Comparative transcriptomic and molecular interaction analysis of olive polyphenols: Unraveling their mechanisms in immune regulation and metabolic processes in adipocytes

Olive polyphenols, including oleacein, oleocanthal, and oleuropein, have demonstrated potential benefits in mitigating the risk of metabolic diseases such as obesity and inflammation by improving glycemic control, reducing blood pressure, and lowering inflammation. This study provides a comprehensive comparative analysis of these compounds using human adipose-derived stem cells (hASCs). Whole-transcriptome analysis was employed to investigate the compounds' effects on adipocytes, focusing on immune system and metabolic pathways. A protein-protein interaction (PPI) network identified key targets and pathways. Oleacein uniquely enriched the PI3K-AKT pathway and demonstrated distinct regulation of inflammation and metabolism. The PPI network pinpointed growth hormone receptor (GHR) and interleukin-6 receptor (IL6R) as significant targets. Quantitative RT-qPCR analysis showed that oleacein significantly increased IL6R expression, while oleocanthal and oleuropein had no effect. GHR expression was also elevated by oleacein, although without statistical significance. Physicochemical properties of the compounds were analyzed for drug development potential. Molecular docking simulations indicated superior binding affinities for oleacein with GHR and IL6R, supported by surface plasmon resonance (SPR) assays, which confirmed stable interactions with both receptors. This study is the first to integrate transcriptomic, molecular, and structural analyses of these bioactive olive-derived compounds. Oleacein emerged as a promising candidate for therapeutic development, with unique molecular interactions and regulatory effects on adipocyte immune and metabolic processes. These findings provide a foundation for future investigations into oleacein's role in treating metabolic and inflammatory disorders.

Previous studies have demonstrated the preventive effects of greenness and greenspace using three classes of exposure metrics (greenspace coverage, greenness, and landscape indices for greenspace) on suicide. Nevertheless, most studies were individually employed for each metric using a single-phase method, representing the short-term exposure. We hierarchically investigated the association between three classes of exposure metrics and suicide using long-term data from 2000 to 2018 in Taiwan. Normalized difference vegetation index (NDVI) was used to represent the greenness exposure, while mean patch area (area and edge), area-weighted mean fractal dimension index (shape), and proximity index (proximity) were opted to represent landscape indices for greenspace. Using the generalized additive mixed model, we revealed that the relative risk (95% confidence interval) of greenspace coverage, NDVI, mean patch area, area-weighted mean fractal dimension index, and proximity index was 0.998 (0.997, 0.999), 0.860 (0.811, 0.912), 0.999 (0.999, 0.999), 1.485 (1.056, 2.088), and 0.999 (0.999, 0.999), respectively. This indicates that long-term exposure to both greenspace coverage and greenness is associated with a reduced suicide ratio. Similarly, long-term exposure to greenspace that is greater in size, simpler in shape, and closer in proximity is associated with a decreased suicide risk. For formulating future afforestation policies, the layout and connectivity of greenspaces should be considered besides increasing green areas. This study not only demonstrated the significance of long-term exposure to greenspace for reducing the suicide ratio, but also illustrated a hierarchical approach to examine the effects of greenspace. © 2023

This study concentrates on the response, data sending, and survey completion rates of a smartphone-based travel survey in a developing country. A smartphone-based survey reinforced by a web-based survey was conducted in 2015 in Kabul, Afghanistan. The aims of this study are: 1) to explore the effects of age and gender on the response rate of the smartphone-based travel survey, and 2) to examine how gender, age, smartphone ownership history, and occupation influence the data sending and completion rates of the smartphone-based travel survey. The results show that gender had no effects on the response rate, while age had a significant impact. In addition, gender had effect on data sending rate, and the respondents who owned smartphones for longer time were more likely to complete the survey successfully. Furthermore, the survey withdraw rate of respondents was higher on weekends in comparison to weekdays.

This study investigated the spatiotemporal variation and long-range transport characteristics of atmospheric speciated mercury (ASM) along the West and East Channels surrounding Taiwan Island toward the South China Sea (SCS) in East Asia. Field sampling was conducted at three remote island sites, and potential sources of ASM were examined using backward trajectory clustering and regional fire maps. Seasonally, ASM concentrations followed the order: spring > winter > fall > summer, with higher ASM levels observed in the western waters of Taiwan Island than those in the eastern waters. Among the mercury species, gaseous elemental mercury (GEM) was predominant, and followed by particulate-bound mercury (PBM) and gaseous oxidized mercury (GOM), with total gaseous mercury (TGM = GEM+GOM) accounting for 92.1–98.4 % of ASM. Clustered trajectory analysis indicated that Asian Northeastern Monsoons (ANMs) may transport ASMs originating from continental sources in North and East Asia to the island sites via long-range transport. Slash-and-burn activities in the Indochina Peninsula and mainland China during spring and winter contributed to elevated ASM levels. The West Channel exhibited a pronounced “channel effect,” amplifying industrial emissions across the Taiwan Strait and resulting in higher ASM levels at the Penghu Islands. Conversely, the East Channel received diluted air masses through long-range transport from the Korean Peninsula and North China via Green Island and the Bashi Channel, leading to relatively lower ASM levels at the Dongsha Island. Overall, this study highlights the importance of seasonal circulation patterns and regional combustion sources in shaping mercury distribution in the marine boundary layer of East Asia.

