YTHDF1 shapes immune-mediated hepatitis via regulating inflammatory cell recruitment and response

Severe immune responses regulate the various clinical hepatic injuries, includingautoimmune hepatitis and acute viral hepatitis. N6-methyladenosine (m6A) modification is acrucial regulator of immunity and inflammation. However, the precise role of YTHDF1 in Tcell-mediated hepatitis remains incompletely characterized. To address this, we utilizedConcanavalin A (ConA)-induced mouse liver damage as an experimental model for T cell-mediated hepatitis. Our findings found that hepatic YTHDF1 protein rapidly decreased during ConAinduced hepatitis, and YTHDF1-deficient (Ythdf1^(-/-)) mice showed more susceptibility toConA-induced liver injury, along with an intensified inflammatory storm accompanied by aggravated hepatic inflammatory response via ERK and NF-κB pathways. Interestingly, hepatic-specific over-expression or deletion of YTHDF1 exhibited redundancy in ConA-induced liver *** in bone marrow chimeric mice confirmed the necessity of YTHDF1 in hematopoieticcells for controlling the response to ConA-induced hepatitis. Additionally, our data revealedthat YTHDF1 deletion in macrophages exacerbated the inflammatory response induced by lipopolysaccharide. In summary, our study uncovered that YTHDF1 deficiency exacerbates the immune response in ConA-induced hepatitis by modulating the expression of inflammatorymediators, highlighting the potential of YTHDF1 as a therapeutic target for clinical hepatitis.

In recent years, autonomous rehabilitation training systems with no or reduced supervision have attracted much research attentions. It is essential to automatically record and recognize motions so as to realize autonomous rehabilitation training. In this paper, we propose a motion recognition model of upper-limb rehabilitation exercise (MRURE) for post-strokes. Wearable inertial measurement unit (IMU) is used to collect motion data including tri-axial acceleration and angular velocity. A Mahalanobis distance-based dynamic time warping (MDDTW) technique and an improved metric learning algorithm Nearest Neighbors Constraint (NNC) are built to measure the similarity between motions. Then, the kNN classifier is adopted to classify motions based on Mahalanobis distance. In order to verify and validate the proposed motion recognition model, we conduct extensive experiments by motion samples collected from healthy participants. The overall recognition accuracy for five motions achieves 99.34%, which demonstrates that MRURE is effective for motion recognition of upper-limb exercises. The performance of MRURE is also compared with kNN based on Euclidean and Manhattan distance and the state-of-the-art motion recognition methods. Experiment results show that the accuracy of kNN based on Mahalanobis distance is better than that of Euclidean and Manhattan distance, and MRURE outperforms methods based on feature extraction. © 2023, The Author(s), under exclusive licence to Springer-Verlag GmbH Germany, part of Springer Nature.

Chirp-free hundred-picosecond pulse fiber laser based on carbon nanotubes mode-locker

We demonstrate a narrow bandwidth passively erbium-doped mode-locked fiber laser based on single-wall carbon nanotubes (SWCNTs). The center wavelength is 1546.61 nm. The strong filtering effect and low saturable absorber (SA) modulation depth provide advantages for a long pulse duration of 323 ps. An overall spectral bandwidth of 1.3 GHz (0.01 nm) is measured by the optical heterodyne method, representing the narrowest spectral width based on SWCNT under a single filter structure. The obtained spectral width is very close to the transform-limited bandwidth of 8 pm, indicating that the pulse is chirp-free. Meanwhile, the generation of beat signals reveals the strong coherence between pulses. The repetition frequency of the laser is 7.8 MHz, and the number of longitudinal mode locks is only 166, which is the minimum number of longitudinal mode-locking in passively mode-locked laser based on material SA. At the same time, numerical simulations further support our experimental results and reveal the pulse evolution, providing evidence that SA and filter are the key factors in the generation of chirp-free long pulse. © 2025 Optica Publishing Group under the terms of the Optica Open Access Publishing Agreement.

Container technology is widely used and improve the efficiency of container real-time migration has become an important research topic. Existing studies mainly focus on optimizing iterative dumping of containers without fully considering the impact of migration node selection on migration efficiency. Software-defined networking (SDN), achieves centralized control of the network by separating the control plane and the forwarding plane. Applying deep learning to SDN makes tasks such as traffic prediction and network optimization more intelligent. This paper proposes Prediction Model based on CLCB (CNN-LSTM-CBAM) uses real-time collected load data as input, extracts feature components of load data using one-dimensional convolutional neural networks, and then predicts using LSTM. To improve prediction accuracy, a lightweight CBAM attention module is introduced, and the prediction results are output through fully connected layers. The Network Traffic Prediction Model based on GCGRU-Conv (GCN-GRU-CNN) considers both spatial and temporal features of network traffic. GCN and GRU are used to extract spatial and temporal features of network traffic, respectively, and then these features are fused together using convolutional neural networks. To address the gradient problem caused by increasing network depth, a residual network structure is introduced. Experimental results show that the method proposed can effectively reduce the container migration time and has better prediction results. © 2024 IEEE.

