BACKGROUND:The main commercially available methods for detecting small molecules of mycotoxins in traditional Chinese medicine (TCM) and functional foods are enzyme-linked immunosorbent assay and mass spectrometry. Regarding the development of diagnostic antibody reagents, effective methods for the rapid preparation of specific monoclonal antibodies are inadequate. METHODS:In this study, a novel synthetic phage-displayed nanobody Golden Glove (SynaGG) library with a glove-like cavity configuration was established using phage display technology in synthetic biology. We applied this unique SynaGG library on the small molecule aflatoxin B1 (AFB1), which has strong hepatotoxicity, to isolate specific nanobodies with high affinity for AFB1. RESULT:These nanobodies exhibit no cross-reactivity with the hapten methotrexate, which is recognized by the original antibody template. By binding to AFB1, two nanobodies can neutralize AFB1-induced hepatocyte growth inhibition. Using molecular docking, we found that the unique non-hypervariable complementarity-determining region 4 (CDR4) loop region of the nanobody was involved in the interaction with AFB1. Specifically, the CDR4's positively charged amino acid arginine directed the binding interaction between the nanobody and AFB1. We then rationally optimized the interaction between AFB1 and the nanobody by mutating serine at position 2 into valine. The binding affinity of the nanobody to AFB1 was effectively improved, and this result supported the use of molecular structure simulation for antibody optimization. CONCLUSION:In summary, this study revealed that the novel SynaGG library, which was constructed through computer-aided design, can be used to isolate nanobodies that specifically bind to small molecules. The results of this study could facilitate the development of nanobody materials to detect small molecules for the rapid screening of TCM materials and foods in the future.

Ovarian cancer remains the third most prevalent and deadliest gynecologic malignancy worldwide, with most patients eventually developing resistance to platinum-based chemotherapy. This highlights a critical unmet need for innovative multitargeted therapies to address current treatment challenges. In this study, we identified 35 differentially expressed genes (DEGs) through integrated analysis of four GEO ovarian cancer datasets, with validation using TCGA data. Gene Ontology (GO) and KEGG enrichment analyses highlighted key tumor-associated pathways, and protein–protein interaction (PPI) network modeling prioritized CDK1 and TTK as high-value therapeutic targets. We evaluated the association between molecular genomic features and drug responses across the NCI-60 ovarian cancer cell line panel (IGROV1, OVCAR-3, OVCAR-4, OVCAR-5, OVCAR-8, NCI/ADR-RES, and SK-OV-3), using a series of salicylanilide-derived compounds and four FDA-approved drugs (cabozantinib, paclitaxel, rapamycin, and niclosamide) from the NCI developmental Therapeutics program (DTP). Among these, NSC765690 (MCC22) emerged as the most promising candidate. It demonstrated potent antiproliferative activity, high target selectivity, and strong binding affinity to both CDK1 and TTK. Multi-omics integration, combined with AI-driven network modeling, further elucidated NSC765690's mechanism of action and its relevance to ovarian cancer pathogenesis. Additionally, ADMET and pharmacokinetic profiling confirmed its favorable drug-like properties and low predicted toxicity. Collectively, these findings establish NSC765690 as a potent dual-target inhibitor and exemplify a rational, data-driven drug discovery pipeline for overcoming chemotherapy resistance in ovarian cancer. © 2025 The Authors

The nitrogen assimilation pathway in cassava was reconstructed by comparative genomics approach to understand the underlying metabolism as well as the interaction between carbon and nitrogen assimilation towards the synthesis of metabolic phenotype. First, theproteins of cassava were annotated via sequence similarity search against genes of 11 template plants obtained from KEGG and PMN databases, employing reciprocal BLASTp(E-value ≤ 1x10-10, identity percentage ≥ 60, and coverage percentage ≥ 80). The template plants comprised well-known plant, starchy crops, nitrogen-fixing crops and crops that are evolutionarily related to cassava and includedArabidopsis thaliana, Oryzasativa, Zea mays, Ricinuscommunis, Solanumtuberosum, Brassica rapa, Cicerarietinum, Jatrophacurcas, Medicagotruncatula, Phaseolus vulgaris and Glycine *** pathway was then curatedwith reactions obtained from the CassavaCyc database to ensure full pathway *** was subsequently validated with cloned sequence of cassava from the GenBank and cassava transcriptome data from literature. The resulting N-assimilation pathway, covering the conversion of nitrate to amino acids (glutamine and glutamate),consists of 14 biochemical reactions corresponding to 59 genes, 73 proteins and 2 transport reactions. At least 92 percent of the identified proteins in the pathway were supported by the transcriptome data. In addition, the proposed N-assimilation pathway contains four additional enzymes, including glutamate synthase, nitrilase, formamidase and carbamoyl phosphate synthasecompared to the existing N-assimilation pathway in CassavaCyc database. Taken together, the N-assimilation pathway herein proposed identified reactions involved in N-assimilation and represents a forward step towards understanding metabolic basis for cassava yield as well as its phenotypic plasticity and adaptation to stress. © Published under licence by IOP Publishing Ltd.

