Despite substantial declines since 2000, lower respiratory infections (LRIs), diarrhoeal diseases, and malaria remain among the leading causes of nonfatal and fatal disease burden for children under 5 years of age (under 5), primarily in sub-Saharan Africa (SSA). The spatial burden of each of these diseases has been estimated subnationally across SSA, yet no prior analyses have examined the pattern of their combined burden. Here we synthesise subnational estimates of the burden of LRIs, diarrhoea, and malaria in children under-5 from 2000 to 2017 for 43 sub-Saharan countries. Some units faced a relatively equal burden from each of the three diseases, while others had one or two dominant sources of unit-level burden, with no consistent pattern geographically across the entire subcontinent. Using a subnational counterfactual analysis, we show that nearly 300 million DALYs could have been averted since 2000 by raising all units to their national average. Our findings are directly relevant for decision-makers in determining which and targeting where the most appropriate interventions are for increasing child survival. © 2022, The Author(s).

In this review paper, we will present different data-driven dimension reduction techniques for dynamical systems that are based on transfer operator theory as well as methods to approximate transfer operators and their eigenvalues, eigenfunctions, and eigenmodes. The goal is to point out similarities and differences between methods developed independently by the dynamical systems, fluid dynamics, and molecular dynamics communities such as time-lagged independent component analysis (TICA), dynamic mode decomposition (DMD), and their respective generalizations. This paper considers the reconstruction problem in Acousto-Electrical Tomography, i.e., the problem of estimating a spatially varying conductivity in a bounded domain from measurements of the internal power densities resulting from different prescribed boundary conditions. Particular emphasis is placed on the limited angle scenario, in which the boundary conditions are supported only on a part of the boundary. The reconstruction problem is formulated as an optimization problem in a Hilbert space setting and solved using Landweber iteration. The resulting algorithm is implemented numerically in two spatial dimensions and tested on simulated data. The results quantify the intuition that features close to the measurement boundary are stably reconstructed and features further away are less well reconstructed. Associations between the gut microbiome and colorectal cancer (CRC) have been uncovered, but larger and more diverse studies are needed to assess their potential clinical use. We expanded upon 12 metagenomic datasets of patients with CRC (n = 930), adenomas (n = 210) and healthy control individuals (n = 976; total n = 2,116) with 6 new cohorts (n = 1,625) providing granular information on cancer stage and the anatomic location of tumors. We improved CRC prediction accuracy based solely on gut metagenomics (average area under the curve = 0.85) and highlighted the contribution of 19 newly profiled species and distinct Fusobacterium nucleatum clades. Specific gut species distinguish left-sided versus right-sided CRC (area under the curve = 0.66) with an enrichment of oral-typical microbes. We identified strain-specific CRC signatures with the commensal Ruminococcus bicirculans and Faecalibacterium prausnitzii showing subclades associated with late-stage CRC. Our analysis confirms that the microbiome can be a clinical target for CRC screening and characterizes it as a biomarker for CRC progression.

Context. Number counts of galaxy clusters across redshift are a powerful cosmological probe, if a precise and accurate reconstruction of the underlying mass distribution is performed - a challenge called mass calibration. With the advent of wide and deep photometric surveys, weak gravitational lensing by clusters has become the method of choice to perform this measurement. Aims. We measure and validate the weak gravitational lensing (WL) signature in the shape of galaxies observed in the first 3 years of the Dark Energy Survey (DES Y3) caused by galaxy clusters and groups selected in the first all-sky survey performed by SRG (Spectrum Roentgen Gamma)/eROSITA (eRASS1). These data are then used to determine the scaling between X-ray photon count rate of the clusters and their halo mass and redshift. Methods. We empirically determine the degree of cluster member contamination in our background source sample. The individual cluster shear profiles are then analysed with a Bayesian population model that self-consistently accounts for the lens sample selection and contamination, and includes marginalization over a host of instrumental and astrophysical systematics. To quantify the accuracy of the mass extraction of that model, we perform mass measurements on mock cluster catalogs with realistic synthetic shear profiles.