The precise forecasting of traffic data holds significant importance for numerous practical applications in the real world. The challenge lies in the nonlinear spatio-temporal dependencies of the data, as well as rapid error accumulations for multi-step predictions. Therefore, we propose a Generative Adversarial Nets model based on a Spatio-Temporal Graph Attention Network (STGAT-GAN). Utilizing spatio-temporal graph attention proves to be an effective method for capturing the spatio-temporal dependencies, while GANs can learn a rich distribution over spatio-temporal data implicitly to produce realistic data adversarially. Experiments are carried out on two public traffic datasets. Comparative experimental analysis reveals that the proposed STGAT-GAN has better performance. © 2023 IEEE.

The promptness of decision-making is of paramount importance in coordinate decision-making communication systems. In this paper, we envision a system in which a sender transmits information to a receiver for decision-making. The decision-making process encompasses the extraction of decision-related information followed by subsequent computation. To assess the impact of communication delays, randomness of decision-making and computational time on the immediacy of decisions, we introduce a novel performance metric, namely the age of decision (AoD). Specifically, AoD is defined as the time elapsed since the generation of the update upon which the most recent decision is based. Considering the stochastic nature of decision-making, we analyze the average peak AoD in systems where the arrival and service processes follow general processes or Poisson processes. To facilitate our derivations, we introduce the concept of effective decisions to denote those decisions that can lead to the system's peak AoD. Simulation results confirm the validity of our derivations and illustrate that appropriately increasing the rate of decision-making can significantly reduce the system's average peak AoD. © 2024 IEEE.

Monitoring vital signs such as Heart Rate (HR) and Respiratory Rate (RR) is critical for veterinary medicine. The existing contact based systems are difficult to use on animals for longer periods as they may cause movement restriction. Contactless solutions have recently gained more popularity due to their ease-of-use. However, there are very few validated systems for animals. In this study, we propose a contactless vital signs monitoring system, CageDot for animals. Our system provides continuous real-time monitoring of HR and RR during hospitalization. The CageDot is placed under the animal cage and detects the heart vibrations using a geophone sensor. The CageDot also has a signal quality control algorithm to address the problem of obtaining high-quality cardiac data in real-life noisy environments such as hospitals. Compared to the existing works that use controlled environments, this is especially significant. The algorithm includes several steps that include background noise/movement removal, subject movement detection, heartbeat extraction and vital signs estimation. The experimental results on 16 hospitalized dogs and cats show that this system can achieve high accuracy for vital signs monitoring with a Mean Absolute Error (MAE) of 4.71 for Heart Rate (3.8 percent error rate) and 1.28 (8 percent error rate) for Respiratory Rate. © 2014 IEEE.

Neuroinflammation,mediated by the nucleotide-binding oligomerization domain-like receptor family pyrin domain-containing-3(NLRP3)inflammasome,is a significant contributor to the pathogenesis of neurodegenerative diseases(NDDs).Reynos-in,a natural sesquiterpene lactone(SL),exhibits a broad spectrum of pharmacological effects,suggesting its potential therapeutic ***,the effects and mechanism of reynosin on neuroinflammation remain *** current study explores the effects and mechanisms of reynosin on neuroinflammation using mice and BV-2 microglial cells treated with lipopolysaccharide(LPS).Our findings reveal that reynosin effectively reduces microglial inflammation in vitro,as demonstrated by decreased CD11b expression and lowered interleukin-1 beta(IL-1β)and interleukin-18(IL-18)mRNA and protein ***,in vivo,results showed a re-duction in the number of Iba-1 positive cells and alleviation of morphological alterations,alongside decreased expressions of IL-1βand *** analysis indicates that reynosin inhibits NLRP3 inflammasome activation,evidenced by reduced transcription of NLRP3 and caspase-1,diminished NLRP3 protein expression,inhibited apoptosis-associated speck-like protein containing a CARD(ASC)oli-gomerization,and decreased caspase-1 ***,reynosin curtailed the activation of nicotinamide adenine dinuc-leotide phosphate(NADPH)oxidase,demonstrated by reduced NADP^(+)and NADPH levels,downregulation of gp91^(phox) mRNA,pro-tein expression,suppression of p47^(phox) expression and translocation to the ***,reynosin exhibited a neuroprotective effect against microglial inflammation in vivo and in *** collective findings underscore reynosin’s capacity to mitigate mi-croglial inflammation by inhibiting the NLRP3 inflammasome,thus highlighting its potential as a therapeutic agent for managing neuroinflammation.
