Context. The BL Lac object 1ES 0647+250 is one of the few distant γ-ray emitting blazars detected at very high energies (VHEs;&100 GeV) during a non-flaring state. It was detected with the MAGIC telescopes during a period of low activity in the years 2009-2011 as well as during three flaring activities in the years 2014, 2019, and 2020, with the highest VHE flux in the last epoch. An extensive multi-instrument data set was collected as part of several coordinated observing campaigns over these years. Aims. We aim to characterise the long-term multi-band flux variability of 1ES 0647+250, as well as its broadband spectral energy distribution (SED) during four distinct activity states selected in four different epochs, in order to constrain the physical parameters of the blazar emission region under certain assumptions. Methods. We evaluated the variability and correlation of the emission in the different energy bands with the fractional variability and the Z-transformed discrete correlation function, as well as its spectral evolution in X-rays and γ rays. Owing to the controversy in the redshift measurements of 1ES 0647+250 reported in the literature, we also estimated its distance in an indirect manner through a comparison of the GeV and TeV spectra from simultaneous observations with Fermi-LAT and MAGIC during the strongest flaring activity detected to date. Moreover, we interpret the SEDs from the four distinct activity states within the framework of one-component and two-component leptonic models, proposing specific scenarios that are able to reproduce the available multi-instrument data. Results. We find significant long-term variability, especially in X-rays and VHE γ rays. Furthermore, significant (3-4σ) correlations were found between the radio, optical, and high-energy (HE) γ-ray fluxes, with the radio emission delayed by about ∼400 days with respect to the optical and γ-ray bands. In recent years, a new generation of optical intensity interferometers has emerged, leveraging the existing infrastructure of Imaging Atmospheric Cherenkov Telescopes (IACTs). The MAGIC telescopes host the MAGIC-SII system (Stellar Intensity Interferometer), implemented to investigate the feasibility and potential of this technique on IACTs. After the first successful measurements in 2019, the system was upgraded and now features a real-time, dead-time-free, 4-channel, GPU-based correlator. These hardware modifications allow seamless transitions between MAGIC’s standard very-high-energy gamma-ray observations and optical interferometry measurements within seconds. We establish the feasibility and potential of employing IACTs as competitive optical Intensity Interferometers with minimal hardware adjustments. The measurement of a total of 22 stellar diameters are reported, 9 corresponding to reference stars with previous comparable measurements, and 13 with no prior measurements. A prospective implementation involving telescopes from the forthcoming Cherenkov Telescope Array Observatory’s northern hemisphere array, such as the first prototype of its Large-Sized Telescopes, LST-1, is technically viable. This integration would significantly enhance the sensitivity of the current system and broaden the UV-plane coverage. This advancement would enable the system to achieve competitive sensitivity with the current generation of long-baseline optical interferometers over blue wavelengths. Copyright © 2024, The Authors. Extreme high-frequency-peaked BL Lac objects (EHBLs) are the most energetic persistent sources in the universe. This contribution reports on long-term observing campaigns of tens of EHBLs that have been organized by the MAGIC collaboration to enlarge their population at VHE and understand the origin of their extreme properties. EHBLs are characterized by a spectral energy distribution (SED) featuring a synchrotron peak energy above 1 keV. Several EHBLs display a hard spectral index at very high energies (VHE;E > 100 GeV), suggesting a gamma-ray SED component peaking significantly above 1 TeV. Such extreme properties are challenging current standard emission and acceleration mechanisms. Recent studies have also unveiled intriguing disparities in the temporal characteristics of EHBLs. Some sources seem to display a persistent EHBL behaviour, while others belong to the EHBL family only temporarily. Here, we present recent results of the first hard-TeV EHBL catalog. The MAGIC observations are accompanied by an extensive multiwavelength coverage to obtain an optimal determination of the SED. This allow us to investigate leptonic and hadronic scenarios for the emission. We also present the recent detection of the EHBL RX J0812.0+0237 in the VHE band by MAGIC. Finally, we discuss a broad multiwavelength campaign on the BL Lac type object 1ES 2344+514, which showed intermittent EHBL characteristics in August 2016. © Copyright owned by the author(s).

