Purpose: Generating brain tumor volume measurements in a reproducible and efficient manner is a difficult, yet necessary, component of response assessment. The purpose of this study was to adapt and validate a multilevel Fuzzy C-means clustering algorithms for ROI tumor segmentation to allow consistent volumetric comparisons at multiple sites. Methods: Preoperative contrast-enhanced T1W images from 37 glioblastoma cases were segmented using Fuzzy C-means clustering-based methods and compared to manually contoured volumes created by specialists. The same was done post-operatively, using subtracted images to eliminate intrinsically T1-hyperintense material (blood). Volume computations based on the MacDonald criteria were also used for comparison. Agreement and inter-rater variability between volumes produced with each method was assessed by determining the concordance correlation coefficient (CCC). Results: The MacDonald criteria method had poor agreement (CCC=0.350–0.972) with manual contouring pre- and postoperatively, while the proposed semi-automated methods exhibited very high agreement (CCC=0.839–0.995) with manual contouring before and after resection. Fuzzy C-means clustering with three classes was the most robust semi-automated method, showing better inter-rater agreement than the MacDonald criteria method for both pre- (CCC of 0.990 and 0.975, respectively) and post-operative cases (CCC of 0.983 and 0.576, respectively). Post-operative inter-rater agreement was significantly different between these methods (p < 0.001). Conclusion: The proposed semi-automated segmentation methods allow tumor volume measurements of MR images in a reliable and reproducible fashion necessary for measuring treatment response in glioblastoma patients in multicenter clinical trials.

Cerebral information processing involves the activation of brain regions and the exchange of information between them. As metric for intra-cortical communication, coherence, a frequency specific measure quantifying the correlation of the amplitudes across trials of two phase-locked signals, has been suggested. A value of 0 indicates that both signals are unrelated, a value of 1 that both signals are fully synchronized. It is evidenced by the plethora of findings that the functional connectivity between brain regions is modulated by brain states, mental processing and the planning and execution of actions. Thus, it might be hypothesized that volitionally modulating the functional connectivity between two interacting brain regions should have an impact either on behavior or cognition. The purpose of the study was to test whether subjects can learn to volitionally modulate brain functional connectivity between left and right motor cortices employing a neurofeedback training and whether the neurofeedback training induces specific effects on motor function. Thirty right-handed subjects participated in the study. The subject group was partitioned into two sub-groups, namely contingent feedback group and sham feedback group, consisting of fifteen subjects. During the neurofeedback training the magnetic brain activity originating from left and right primary motor cortices was recorded using magnetoencephalography (MEG) and a feedback signal representing the coherence between the primary motor cortices of both hemispheres was presented to the subjects on a screen. Subjects receiving contingent feedback were trained to increase coherence while the discriminative stimulus S D+ was presented and to decrease coherence while the S D− was presented. A pretest and posttest examination served to examine whether acquired control on functional connectivity evidences any behavioral effects. Pancreatic ductal adenocarcinoma (PDAC) is characterized by early recurrence following pancreatectomy, rapid progression, and chemoresistance. Novel prognostic and predictive biomarkers are urgently needed to both stratify patients for clinical trials and select patients for adjuvant therapy regimens. This study sought to determine the biological significance of RABL6A (RAB, member RAS oncogene family-like protein 6 isoform A), a novel pancreatic protein, in PDAC. Analyses of RABL6A protein expression in PDAC specimens from 73 patients who underwent pancreatic resection showed that RABL6A levels are altered in 74% of tumors relative to adjacent benign ductal epithelium. Undetectable RABL6A expression, found in 7% (5/73) of patients, correlated with improved overall survival (range 41 to 118 months with 3/5 patients still living), while patients with RABL6A expression had a worse outcome (range 3.3 to 100 months, median survival 20.3 months) (P = 0.0134). In agreement with those findings, RABL6A expression was increased in pancreatic cancer cell lines compared to normal pancreatic epithelial cells, and its knockdown inhibited pancreatic cancer cell proliferation and induced apoptosis. Moreover, RABL6A depletion selectively sensitized cells to oxaliplatin-induced arrest and death. This work reveals that RABL6A promotes the proliferation, survival, and oxaliplatin resistance of PDAC cells, whereas its loss is associated with extended survival in patients with resected PDAC. Such data suggest RABL6A is a novel biomarker of PDAC and potential target for anticancer therapy. © The Author(s) 2013.

Natural T helper 17 (nTH17) cells are a population of interleukin 17 (IL-17)-producing cells that acquire effector function in the thymus during development. Here we demonstrate that the serine/threonine kinase Akt has a critical role in regulating nTH17 cell development. Although Akt and the downstream mTORC1-ARNT-HIFα axis were required for generation of inducible TH17 (iTH17) cells, nT H17 cells developed independently of mTORC1. In contrast, mTORC2 and inhibition of Foxo proteins were critical for development of nTH17 cells. Moreover, distinct isoforms of Akt controlled the generation of T H17 cell subsets, as deletion of Akt2, but not of Akt1, led to defective generation of iTH17 cells. These findings define mechanisms regulating nTH17 cell development and reveal previously unknown roles of Akt and mTOR in shaping subsets of T cells. © 2013 Nature America, Inc. All rights reserved.

