Genetic purity and population structure of potato variety Granola L

The phenotypic performances have formed the basis for genetic purity evaluation. However, the phenotypic based the genetic purity assessment are time-consuming, expensive, and not stable due to a great environmental effect on traits expression. While, the genotypic based genetic purity evaluation of a variety offers an efficient, more stable, and precise result than those phenotypic method due to no environmental factors involved. The objectives of this study were to determine genetic purity and population structure of Granola L. potato variety derived from twelve collection sources using microsatellite markers. This study consisted of following steps, such as cultivation of Granola L. potato varieties in the field, DNA isolation, PCR analysis, amplified DNA fragment analysis. The results showed that Granola L. variety derived from twelve sources had high level of genetic purity which indicated by genetic similarity value of 0.96 observed in present study. Based on population structure analysis, Granola L. variety might have consisted of two sub-populations of which one sub-population was more dominant than other sub-population. Therefore, genetic purification activities for Granola L. variety are still needed to increase genetic purity and to reduce phenotypic differences in future.

Compatibility of Rimau Gerga Lebong (RGL) mandarin grafted onto three genotypes of citrus rootstock

Introduction of Citrumelo and Kanci, an alternative citrus rootstock, into citrus cultivation system in Indonesia as complement of Japansche citroen (JC) is very appreciated. Compatibility of Citrumelo and Fortunella to commercial citrus in Indonesia have not been evaluated. The aim of the study is to assess the compatibility of Rimau Gerga Lebong (RGL) mandarin citrus grafted onto JC, Citrumelo, and Kanci based on anatomical, morphological and physiological parameters. The results showed that 100% of RGL mandarin shoot bud grew on Citrumelo and JC rootstocks, but only 88.75% on Kanci rootstock. Anatomically, RGL mandarin at 60 days after budding (DAB) had perfectly adhered on Citrumelo rootstock and not yet fully adhered on JC and Kanci rootstock. Morphologically, RGL-Citrumelo and RGL-JC combinations at three years old plant resulted in the same plant growth parameters, better than RGL-Kanci combination. Physiologically, RGL-Citrumelo and RGL-JC combinations had the same chlorophyll index, better than RGL-Kanci combination. RGL-Citrumelo and RGL-JC combinations showed graft compatible and RGL-Kanci combination showed late incompatibility.

Conceptual framework for the development of West java Zero Waste Management Index

Over the past decade, zero waste emerged as one of the promising concepts for waste management. Many parties have introduced this concept as a new way of waste management, aiming to reduce as little amount of waste generated as possible and send nothing to landfills. To measure the success of the zero waste implementation, some set of indicators have been developed. Although these were claimed to be successful after their implementation, very often they are not applicable to other regions or countries. Therefore, this study recommends developing the Zero Waste Management Index, which fits with local or regional conditions. This paper will discuss a framework for developing the West java Zero Waste Management Index (WJZWMI). It will outline urban solid waste problem in West java, and a brief review of existing zero waste indexes. Further, it will emphasize on the conceptual framework of the WJZWMI, which then results in potential long list indicators as preliminary results of this study, while also charting pathways for future works in developing a comprehensive zero waste management index for West java province.

The low efficiency of fertilizer application in association with the inappropriate practice cropping system is two of the reasons for the low productivity of rice in tropical riparian wetland ecosystem. This study evaluated the response of rice to the timing and splitting of fertilizer application at different cropping patterns in a tropical riparian wetland ecosystem. Ciherang, a commonly cultivated rice variety, was utilized during this study. The field experiment was laid on a split-plot design with timing and splitting of fertilizer applications on different days after transplanting (DAT) as the main plot and cropping system as a subplot. The main plot consisted of T1=15 DAT, T2=30 DAT, T3=45 DAT, T4=15+30 DAT, T5=15+45 DAT, T6=30+45 DAT, and T7=15+30+45 DAT. The subplots included: P1=Common farmers’ practice and P2= Double row cropping system (P2). The results showed the highest plant height, shoot dry weight, grains per panicle, and yield of 98.89 cm, 66.58 g, 3.14 g, and 673 g m−2, respectively, were obtained in the T7 treatment. Meanwhile, regarding cropping pattern, the highest plant height, number of tillers, root dry weight, total dry weight, number of productive tillers, and grains per panicle of 96.46 cm, 29.01, 28.50 g, 85.7 g, 23.23, and 83.74 g respectively, were obtained in P1. The combination of T7 and P2 resulted in the highest yield (745 g m−2). Therefore, the fertilizer application at 15+30+45 DAT (T7) and utilization of a double-row cropping system (P2) is recommended for rice cultivation in the tropical riparian wetland area.

