricultural sector, sugarcane industry, has shown promise in supplying sugarcane bagasse (SCB) as a feedstock for the second generation of bioethanol production. In this study, the techno-economic analysis of the sugarcane bagasse from the bioethanol production plant was explored, and the viability of bioethanol production based on the biomass generated from a local sugarcane factory was evaluated. All the costs that will be incurred during a 20-year operating period for a 203 ktons bioethanol production plant from SCB is $231 million, with a payback period of 6.19 years. The potential amount of bioethanol produced from SCB of this plant is 30.19 ML/year with the production cost (levelized cost of bioethanol, LCOB) of $0.59 per liter of ethanol produced. Overall, SCB bioethanol still carries better competitiveness at lower selling price in comparison with the latest price regulation by the Indonesian government ($0.85/L). © 2023, The Authors. Anchorene,identified as an endogenous bioactive carotenoid-derived dialdehyde and diapocarotenoid,affects root development by modulating auxin ***,the precise interaction between anchorene and auxin,as well as the mechanisms by which anchorene modulates auxin levels,remain largely *** this study,we conducted a comparative analysis of anchorene's bioactivities alongside auxin and observed that anchorene induces multifaceted auxin-like *** genetic and pharmacological examinations,we revealed that anchorene's auxin-like activities depend on the indole-3-pyruvate-dependent auxin biosynthesis pathway,as well as the auxin inactivation pathway mediated by Group II Gretchen Hagen 3(GH3)proteins that mainly facilitate the conjugation of indole-3-acetic acid(IAA)to amino acids,leading to the formation of inactivated storage *** measurements indicated that anchorene treatment elevates IAA levels while reducing the quantities of inactivated IAA–amino acid conjugates and ox *** sequencing further revealed that anchorene triggers the expression of numerous auxin-responsive genes in a manner reliant on Group II ***,our in vitro enzymatic assays and biolayer interferometry(BLI)assay demonstrated anchorene's robust suppression of GH3.17-mediated IAA conjugation with ***,our findings highlight the significant role of carotenoid-derived metabolite anchorene in modulating auxin homeostasis,primarily through the repression of GH3-mediated IAA conjugation and inactivation pathways,offering novel insights into the regulatory mechanisms of plant bioactive apocarotenoids.

In its competitive form, CrossFit® intends on assessing the performance of athletes in a wide variety of aspects that determine their conditioning. CrossFit Games is an official competition and intends to recognize the best conditioned athlete in the world to each class. Thus, measuring an athlete’s Physical Conditioning is, in a sense, assigning a set of performance outputs that can determine the efficiency and efficacy of the athlete, discriminating the performance of one or more athletes. Since the scores obtained by the athletes in the various workouts are directly identifiable, the conditioning can be seen through the performance in a competition, it is, therefore, a result of the interpretation and scope of the workouts in measuring the performance. This work analyzed data form "CrossFit Open" and has as an objective to propose a new theoretical arrangement to the sport discipline while being of the Item Response Theory which is capable of providing data such as, discriminatory capacity and difficulty level of the workouts, as well as, an assessment of the competition. In other words, intends to describe the probability of an athlete performing a workout and obtaining a score, given his/her physical conditioning. Analysis of the main indications that refer to a good quality of the measurement tool indicates it is a high quality competition. Lastly, this work accomplished both objectives proposed, methodological as well as practical, and recognizes the limitations derived from the reduced amount of qualitative data on the topic and the little use of applied probability models. © 2022, CC BY.