Background: Diabetes is one of the leading causes of death and disability worldwide, and affects people regardless of country, age group, or sex. Using the most recent evidentiary and analytical framework from the Global Burden of Diseases, Injuries, and Risk Factors Study (GBD), we produced location-specific, age-specific, and sex-specific estimates of diabetes prevalence and burden from 1990 to 2021, the proportion of type 1 and type 2 diabetes in 2021, the proportion of the type 2 diabetes burden attributable to selected risk factors, and projections of diabetes prevalence through 2050. Methods: Estimates of diabetes prevalence and burden were computed in 204 countries and territories, across 25 age groups, for males and females separately and combined;these estimates comprised lost years of healthy life, measured in disability-adjusted life-years (DALYs;defined as the sum of years of life lost [YLLs] and years lived with disability [YLDs]). We used the Cause of Death Ensemble model (CODEm) approach to estimate deaths due to diabetes, incorporating 25 666 location-years of data from vital registration and verbal autopsy reports in separate total (including both type 1 and type 2 diabetes) and type-specific models. Other forms of diabetes, including gestational and monogenic diabetes, were not explicitly modelled. Total and type 1 diabetes prevalence was estimated by use of a Bayesian meta-regression modelling tool, DisMod-MR 2.1, to analyse 1527 location-years of data from the scientific literature, survey microdata, and insurance claims;type 2 diabetes estimates were computed by subtracting type 1 diabetes from total estimates. Mortality and prevalence estimates, along with standard life expectancy and disability weights, were used to calculate YLLs, YLDs, and DALYs. Starch molecules in black rice offer exceptional nutritional benefits, however, the effects of processing-induced structural changes on their functional properties remain underexplored. This study evaluated the effects of pressure cooking (PC) and electric rice cooking (RC), combined with three drying methods (hot air, vacuum, and freeze-drying), influences the quality of instant rice from two Thai black rice varieties, Jao Kham Hom Maejo 1A (M1A) and Thai Rice Berry (TRB). Among the 40 treatment combinations, only M1A exhibited starch f for instant rice production. Specifically, pressure cooking for 4 min followed by freeze-drying at −20 °C for 12 h (M1A_PC4_F) most effectively preserved starch functionality while retaining high bioactive compounds, including total phenolics, anthocyanins, and antioxidant activities. Structural analysis using X-ray diffraction and differential scanning calorimetry confirmed that this thermal process initiated partial starch gelatinization while maintaining amylose-amylopectin crystallinity. These structural modifications increased resistant starch content, improved pasting properties, and enhanced rehydration characteristics, with the potential to moderate the glycemic response. Collectively, this study introduces pressure cooking (4 min) followed by freeze-drying as a novel approach for producing instant black rice that uniquely preserves starch crystallinity (26 %) while maintaining anthocyanins and resistant starch content for enhanced nutritional value. © 2025 Elsevier B.V.

