More than half of the world’s population still experiences dental and mouth disease which causes disruption to the quality of human life. The number of untreated dental and oral diseases increased by 40% from 1990 to 2015 which reached 3.5 billion people and periodontal disease in severe conditions reached 573 million. One of the cases of periodontal abnormalities is the Guided tissue regeneration (GTR) method. In this study the formation of GTR membranes derived from biocomposite PCL-AgNPs using the electrospinning method. The results obtained through FTIR characterization, degradation, antibacterial, and cytotoxicity that the addition of AgNPs did not show changes in functional groups. All biocomposite GTR membrane PCL-AgNPs samples showed that AgNPs only physically bonded to PCL. Degradation characterization shows that the addition of AgNPs causes the rate of degradation to increase with increasing number of AgNPs. The addition of AgNPs also shows the ability of membranes to inhibit bacteria and viability decreases with increasing number of AgNPs.

Rare earth elements (REEs) are exploited for global use in manufacturing. Such activities result in their release into the environment and the transformation into more stable phosphate deposition. The objective of this study was to evaluate molecular and behavioral changes of zebrafish exposed to the synthesized terbium phosphate (TbPO4) at concentrations of 10, 20, and 50 mg/L and to determine its potential for neurotoxicity. Metabolomics related to neurotransmitters, and assessment of transcripts and proteins were conducted to uncover the molecular mechanisms underlying TbPO4 with emphasis on neurotransmitter systems. Exposure to 20 mg/L TbPO4 induced larval hyperactivity and perturbed the cholinergic system in zebrafish. Based on metabolomics related to neurotransmitters, dopamine (DA), serotonin (5-HT), and many of their precursors and metabolites were decreased in abundance by TbPO4. In addition, the expression levels of transcripts related to the synthesis, transport, receptor binding, and metabolism of DA and 5-HT were analyzed at the mRNA and protein levels. Transcript and protein levels for tyrosine hydroxylase (TH), the rate-limiting enzyme for DA synthesis, were down-regulated in larval fish. Monoamine oxidase (MAO), an enzyme that catabolizes monoamines DA and 5-HT, was also reduced in mRNA abundance. We hypothesize that DA synthesis and monoamine metabolism are associated with behavioral alterations. This study elucidates putative mechanisms and exposure risks to wildlife and humans by characterizing phosphatic REE-induced neurotoxicity in developing zebrafish. © 2023 Elsevier Ltd

Fair Patient Model: Mitigating Bias in the Patient Representation Learned from the Electronic Health Records

Objective: To pre-train fair and unbiased patient representations from Electronic Health Records (EHRs) using a novel weighted loss function that reduces bias and improves fairness in deep representation learning models. Methods: We defined a new loss function, called weighted loss function, in the deep representation learning model to balance the importance of different groups of patients and features. We applied the proposed model, called Fair Patient Model (FPM), to a sample of 34,739 patients from the MIMIC-III dataset and learned patient representations for four clinical outcome prediction tasks. Results: FPM outperformed the baseline models in terms of three fairness metrics: demographic parity, equality of opportunity difference, and equalized odds ratio. FPM also achieved comparable predictive performance with the baselines, with an average accuracy of 0.7912. Feature analysis revealed that FPM captured more information from clinical features than the baselines. Conclusion: FPM is a novel method to pre-train fair and unbiased patient representations from EHR data using a weighted loss function. The learned representations can be used for various downstream tasks in healthcare and can be extended to other domains where bias and fairness are important. Copyright © 2023, The Authors. Bacterial cellulose composite-collagen-glycerol as an innovation of artificial dura mater candidates for open head trauma case

Head injury is one of the leading causes of death and disability in the productive age. According to the World Health Organization (WHO), 90% of the causes of head injuries in Indonesia are caused by traffic accidents. It causes tearing of the dura mater layer on the head and will be fatal if not treated immediately. The aim of this study is finding out the replacement composites of artificial head dura mater layers which are compatible. In this study, artificial dura mater derived from the bacterial cellulose Acetobacter xylinum which was synthesized on coconut water media. In this study, four variations of composition were used, namely bacterial cellulose (control), bacterial cellulose-glycerol, bacterial cellulose – collagen, and bacterial cellulose-collagen-glycerol. Characterization of the river group (FTIR), tensile strength, swelling and MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) tetrazolium reduction assay. Assay was conducted in this study. Characterization of functional groups shows the functional groups characteristic of bacterial cellulose, collagen, and glycerol. The results of the tensile strength characterization show that of the four variations in sample composition have average UTS ( Ultimate Tensile Strength) and Elongation values that are close to the standard value of artificial dura mater. The best results in the swelling test were obtained on the composition of the bacterial cellulose sample-Collagen-Glycerol with a swelling value of 28.30%. These results did not exceed the membrane swelling ratio limit of 70%. For the MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) tetrazolium reduction assay test, the percentage of living cells from each sample is above 50% which means that the material produced is not toxic when applied to the body. We introduce cytoNet, a cloud-based tool to characterize cell populations from microscopy images. cytoNet quantifies spatial topology and functional relationships in cell communities using principles of network science. Capturing multicellular dynamics through graph features, cytoNet also evaluates the effect of cell-cell interactions on individual cell phenotypes. We demonstrate cytoNet’s capabilities in four case studies: 1) characterizing the temporal dynamics of neural progenitor cell communities during neural differentiation, 2) identifying communities of pain-sensing neurons in vivo , 3) capturing the effect of cell community on endothelial cell morphology, and 4) investigating the effect of laminin α4 on perivascular niches in adipose tissue. The analytical framework introduced here can be used to study the dynamics of complex cell communities in a quantitative manner, leading to a deeper understanding of environmental effects on cellular behavior. The versatile, cloud-based format of cytoNet makes the image analysis framework accessible to researchers across domains.

Colorectal cancer is a leading cause of cancer mortality worldwide, with an increasing incidence rate in developing countries. Integration of genetic information with cancer therapy guidance has shown promise in cancer treatment, indicating its potential as an essential tool in translation oncology. However, the high-throughput analysis and variability of genomic data poses a major challenge to conventional analytic approaches. In this study, we propose an advanced analytic approach, named "Fuzzy-based RNNCoxPH," incorporated fuzzy logic, recurrent neural networks (RNNs), and Cox proportional hazards regression (CoxPH) for detecting missense variants associated with high-risk of all-cause mortality in rectum adenocarcinoma. The test data set was downloaded from "Rectum adenocarcinoma, TCGA-READ" the Genomic Data Commons (GDC) portal. In this study, four model-based risk score models were derived using RNN, CoxPH, RNNCoxPHAddition, and RNNCoxPHMultiplication. The RNNCoxPHAddition and RNNCoxPHMultiplication models were obtained as the sum and product of the RNN risk degree matrix and the CoxPH risk degree matrix, respectively. Moreover, the fuzzy logic system was used to calculate the survival risk values of missense variants and classified their membership grade to improve the identification of high-risk gene variation locations associated with cancer mortality. The four models were integrated to develop an advanced risk estimation model. There were 20 028 variants associated with survival status, amongst 17 638 variants were associated with survival and 2390 variants associated with mortality. The proposed Fuzzy-based RNNCoxPH model obtained a balanced accuracy of 93.7%, which was significantly higher than that of the other four test methods. In particular, the CoxPH model is commonly used in medical researches and the XGBoost model is famous for its high accuracy in machine learning.
