Bioethanol Production from Oil Palm Frond by Simultaneous Saccharification and Fermentation

Ethanol production from oil palm frond (OPF) by simultaneous saccharification and Saccharomyces cerevisiae TISTR5048 fermentation was investigated. Solid fraction of OPF (20% TS) was pretreated by 2% H 2 SO 4 , 2% NaOH and 2% NaOH in H 2 O 2 presoaking at room temperature for 24 hours. Pretreated OPF by presoaking in 2% H 2 SO 4 , 2% NaOH and 2% NaOH in H 2 O 2 contained 37%, 42% and 49% of cellulose, respectively. Pretreated OPF was simultaneous saccharification by cellulase enzyme (Cellic CTec2, Novozymes) and sequentially fermentation. Sugar concentration in OPF cellulose hydrolysis of 2% H 2 SO 4 , 2% NaOH and 2% NaOH in H 2 O 2 presoaking was 45.72, 55.73 and 56.94 g/l, respectively. Ethanol concentration of 2% H 2 SO 4 , 2% NaOH and 2% NaOH in H 2 O 2 presoaking was 14.5, 15.0 and 17.2 g/L, respectively. 2% NaOH in H 2 O 2 presoaking was the best pretreatment with 82.11% of total solids recovery and containing 49.9% of cellulose with enzyme digestion ability of 37.6%.

The molecular epidemiology data of torque teno virus (TTV) in Indonesia is very rare. This study evaluated the prevalence of TTV in male commercial sex workers, as one of the high risk community for blood borne viruses pathogens in Surakarta, Indonesia. All blood samples collected from male commercial sex workers in Surakarta in 2009-2013 were tested by nested polymerase chain reaction (PCR). The amplified PCR products were molecularly cloned and subjected to sequence analysis. TTV DNA was detected in 80.9% (72/89) samples. Furthermore, the molecular characterization revealed that the most prevalent was genogroup 3, followed by genogroup 2 and l, respectively. TTV was detected in male commercial sex workers in Surakarta with high infection rate. Further investigation about TTV circulation in Indonesian population is needed in order to provide additional information about the genetic variability and TTV epidemiology in Indonesia, especially in the high risk communities.

Effect of Operating Parameters on Process Stability of Continuous Biohydrogen Production from Palm Oil Mill Effluent under Thermophilic Condition

Effect of organic loading rate (OLR), hydraulic retention time (HRT) and temperature variation on process stability of fermentative hydrogen production from palm oil mill effluent (POME) under thermophilic condition was investigated. High OLR (>70 gCOD/l/d) and low HRT (4 d) has significant affected on hydrogen productivity, process stability and microbial community. Hydrogen production at OLR of 55, 60, 65 and 70 gCOD/l/d was 2.4, 3.4, 4.9 and 3.7 L H 2 /L-POME, respectively. The decreasing in pH and alkalinity and increasing in total VFA could cause the instability of process. The microbial community was changed at OLR >70 gCOD/l/d, HRT 4 d and stop feeding. The dominant microbial community changed from Thermoanaerobacterim sp. to Clostridium sp., Thermoanaerobacterim sp. and Bacillus sp. Hydrogen production at HRT of 2, 4, stop feeding and 2 d was 2.7, 0.8, 0, and 2.6 L H 2 /L-POME, respectively. Hydrogen production, process stability and microbial community was not affected by variation of temperature. Hydrogen production before and after temperature variation was 2.5 and 2.45 L H 2 /L-POME.