Sublethal thermal stress induces accelerated proliferation and growth factor expression in hepatocellular carcinoma

Purpose Tumor recurrence following percutaneous thermal ablation of hepatocellular carcinoma (HCC) remains a significant clinical problem, particularly in tumors ≥3.0cm. The aim of the present study was to test the hypothesis that sublethal heat stress of HCC cells or hepatocytes stimulates accelerated proliferation of non-heat stressed HCC cells and to identify candidate heat-stress induced pro-tumorigenic growth factors. Materials and Methods A small number of HCC cells stably transfected to constitutively express firefly luciferase (N1S1luc and AS30Dluc) were co-cultured with a feeder layer of non-transfected HCC cells (N1S1 or AS30D) or hepatocytes (Clone9) that were heat stressed to induce complete (50 C), partial (45 C) or no (37 C) cell death (N=3). Proliferation of non-heat stressed HCC cells was monitored by non-invasive molecular bioluminescence imaging from 0-6 days using IVIS 200 and a standard curve was generated to estimate cell number in vitro. The two HCC cell lines and the hepatocyte cell line underwent sublethal heat stress (45 C) or control (37 C) and recovered after 6 hours at 37 C for whole transcriptome microarray analysis to assess heat stress induced growth factor expression (N=4). Results Sublethal heat stress (45 C) of both HCC cells and hepatocytes induced a significant increase in proliferation of non-heat stressed N1S1luc (p<0.01) and AS30Dluc (p<0.01) HCC cells as early as 4-6 days after plating compared to the no feeder, no cell death (37 C) and complete cell death (50 C) groups. Whole transcriptome microarray analysis identified a rapid, significant, differential increase in the expression of 11 pro-tumorigenic growth factors and cytokines (2.0-45.2 fold increase over baseline; p<0.05) 6 hours post sublethal heat stress including PDGF , EGF, TGFα, VEGF, HB-EGF and IGF in both HCC cells and hepatocytes. Conclusion These data suggest that sublethal thermal injury to HCC cells and hepatocytes stimulates proliferation of residual HCC cell

Revealing neuronal circuitry using stem cell-derived neurons

Mouse embryonic stem cell (mESC)-derived neurons are a renewable cell source for investigation of neuronal circuits. Engineering circuit-tracing components into stem cells facilitates studies on mechanisms of synaptic coupling and circuitogenesis. This unit details methods for the generation of mESC-derived neurons harboring trans-synaptic viral tracing elements, which are used for investigation of synaptic connections within circuits in vitro, ex vivo, and in vivo. The first protocol describes procedures for feederfree passaging of mESCs, modified to carry reporter and rabies virus tracing elements. The second protocol describes in vitro generation of neurons from these ESCs. The last protocols describe the use of ESC-derived neurons as "source cells" for rabies virus circuit-tracing to identify inputs onto synaptically connected neurons. Given the broad applicability, these protocols can be applied to investigate the ability of in vitro-derived neurons to establish/maintain synaptic connections in disease models, and/or with humaninduced pluripotent stem cells. © 2013 John Wiley & Sons, Inc.

Coagulation Factor VIII Is Synthesized In Endothelial Cells

Combined deficiency of coagulation factors V and VIII (F5F8D) is an autosomal recessive bleeding disorder resulting from mutations in Lman1. This gene encodes a cargo receptor in the early secretory pathway that is responsible for the efficient secretion of factor V (FV) and factor VIII (FVIII) to the plasma. F5F8D is characterized by levels of both FV and FVIII reduced to ∼5-30% of normal. In contrast, Lman1 knockout mouse models of F5F8D exhibit FV and FVIII activities that are ∼50% of normal, relative to wildtype mice. Though FV and FVIII are synthesized at markedly different levels and potentially in different tissues, loss of the LMAN1 cargo receptor leads to parallel reductions in both FV and FVIII activity. FV is synthesized in hepatocytes (as well as megakaryocytes in the mouse). However, the primary cellular source of FVIII biosynthesis is controversial, with contradictory evidence supporting an endothelial or hepatocyte origin. We took advantage of the dependence of efficient FV and FVIII secretion on LMAN1 to examine the cellular source of each protein. FV and FVIII secretion profiles of conditional Lman1 knockout mice were characterized, relative to that of wildtype mice and ubiquitous Lman1 null mice (Lman1-/-). In order to generate mice with Lman1 expression specifically deleted in the endothelium or the hepatocytes, either a Tie2-Cre or Albumin-Cre transgene was crossed into Lman1 conditional mice (Lman1fl). FV and FVIII activity levels were measured by functional coagulation activity assays. Though Lman1fl/fl/Tie2-Cre+ mice (endothelial-specific knockout) exhibit normal plasma FV activity (99.9%) relative to wildtype mice (set to 100%), FVIII activity is reduced to 53.5% (p < 2.5 x 10-6). In contrast, Lman1fl/fl/Alb-Cre+ (hepatocyte-specific knockout) mice demonstrate normal FVIII activity (89.0%) and reduced FV activity (37.0%) (p < 1.4 x 10-10).
