International benchmarking competitions have become fundamental for the comparative performance assessment of image analysis methods. However, little attention has been given to investigating what can be learnt from these competitions. do they really generate scientific progress? What are common and successful participation strategies? What makes a solution superior to a competing method? To address this gap in the literature, we performed a multicenter study with all 80 competitions that were conducted in the scope of IEEE ISBI 2021 and MICCAI 2021. Statistical analyses performed based on comprehensive descriptions of the submitted algorithms linked to their rank as well as the underlying participation strategies revealed common characteristics of winning solutions. These typically include the use of multi-task learning (63%) and/or multi-stage pipelines (61%), and a focus on augmentation (100%), image preprocessing (97%), data curation (79%), and post-processing (66%). The “typical” lead of a winning team is a computer scientist with a doctoral degree, five years of experience in biomedical image analysis, and four years of experience in deep learning. Two core general development strategies stood out for highly-ranked teams: the reflection of the metrics in the method design and the focus on analyzing and handling failure cases. According to the organizers, 43% of the winning algorithms exceeded the state of the art but only 11% completely solved the respective domain problem. The insights of our study could help researchers (1) improve algorithm development strategies when approaching new problems, and (2) focus on open research questions revealed by this work.

Using the example of the 'h-related' publication dataset created for a previous study on the literature of Hirsch-type measures (Zhang et al. in J Informetri 5(3):583-593, 2011) and updated for the present paper, we attempt to study the robustness and dynamic evolution of 'core documents'. With using two different methods that have been recently introduced in the context of the detection and analysis of emerging topics (Glanzel and Thijs in Scientometrics 91(2):399-416, 2012), we show that the applied methods prove both stable and representative. Furthermore, the evolution of the core-document network represents and follows the general trends topic development in terms of focus shift from theory to application in an adequate manner.

Prasugrel is a third-generation thienopyridine that achieves potent platelet inhibition with less pharmacological variability than other thienopyridines. However, clinical experience suggests that prasugrel may be associated with a higher risk of de novo and recurrent bleeding events compared with clopidogrel in Japanese patients undergoing percutaneous coronary intervention (PCI). In this review, we evaluate the risk of bleeding in Japanese patients treated with prasugrel at the doses (loading/maintenance doses: 20/3.75 mg) adjusted for Japanese patients, evaluate the risk factors for bleeding in Japanese patients, and examine whether patients with a bleeding event are at increased risk of recurrent bleeding. This review covers published data and new analyses of the PrASFIT (PrASugrel compared with clopidogrel For Japanese patIenTs) trials of patients undergoing PCI for acute coronary syndrome or elective reasons. The bleeding risk with prasugrel was similar to that observed with the standard dose of clopidogrel (300/75 mg), including when bleeding events were re-classified using the Bleeding Academic research Consortium criteria. The pharmacodynamics of prasugrel was not associated with the risk of bleeding events. The main risk factors for bleeding events were female sex, low body weight, advanced age, and presence of diabetes mellitus. Use of a radial puncture site was associated with a lower risk of bleeding during PCI than a femoral puncture site. Finally, the frequency and severity of recurrent bleeding events during continued treatment were similar between prasugrel and clopidogrel. In summary, this review provides important insights into the risk and types of bleeding events in prasugrel-treated patients.

Effects of Topical diclofenac Plus Heparin (dhep plus H Plaster) on Somatic Pain Sensitivity in Healthy Subjects With a Latent Algogenic Condition of the Lower Limb