The human leukocyte antigen (HLA)-DQB1 gene polymorphisms may associated with the infection risk of Toxoplasma gondii in HIV patients. The HLA-DQB1*0402 in HIV-1-positive patients could be considered risk factors for developing neurological opportunistic infections, mainly Toxoplasma encephalitis. However, the HLA-DQB1*0402 gene polymorphisms status in the Javanese HIV patients is unknown. This study evaluated the prevalence of HLA-DQB*0402 alleles polymorphisms in Javanese HIV patients with positive anti-Toxoplasma gondii IgM status. Since 2009 our research group performing a molecular epidemiology of blood borne viruses in Central Java Indonesia, by collecting the epidemiological and clinical data from the high risk communities. All blood samples were screened for blood borne pathogens by serological and molecular assays including for HIV and Toxoplasma gondii. The genomic DNA was isolated from the whole blood samples. Genetic polymorphisms of HLA-DQB1*0402 alleles were detected with polymerase chain reaction-sequence-specific primers (PCR-SSPs) technique. The genotypes were defined according to generated fragment patterns in the agarose gel electrophoresis analysis of PCR products. All of the samples were tested at least in duplicate. HLA-DQB1*0402 alleles were detected in 20.8% (16/77) patients and not detected in all HIV positive samples with negative anti-Toxoplasma gondii IgM status (n= 200). The HLA-DQB1*0402 alleles polymorphisms were detected in Javanese HIV patients with positive anti-Toxoplasma gondii IgM. The polymorphisms found may have association with the infection risk of Toxoplasma gondii in HIV patients.

To evaluate associations by EGFR mutation status for lung adenocarcinoma risk among never-smoking Asian women, we conducted a meta-analysis of 11 loci previously identified in genome-wide association studies (GWAS). Genotyping in an additional 10,780 never-smoking cases and 10,938 never-smoking controls from Asia confirmed associations with eight known single nucleotide polymorphisms (SNPs). Two new signals were observed at genome-wide significance (P < 5 × 10-8), namely, rs7216064 (17q24.3, BPTF), for overall lung adenocarcinoma risk, and rs3817963 (6p21.3, BTNL2) which is specific to cases with EGFR mutations. In further sub-analyses by EGFR status, rs9387478 (ROS1/DCBLD1) and rs2179920 (HLA-DPB1) showed stronger estimated associations in EGFR-positive compared to EGFR-negative cases. Comparison of the overall associations with published results in Western populations revealed that the majority of these findings were distinct, underscoring the importance of distinct contributing factors for smoking and non-smoking lung cancer. Our results extend the catalogue of regions associated with lung adenocarcinoma in non-smoking Asian women and highlight the importance of how the germline could inform risk for specific tumour mutation patterns, which could have important translational implications.

RNA in situ hybridization （ISH）, including chromogenic ISH （CISH） and fluorescent ISH （FISH）, has become a powerful tool for revealing the spatial distribution ofgene transcripts in model organisms. Previously, we developed a robust protocol for wholemount RNA CISH in the pea aphid Acyrthosiphon pisum, an emerging insect genomicmodel. In order to improve the resolving capacity of gene detection, we comprehensively surveyed current protocols of whole-mount RNA-FISH and developed protocols that allow,using confocal microscopy, clearer visualization of target messenger RNAs （mRNAs） - including those subcellularly localized and those with spatially overlapping expression. Wefind that Fast dye-based substrate fluorescence （SF）, tyramide signal amplification （TSA）, and TSA Plus all enable identifying gene expression thanks to multiplex amplificationof fluorescent signals. By contrast, methods of direct fluorescence （DF） do not allow visualizing signals. Detection of a single gene target was achieved with SF and TSA Plusfor most mRNAs, whereas TSA only allowed visualization of abundant transcripts such as Apvasl andAppiwi2 in the germ cells. For detection of multiple gene targets using doubleFISH, we recommend：（i） TSA/TSA, rather than TSA Plus/TSA Plus for colocalized mRNAs abundantly expressed in germ ceils, as proteinase K treatment can be omitted;and （ii） SF/TSA Plus for other gene targets such as Apenl and Apen2 as inactivation of enzyme conjugates is not required. SF/SF is not ideal for double FISH experiments due tosignal blurring. Based on these new conditions for RNA-FISH, we have obtained a better understanding of germline specification and embryonic segmentation in the pea *** anticipate that the RNA-FISH protocols for the pea aphid may also be used for other aphids and possibly other insect species, thus expanding the range of species from whichuseful insights into development and evolution may be obtained.