The translation of AI-generated brain metastases (BM) segmentation into clinical practice relies heavily on diverse, high-quality annotated medical imaging datasets. The BraTS-METS 2023 challenge has gained momentum for testing and benchmarking algorithms using rigorously annotated internationally compiled real-world datasets. This study presents the results of the segmentation challenge and characterizes the challenging cases that impacted the performance of the winning algorithms. Untreated brain metastases on standard anatomic MRI sequences (T1, T2, FLAIR, T1PG) from eight contributed international datasets were annotated in stepwise method: published UNET algorithms, student, neuroradiologist, final approver neuroradiologist. Segmentations were ranked based on lesion-wise Dice and Hausdorff distance (HD95) scores. False positives (FP) and false negatives (FN) were rigorously penalized, receiving a score of 0 for Dice and a fixed penalty of 374 for HD95. The mean scores for the teams were calculated. Eight datasets comprising 1303 studies were annotated, with 402 studies (3076 lesions) released on Synapse as publicly available datasets to challenge competitors. Additionally, 31 studies (139 lesions) were held out for validation, and 59 studies (218 lesions) were used for testing. Segmentation accuracy was measured as rank across subjects, with the winning team achieving a LesionWise mean score of 7.9. The Dice score for the winning team was 0.65 ± 0.25. Common errors among the leading teams included false negatives for small lesions and misregistration of masks in space. The Dice scores and lesion detection rates of all algorithms diminished with decreasing tumor size, particularly for tumors smaller than 100 mm3. In conclusion, algorithms for BM segmentation require further refinement to balance high sensitivity in lesion detection with the minimization of false positives and negatives. OT 081 is a well-known, luminous blazar that is remarkably variable in many energy bands. We present the first broadband study of the source, which includes very high energy (VHE, E > 100 GeV) γ -ray data taken by the MAGIC (Major Atmospheric Gamma-ray Imaging Cherenkov telescopes) and H.E.S.S. (High Energy Stereoscopic System) imaging Cherenkov telescopes. The discovery of VHE γ -ray emission happened during a high state of γ -ray activity in July 2016, observed by many instruments from radio to VHE γ -rays. We identify four states of activity of the source, one of which includes VHE γ -ray emission. Variability in the VHE domain is found on daily time-scales. The intrinsic VHE spectrum can be described by a power law with index 3.27 ± 0.44stat ± 0.15sys (MAGIC) and 3.39 ± 0.58stat ± 0.64sys (H.E.S.S.) in the energy range of 55–300 and 120–500 GeV, respectively. The broadband emission cannot be successfully reproduced by a simple one-zone synchrotron self-Compton model. Instead, an additional external Compton component is required. We test a lepto-hadronic model that reproduces the data set well and a proton-synchrotron-dominated model that requires an extreme proton luminosity. Emission models that are able to successfully represent the data place the emitting region well outside of the broad-line region to a location at which the radiative environment is dominated by the infrared thermal radiation field of the dusty torus. In the scenario described by this flaring activity, the source appears to be a flat spectrum radio quasar (FSRQ), in contrast with past categorizations. This suggests that the source can be considered to be a transitional blazar, intermediate between BL Lac and FSRQ objects. © 2024 The Author(s).