ObjectiveTo evaluate whether a diclofenac epolamine+ heparin topical (plaster) is more effective than diclofenac plaster alone in reducing deep somatic hyperalgesia in subjects without spontaneous pain and whether the effect is linked to or independent of the anti-edematous action of heparin. designProspective, double-blind, randomized and controlled, four-arm parallel design trial. SubjectsOne hundred and four patients (84 women, 20 men, mean age 42.213.3years), with deep somatic hyperalgesia in one thigh, randomly assigned to one of 4 groups of 26 each. InterventionEach group underwent one of the following plaster treatments on one thigh: diclofenac+heparin;diclofenac;heparin;placebo, for 7days, renewing the plaster every 24hours. Outcome MeasuresBefore treatment (day 1), at day 4 and day 8, assessment of (a) pressure and electrical pain thresholds of vastus lateralis and overlying subcutis and skin;and (b) structure/thickness of subcutis and muscle with ultrasounds at the same level. resultsduring treatment, in placebo and heparin, no significant threshold changes, except subcutis thresholds which increased slightly (P<0.02);in diclofenac and diclofenac+heparin, significant increase in all thresholds (0.0001 rical muscle pain thresholds increased significantly more in diclofenac+heparin than in diclofenac, heparin, and placebo (0.0001roups: no edema and thickness changes at ultrasounds in muscle and subcutis. ConclusionsTopical diclofenac+heparin is significantly more effective than diclofenac alone in reducing muscle hyperalgesia in subjects without spontaneous pain, independently of the anti-edematous action of heparin. The results provide a rationale for the use of diclofenac+heparin also in algogenic conditions without evident signs of injury/edema/hematoma.

Background Palbociclib, a cyclin-dependent kinase 4 and 6 inhibitor, is approved in many countries for the treatment of Er-positive and HEr2-negative advanced breast cancer in combination with endocrine therapy. We aimed to evaluate the pharmacokinetics, safety, and efficacy of palbociclib combined with letrozole as first-line treatment for Er-positive and HEr2-negative advanced breast cancer. Methods This is a phase 1, open-label, single-arm study in Chinese women with Er-positive and HEr2-negative advanced breast cancer. From cycle 1 day 1, patients received letrozole 2·5 mg orally once daily continuously plus palbociclib 125 mg orally once daily for 3 weeks followed by 1 week off treatment. Blood samples for pharmacokinetic evaluation of palbociclib were collected up to 120 h after a single dose of palbociclib in a lead-in phase and after palbociclib dose on cycle 1 day 21 for multiple dose pharmacokinetic evaluation. Predose blood samples for pharmacokinetic evaluation of palbociclib and letrozole were collected on days 19–21 of cycle 1 and cycle 2 day 1 (letrozole only). disease assessments were done every 12 weeks from cycle 1 day 1. Safety was assessed per Common Terminology Criteria for Adverse Events, version 4.0. This study is registered with ClinicalTrials, number NCT02499146 , and is ongoing but not recruiting. Findings As of March 16, 2016, the cutoff date, the study completed enrolment with 26 patients. All patients completed single dose pharmacokinetic evaluation and 13 completed multiple dose pharmacokinetic evaluation. After multiple doses, the steady-state palbociclib geometric mean area under the concentration–time curve from 0 to 24 h was 2005 ng × h/mL and maximum observed concentration was 112·3 ng/mL, similar to those obtained in Caucasian patients following the same dosing regimen. Measurement and interpolation uncertainties in rainfall maps from cellular communication networks

Accurate measurements of rainfall are important in many hydrological and meteorological applications, for instance, flash-flood early-warning systems, hydraulic structures design, irrigation, weather forecasting, and climate modelling. Whenever possible, link networks measure and store the received power of the electromagnetic signal at regular intervals. The decrease in power can be converted to rainfall intensity, and is largely due to the attenuation by raindrops along the link paths. Such an alternative technique fulfils the continuous effort to obtain measurements of rainfall in time and space at higher resolutions, especially in places where traditional rain gauge networks are scarce or poorly maintained. rainfall maps from microwave link networks have recently been introduced at country-wide scales. despite their potential in rainfall estimation at high spatiotemporal resolutions, the uncertainties present in rainfall maps from link networks are not yet fully comprehended. The aim of this work is to identify and quantify the sources of uncertainty present in interpolated rainfall maps from link rainfall depths. In order to disentangle these sources of uncertainty, we classified them into two categories: (1) those associated with the individual microwave link measurements, i.e. the errors involved in link rainfall retrievals, such as wet antenna attenuation, sampling interval of measurements, wet/dry period classification, dry weather baseline attenuation, quantization of the received power, drop size distribution (dSd), and multi-path propagation;and (2) those associated with mapping, i.e. the combined effect of the interpolation methodology and the spatial density of link measurements. We computed similar to 3500 rainfall maps from real and simulated link rainfall depths for 12 days for the land surface of the Netherlands.