Cardiovascular diseases (CVDs) are still the leading mortality causes in the last decades. The ailments are multifactorial characterized by excessive clot (thrombus) formation in the blood vessels. Thrombus could be degraded through thrombolysis mechanism by plasmin activated by various fibrinolysis agents including, urokinase, nattokinase, or streptokinase. However, the use of these agents is restricted by relatively high cost, short half-life, allergic reaction, and bleeding effects. The search for more economical and safer thrombolytic (clot lysis) agents are essential to address the underlying problem in CVD therapy. Among Holothurians, H. scabra has been known to have the highest protein content making it ideal substrate for protease enzymes including fibrinolytic types with clot lysis properties. However, isolation of a proteases with antithrombotic activities either from tissue or from bacteria of H. scabra has not been reported. This study aimed to screen proteolytic and clot lysis activities of crude protease extracts from tissue and bacteria isolated from fermented intestine of H. scabra. Crude protease of tissue of H. scabra was extracted by cold centrifugation, which activity was measured using UV-spectrophotometer. Crude protease of proteolytic bacteria selected by clear zones on skim milk agar (SMA) medium was isolated from nutrient broth (NB). Extracts showing proteolytic activity were subjected to gravimetry-based clot lysis test. As results, crude proteases isolated directly from the H. scabra's tissue showed low proteolytic activities, thus were no proceed to clot lysis activity test. Crude protease extracted bacteria could show both proteolytic and clot lysis activities. In conclusion, based on this screening study, intestine of H. scabra is a rich source of proteolytic bacteria, some of them could produce crude enzymes showing competitive thrombolysis activities with Nattokinase. This study aimed to develop bioremediation agent with bacterial cells as components for the treatment of biomedical wastes from two hospitals in Semarang City (Central Java), i.e. Roemani Muhammadiyah (coded R1) and Wongsonegoro (coded R2). Single isolates and consortium of indigenous hydrolytic bacteria characterized as multiple hydrolytic enzyme producers with low-to non-pathogenic properties obtained from previous study were tested for their degradation performance. The degradation performance test is necessary to formulate components of bacterial consortium as bioremediation agent. The tests were conducted on the selected bacteria as single isolate and as consortium. The six bacteria tested as single colonies and as consortium were Bacillus velezensis R1.3, B. amyloliquefaciens R1.6, B. amyloliquefaciens R1.14, B. velezensis R1.16, B. licheniformis R2.5, and B. amyloliquefaciens R2.9. Degradation performance on biomedical waste mainly containing organic matters was assessed based on water pollution parameters on 4.0-L samples. They included a control, 6 samples treated with bacteria as single colonies, a sample treated with bacteria as indigenous consortium and a sample treated with bacteria as mixed consortium. Parameters of wastewater pollution measured included COD (Chemical Oxygen Demand), BOD (Biological Oxygen Demand), TSS (Total Soluble Solid), NH3, and PO4. Next, encapsulation of a bacterial consortium as the best condition for degradation was also carried out using maltodextrin to allow storage and preservation of the bioremediation agent for longer period. The encapsulated product was visualized in SEM images to evaluate its quality. The results showed that a consortium comprising 4 indigenous bacterial isolates from R1 hospital could decrease BOD of biomedical wastewater by 85% and TSS by 43%. Cardiovascular diseases (CVD) are still the main cause of death in modern society with thrombosis as their most common underlying pathology. The occurrence of thrombosis is characterized by thrombus formation in the circulatory system of the body causing blood clot blockage. Thrombus blocking blood vessels can be destroyed through fibrinolysis by plasmin. Commonly administered fibrinolytic drugs activate plasminogen and convert plasminogen to plasmin which degrades fibrin. However, these agents have limitations such as higher costs, shorter half-lives, allergic reactions and intravenous administration-caused bleeding. Hence, more effective and safer antithrombotic drugs are needed in anti-thrombosis therapy. This paper was intended to assess the ability of Holothuria scabra tissue fibrinolytic proteases as a thrombolytic agent based on literature studies. Summary of recent studies showed that various strong fibrinolytic enzymes could be produced from various animal tissues such as snake and earthworm, yet from sea organism' tissues are barely reported. In this literature review, the potential of protease enzymes from tissue of H. scabra as thrombolysis agent is summarized and discussed. Among the findings is that H. scabra has the highest protein content among other sea cucumbers in Indonesia, yet the isolation of protease enzymes from H. scabra tissue has not been reported. Hence, there is high possibility in finding novel proteases, which may include serine and metallo-protease known for their thrombolysis activities.

Molecular phylogeny and postharvest morphology of petals in two major Nelumbo nucifera cultivars in Thailand

Nelumbo nucifera is a symbolic flower of Buddhism and widely used for decoration in Asia. In most cases its buds are used as cut-flowers, which usually do not open and the outer petals turn black, leading to loss of visual quality and thus decreasing their economic value in markets. In Thailand, two major cultivars, Sattabongkot and Saddhabutra, are used and exported to foreign countries. To investigate the difference between these two cultivars, their molecular phylogeny and postharvest morphology were examined. Using 25 simple sequence repeat (SSR) markers, the cultivar Saddhabutra was very close to the cultivar Satabuto. These results showed a high bootstrap (BS) value of 96%. In contrast, lotus cultivars in Thailand including Sattabongkot and Saddhabutra, showed a BS value of 90%. Saddhabutra and Satabuto are classified in the same group with high similarity, whereas Sattabongkot shows relatively lower similarity. Petal blackening started earlier in Saddhabutra than Sattabongkot, indicating a vase-life difference among cultivars, with Sattabongkot and Saddhabutra having a vase life of 72 and 54 h, respectively. The epidermal cells of normal petals of Sattabongkot and Saddhabutra showed freshness and a round shape with turgid cells. The petal blackening was accompanied with a reduction of the area and perimeter in both cultivars. Sattabongkot had fewer stomata than Saddhabutra. The study provided a molecular classification of the Thailand lotus cultivars and provides a useful technique for the quantification of the postharvest quality of lotus cultivars. © 2018 Kasetsart University