Play behaviors and signals during playful interactions with juvenile conspecifics are important for both the social and cognitive development of young animals. The social organization of a species can also influence juvenile social play. We examined the relationships among play behaviors, candidate play signals, and play bout termination in Tibetan macaques （Macaca thibetana） during juvenile and infant social play to characterize the species play style. As Tibetan macaques are despotic and live in groups with strict linear dominance hierarchies and infrequent reconciliation, we predicted that play would be at risk of misinterpretation by both the individuals engaged in the play bout and by those watching, possibly leading to injury of the players. Animals living in such societies might need to frequently and clearly signal playful intent to play partners and other group members to avoid aggressive outcomes. We gathered video data on 21 individually-identified juvenile and infant macaques （one month to five years of age） from the Valley of the Wild Monkeys, Mt. Huangshan, China. We used all-occurrence sampling to record play behaviors and candidate play signals based on an ethogram. We predicted that play groups would use multiple candidate play signals in a variety of contexts and in association with the number of audience members in proximity to the players and play bout length. In the 283 playful interactions we scored,juvenile and infant macaques used multiple body and facial candidate play signals. The disposal of spent activated carbon(AC) will inevitably create secondary pollution. In overcoming this problem, the spent AC can be regenerated by means of biological approach. Bioregeneration is the phenomenon in which through the action of microorganisms, the adsorbed pollutants on the surface of the AC will be biodegraded and this enables further adsorption of pollutants to occur with time elapse. This review provides the challenges and perspectives for effective bioregeneration to occur in biological activated carbon(BAC)column. Owing to very few reported works on the bioregeneration rate in BAC column, emphasis is put forward on the recently developed models of bioregeneration kinetic in batch system. All in all, providing potential solutions in increasing the lifespan of AC and the enhancement of bioregeneration rate will definitely overcome the bottlenecks in spent AC bioregeneration.

Spreading marine dead zones (or hypoxia) are threatening coastal ecosystems and affecting billions of people's livelihoods globally. However, the lack of field observations makes it challenging to estimate dead zones with spatial precision and across large scales. While satellites offer great potential for detecting environmental changes through large-scale and temporal consistent data, they have yet to be fully integrated into the spatio-temporal dynamic mapping of hypoxia. To address this limitation, we integrated satellite imagery with field observations in random forest models on the Google Earth Engine platform to characterize dead zone dynamics from 2000 to 2019. We applied the workflow to the Gulf of Mexico, which has the largest dead zones in North America. Our model explained 64 % (± 5 %) of the variance in predicting dead zones using satellite data. The analysis revealed that dead zones in the Gulf peaked in 2009 (17,699 ± 679 km2) and contracted afterward in terms of both size and persistence (% days with hypoxia). Despite this contraction, the average size between 2010 and 2019 was twice that of the coastal reduction goal (2) set by the Gulf of Mexico Hypoxia Task Force. Furthermore, dead zones occurred more frequently in the western Gulf, and nearly half of the western region experienced dead zones annually. In addition to inter-annual changes, our analysis highlighted the intra-annual dynamics of this phenomenon. Notably, dead zones expanded in June, peaking in size from mid-August to early September. The high temporal and spatial resolution of this dataset allows policymakers to develop targeted management plans and environmental policies. Our approach, which incorporates remote sensing for long-term monitoring of coastal dead zones, can be applied to worldwide monitoring initiatives when paired with local field observations. © 2025 Elsevier B.V.

Southeast (SE) Asia is a highly biodiverse region, yet it is also estimated to cumulatively contribute a third of the total global marine plastic pollution. This threat is known to have adverse impacts on marine megafauna, however, understanding of its impacts has recently been highlighted as a priority for research in the region. To address this knowledge gap, a structured literature review was conducted for species of cartilaginous fishes, marine mammals, marine reptiles, and seabirds present in SE Asia, collating cases on a global scale to allow for comparison, coupled with a regional expert elicitation to gather additional published and grey literature cases which would have been omitted during the structured literature review. Of the 380 marine megafauna species present in SE Asia, but also studied elsewhere, we found that 9.1 % and 4.5 % of all publications documenting plastic entanglement (n = 55) and ingestion (n = 291) were conducted in SE Asian countries. At the species level, published cases of entanglement from SE Asian countries were available for 10 % or less of species within each taxonomic group. Additionally, published ingestion cases were available primarily for marine mammals and were lacking entirely for seabirds in the region. The regional expert elicitation led to entanglement and ingestion cases from SE Asian countries being documented in 10 and 15 additional species respectively, highlighting the utility of a broader approach to data synthesis. While the scale of the plastic pollution in SE Asia is of particular concern for marine ecosystems, knowledge of its interactions and impacts on marine megafauna lags behind other areas of the world, even after the inclusion of a regional expert elicitation. Additional funding to help collate baseline data are critically needed to inform policy and solutions towards limiting the interactions of marine megafauna and plastic pollution in SE Asia. © 2023 The Authors
