This review presents the significance and current challenges associated with fossil fuels, highlighting the need for cleaner and more sustainable energy sources. The significant object of the current study is to concentrate on increased interest in utilizing biogas as a viable substitute for traditional fuels, delving into its characteristics, benefits, accessibility, feasibility, and prospects as an environmentally sustainable energy option. The current study extensively analyses the prospect and significance of biogas in mitigating emissions regulations in Internal Combustion Engines (ICEs). This paper provides a comprehensive overview of recent research on incorporating biogas in internal combustion engines, focusing on emission reduction and the corresponding impact on engine performance. The subject matter encompasses a range of facets about biogas, encompassing its origins, chemical composition, purification, enhancement techniques, and requisite engine adjustments to enable the effective utilization of biogas as an alternative fuel. The study also evaluates the performance of biogas-fuelled ICEs based on the results of cross-referencing with contemporary research and investigates peripheral aspects such as additive inclusion to increase biogas capabilities as fuel and potential applications in advanced engine modes of operation. The review identifies research gaps in the domains of biogas processing and utilization in ICEs and acknowledges existing challenges specific to the adoption of biogas as a conventional fuel alternative. Future research directions are also prescribed, further highlighting the importance of transitioning from fossil fuels to meet environmental goals and global energy demands. © 2024 The Institution of Chemical Engineers

As the cost of genetic testing becomes more affordable each year, direct-to-customer (DTC) genetic testing services witness rapid market growth. This has encouraged the development of an easy-to-use website application to optimize potential customers to obtain informed choice regarding the offered DTC genetic testing as well as purchasing information. We present a wireframing process as a part of the agile software development process to build a web portal prototype for an Indonesia-based genetic testing service called DNAku. The approach in building the prototype used in this case is a part of the Agile Software Development Method. In brief, the wireframing stage resulted in this work consists of three main important outcomes. The first one is the use case diagram as a blueprint of this web-based system. A site map is then proposed as an extended version of the use case diagram which visualizes the whole pages with the links. Finally, based on this site map a series of page mock-ups are designed using Hyper Text Marking Language (HTML) and Cascading Style Sheets (CSS). The result of the prototype can be used as the basis for the next development stage, which is the coding stage based on the collected feedback from users. Considering the continuous application development via the wireframing method by taking into account the cycle of feedbacks from involved parties, this approach can strengthen the infrastructure to sustain DTC genetic business model, which has a niche market in Indonesia.

Clinical imaging trials play a crucial role in advancing medical innovation but are often costly, inefficient, and ethically constrained. Virtual Imaging Trials (VITs) present a solution by simulating clinical trial components in a controlled, risk-free environment. The Virtual Lung Screening Trial (VLST), an in silico study inspired by the National Lung Screening Trial (NLST), illustrates the potential of VITs to expedite clinical trials, minimize risks to participants, and promote optimal use of imaging technologies in healthcare. This study aimed to show that a virtual imaging trial platform could investigate some key elements of a major clinical trial, specifically the NLST, which compared Computed tomography (CT) and chest radiography (CXR) for lung cancer screening. With simulated cancerous lung nodules, a virtual patient cohort of 294 subjects was created using XCAT human models. Each virtual patient underwent both CT and CXR imaging, with deep learning models, the AI CT-Reader and AI CXR-Reader, acting as virtual readers to perform recall patients with suspicion of lung cancer. The primary outcome was the difference in diagnostic performance between CT and CXR, measured by the Area Under the Curve (AUC). The AI CT-Reader showed superior diagnostic accuracy, achieving an AUC of 0.92 (95% CI: 0.90-0.95) compared to the AI CXR-Reader’s AUC of 0.72 (95% CI: 0.67-0.77). Furthermore, at the same 94% CT sensitivity reported by the NLST, the VLST specificity of 73% was similar to the NLST specificity of 73.4%. This CT performance highlights the potential of VITs to replicate certain aspects of clinical trials effectively, paving the way toward a safe and efficient method for advancing imaging-based diagnostics. © 2024, CC BY-NC-ND.

Differentially methylated imprint control regions (ICRs) regulate the monoallelic expression of imprinted genes. Their epigenetic dysregulation by environmental exposures throughout life results in the formation of common chronic diseases. Unfortunately, existing Infinium methylation arrays lack the ability to profile these regions adequately. Whole genome bisulfite sequencing (WGBS) is the unique method able to profile the ICRs. However, it is very expensive and it requires not only a high coverage, but it is also computationally intensive to assess these regions. To address this deficiency, we developed a custom methylation array containing 22,819 probes. Among them, 10,438 are CG probes targeting unique CpG sites, with 9,757 probes successfully mapping to 1,088 out of the 1,488 candidate ICRs recently described. To assess the performance of the array, we created matched samples processed with the Human Imprintome array and WGBS, which is the current standard method for assessing the methylation of the Human Imprintome. We compared the methylation levels from the shared CpG sites, and obtained a mean R2 = 0.569. We also created matched samples processed with the Human Imprintome array and the Infinium Methylation EPIC v2 array, and obtained a mean R2 = 0.796. Furthermore, replication experiments demonstrated high reliability (ICC: 0.799–0.945). Our custom array will be useful for replicable and accurate assessment, mechanistic insight, and targeted investigation of ICRs. This tool should accelerate the discovery of ICRs associated with a wide range of diseases and exposures, and advance our understanding of genomic imprinting and its relevance in development and disease formation throughout the life course.