Epilepsy is a highly heritable disorder affecting over 50 million people worldwide, of which about one-third are resistant to current treatments. Here we report a multi-ancestry genome-wide association study including 29,944 cases, stratified into three broad categories and seven subtypes of epilepsy, and 52,538 controls. We identify 26 genome-wide significant loci, 19 of which are specific to genetic generalized epilepsy (GGE). We implicate 29 likely causal genes underlying these 26 loci. SNP-based heritability analyses show that common variants explain between 39.6% and 90% of genetic risk for GGE and its subtypes. Subtype analysis revealed markedly different genetic architectures between focal and generalized epilepsies. Gene-set analyses of GGE signals implicate synaptic processes in both excitatory and inhibitory neurons in the brain. Prioritized candidate genes overlap with monogenic epilepsy genes and with targets of current antiseizure medications. Finally, we leverage our results to identify alternate drugs with predicted efficacy if repurposed for epilepsy treatment.

In 2023, La Niña conditions that generally prevailed in the eastern Pacific Ocean from mid-2020 into early 2023 gave way to a strong El Niño by October. Atmospheric concentrations of Earth’s major greenhouse gases—carbon dioxide, methane, and nitrous oxide—all increased to record-high levels. The annual global average carbon dioxide concentration in the atmosphere rose to 419.3±0.1 ppm, which is 50% greater than the pre-industrial level. The growth from 2022 to 2023 was 2.8 ppm, the fourth highest in the record since the 1960s. The combined short-term effects of El Niño and the long-term effects of increasing levels of heat-trapping gases in the atmosphere contributed to new records for many essential climate variables reported here. The annual global temperature across land and oceans was the highest in records dating as far back as 1850, with the last seven months (June–December) having each been record warm. Over land, the globally averaged temperature was also record high. Dozens of countries reported record or near-record warmth for the year, including China and continental Europe as a whole (warmest on record), India and Russia (second warmest), and Canada (third warmest). Intense and widespread heatwaves were reported around the world. In Vietnam, an all-time national maximum temperature record of 44.2°C was observed at Tuong Duong on 7 May, surpassing the previous record of 43.4°C at Huong Khe on 20 April 2019. In Brazil, the air temperature reached 44.8°C in Araçuaí in Minas Gerais on 20 November, potentially a new national record and 12.8°C above normal. The effect of rising temperatures was apparent in the cryosphere, where snow cover extent by June 2023 was the smallest in the 56-year record for North America and seventh smallest for the Northern Hemisphere overall. Background: Decades of steady improvements in life expectancy in Europe slowed down from around 2011, well before the COVID-19 pandemic, for reasons which remain disputed. We aimed to assess how changes in risk factors and cause-specific death rates in different European countries related to changes in life expectancy in those countries before and during the COVID-19 pandemic. Methods: We used data and methods from the Global Burden of Diseases, Injuries, and Risk Factors Study 2021 to compare changes in life expectancy at birth, causes of death, and population exposure to risk factors in 16 European Economic Area countries (Austria, Belgium, Denmark, Finland, France, Germany, Greece, Iceland, Ireland, Italy, Luxembourg, the Netherlands, Norway, Portugal, Spain, and Sweden) and the four UK nations (England, Northern Ireland, Scotland, and Wales) for three time periods: 1990–2011, 2011–19, and 2019–21. Changes in life expectancy and causes of death were estimated with an established life expectancy cause-specific decomposition method, and compared with summary exposure values of risk factors for the major causes of death influencing life expectancy. Findings: All countries showed mean annual improvements in life expectancy in both 1990–2011 (overall mean 0·23 years [95% uncertainty interval [UI] 0·23 to 0·24]) and 2011–19 (overall mean 0·15 years [0·13 to 0·16]). The rate of improvement was lower in 2011–19 than in 1990–2011 in all countries except for Norway, where the mean annual increase in life expectancy rose from 0·21 years (95% UI 0·20 to 0·22) in 1990–2011 to 0·23 years (0·21 to 0·26) in 2011–19 (difference of 0·03 years). In other countries, the difference in mean annual improvement between these periods ranged from –0·01 years in Iceland (0·19 years [95% UI 0·16 to 0·21] vs 0·18 years [0·09 to 0·26]), to –0·18 years in England (0·25 years [0·24 to 0·25] vs 0·07 years [0·06 to 0·08]).