Support Vector Regression (SVR) is often used in forecasting. Adjustment of parameters in the SVR affects the results of forecasting. This study aims to analyze the SVR method that is optimized using Harris Hawks Optimization (HHO), hereinafter referred to as HHO-SVR. The HHO-SVR was evaluated using five benchmark datasets to determine the performance of this method. The HHO process is also compared based on the type of kernel and other metaheuristic algorithms. The results showed that the HHO-SVR has almost the same performance as other methods but is less efficient in terms of time. In addition, the type of kernel also affects the process and results.

Pregnant women are regularly exposed to a variety of environmental toxicants in daily life, posing a potential threat of their kidney injury before presence of clinical manifestations. There is a paucity of studies employing an exposome-based approach of kidney health in pregnant women. Thus, this study utilizes the above-mentioned strategy to identify the most significant environmental toxicants associated with early kidney injury in pregnant women in the cohort of TMICS (Taiwan Maternal and Infant Cohort Study). A total of 1,139 third-trimester pregnant women (week 29-40) were recruited between 2012 and 2015 and successfully collected one-spot urine sample for study. Exposure and outcome of interest in urine covered 16 biomonitoring chemicals, including melamine, 9 phthalate metabolites, nonylphenol (NP), bisphenol A (BPA), methylparaben (MP), ethylparaben (EP), propylparaben (PP), and butylparaben (BP), for exposure variables and NAG (N-acetyl-β-D-glucosaminidase) and albumin-to-creatinine ratio (ACR) for outcome variables. A two-tier strategy of statistical analyses was employed and data was randomly and evenly splitted to the training set (n = 569) and the validation set (n = 570). Using a weighted quantile sum (WQS) regression in the training set and subsequently a multivariate regression in the validation set, we found that NP is the most important chemical to link with markers of early kidney injury, both ACR and NAG. Other chemicals, including BPA and MP, are still needed to pay an attention on their potential nephrotoxicity in the pregnant women. The potential applications of fungi in the development of new biomaterials derived from fungal mycelium have captured the attention from both the scientific community and the society. The notable ability of mycelium networks to self-construct and aggregate can be used to produce diverse biomaterials. These biomaterials can be created in a pure state, or both in conjunction with other organic/inorganic compounds. Recent advancements in mycomaterials have gained attention due to their sustainability and mechanical, thermogravimetric, and compression properties. Such properties contribute to reducing the reliance on environmentally problematic substrates within the industry. After a standardized and comprehensive review of publications on mycomaterials across different fields, such as biology, health, agriculture, engineering, and material sciences, we detected that publications on this theme are utterly scattered. This critical review enabled us to also propose a novel classification system for these fungal-derived materials to help to structure and standardize this emerging transdisciplinary field of knowledge. We employed Periodic Density Functional Theory (DFT) to investigate the catalytic activity of single and double dopant of boron (B), nitrogen (N), and phosphorous (P) anchored on diamond surfaces for the CO2 reduction reaction (CO2RR) by analyzing reaction energy profiles. Our findings show that the double-doped catalyst exhibits lower energy barriers in CO2RR compared to the single dopants. Specifically, when considering BB and NN configurations, there is a thermodynamic preference towards the formation of formic acid (HCOOH) with overpotentials of 0.40 and 0.09 V, respectively. The co-doped catalyst comprising B and N (BN) demonstrates a tendency towards the formation of HCOOH without requiring any applied overpotentials. Notably, BN outperforms other catalysts, occupying the top position on the volcano plot, indicating the lowest limiting potential (UL), remarkable thermal stability, and the ability to suppress the competing hydrogen evolution reaction (HER). This research provides valuable insights into the product differentiation in the electroreduction of CO2. © 2023, The Authors. All rights reserved.

Mosaic Mutagenesis In Vivo Reveals Mutant Blood Stem Cells Intrinsically Resistant to Inflammatory Mediators in Clonal Hematopoiesis

Clonal hematopoiesis (CH) is a state of clonal dominance of a mutant hematopoietic stem and progenitor cell (HSPC) promoted by unknown mechanisms. To study the competitive behavior and the mechanism of such expansion in a native environment, we used a combinatorial approach of HSPCs labeling and mutagenesis in vivo in a technique we called T issue editing W ith I nducible S tem cell T agging via R ecombination (TWISTR). TWISTR utilizes Zebrabow zebrafish to fluorescently label endogenous HSPCs, and allows following of the clonal activity and sorting of labeled HSPCs within the hematopoietic system. CRISPR/Cas9 mutagenesis was used to simultaneously induce mosaic insertions/deletions (indels) in zebrafish orthologs of 12 human CH genes during development, and zebrafish were followed for 8 months. As such, mutant stem cells were directly competed against endogenous wildtype stem cells. We achieved a high degree of mosaic mutagenesis with heterozygous edits, with a median of 5 targeted genes per zebrafish in total marrow cells at variant allele frequency (VAF) of 5% or greater. Serial sampling and sequencing of peripheral blood cells showed selective expansion of clones with frameshift indels in exon 12 of asxl1 , while clones with other CH mutations did not show a significant change over the 4 months period evaluated. No changes were noted in control gene indels or in the CH gene indels in non-hematopoietic tissue in the same time period. We identified significant enrichment of frameshift indels in asxl1 in sorted dominant clones compared to sorted smaller clones in the same zebrafish ( p <0.01) at 8 months, while indels in DNMT3A ortholog dnmt8 were present in clones of various competitive nature. Introduction of asxl1 mutations singly in a mosaic fashion also resulted in clonally dominant states.