Determination of priority SNI-certified-products with AHP approach – A case study on food and beverage sector

Understanding consumers’ knowledge about standard-certified products is necessary for a standard implementation to protect the community from unsafe products; more specifically, regarding consumers’ preferences for products with SNI marking. However, with a large number of SNI in one sector, the research needs to make prioritization the study’s object(s). This study then aims to provide recommendations for priority SNI-certified products with a case study on the food and beverage sector. To achieve the aim, this study used the AHP approach with selected criteria of readiness of the industry, the readiness of the standardization and conformity assessment infrastructures and human resources, number of users, risk for consumers, and readiness for consumption. The criteria are used to weigh 5 products, namely bottled drinking water, iodized table salt, rice, canned fish, and yogurt. Risk for consumers is considered the most important criteria and rice was weighted as the priority of SNI-certified-product to be further researched in the food and beverage sector.

Soybean production can be increased through the assembly of new superior varieties. In a plant gene assembly, it is necessary first to discover the characteristics of the parents. One way to find out these characteristics is to conduct a morphological study. The current study employed a randomized complete block design with three replications to 10 soybean lines. The longest young and mature pod was found in MLGG 0582. The thickest young pod was observed in MLGG 0583, while the widest was reported by MLGG 0617. MLGG 0276 had the thickest pod skin of all varieties. In mature plants, the widest and the thickest pods were found in MLGG 0582 and MLGG 0707, respectively. MLGG 0617 reported the longest, the widest, and the thickest seed. There was no positive correlation found among the characteristics of the young pods, except the thickness of the pods and the skin (0.816). However, in the mature plants, the characteristics of the pods were positively correlated with the characteristics of the seeds. © 2019 IOP Publishing Ltd. All rights reserved.

Screening genetic resources of local accessions of Capsicum originated from east java

Inventory and collection of local chili varieties in production centers in east java had been carried out in the period 2013–2015 by east java Assessment Institute for Agricultural Technology (AIAT). A total of 143 accessions of five chili species (Capsicum annuum L., C. frutescens L., C. chinense Jacq., C. baccatum L. and C. pubescens L) was collected from eight chili production centers in east java and has been maintained by east java AIAT. A set of 85 local chili accessions was selected and evaluated for agronomic performance and biochemical compounds. Among the five species, C. annuum L. and C. frutescens L. dominated the distribution of chili species in east java. The other three species were still found in some spots of farm households and highlands. C. pubescens, locally known as 'Bodong' or 'Wudel' chili, was found on the high slopes of Mount Semeru, whereas C. chinense, known as 'Cotoh' chili by the locals, was also found in the highlands of Batu City. Based on agronomic performance there were several accessions with high-yielding potential (≥12 t/ha). Five accessions of C. frutescens collected from planting areas in the dryland with dry climate of Blitar not only had high yield potential, but also had high capsaicinoid content, low-fat content, low quercetin and high flavonoids and polyphenols. Phenotypic diversity and geographic origin may be useful as the criteria for selecting a good set of chili accessions.

The impact of the Covid 19 pandemic on the performance of SNI certified SMEs

The COVID-19 pandemic is causing unprecedented economic disruption and downturn in communities and organizations globally, including small and medium enterprises (SMEs). Indonesian National Standard (SNI) certified products have good quality and are competitive in trade. The purpose of this study was to determine the impact of the COVID-19 pandemic on the performance of SNI-certified SMEs. The research method used is descriptive quantitative involving 34 SMEs that apply SNI. The data collection method is an online interview using a structured questionnaire to SMEs implementing SNI with research variables covering four independent variables, namely financial, operational, supply chain, and reputation of SMEs. The analysis used is using quantitative analysis techniques using a Likert scale. The COVID-19 pandemic has harmed the performance of SMEs from the financial, operational, supply chain, and reputation aspects. The percentage of the impact caused is 70% with the information level being high. Financial and operational impacts have a high level of impact with a percentage of 72% and 73%, while the supply chain and reputation aspects have a moderate level of impact with an impact percentage of 68% and 63%, respectively. In the aspect of reputation, brand image, product quality, and competitiveness can be maintained by the implementation of SNI, so that it has the lowest percentage value. SNI certification also helps SMEs in product promotion and sales. Although the majority of SMEs stated that recertification was a priority, not a few SMEs experienced difficulties and incompetence in the SNI certification process.

Extraction Treatments Affect Total Flavonoid and Phenolic Contents of Cowpea (Vigna Unguiculata L. Walp.)

Plant secondary metabolites exist during normal plant growth. The production increases remarkably when the plants are challenged by environmental stress. Quantification of these metabolites is affected by extraction solvents and preparation treatments. This study aims to select extraction solvents and treatments which are able to extract high total flavonoid and phenolic contents in cowpea seeds. Acetone at concentrations of 70-80% produced higher total flavonoid and phenolic contents of 10.37-11.93 mg CE/g and 18.20 – 20.20 mg GAE/g in two cowpea cultivars. Antioxidant activities were in the range from 115.9 to 126.1 umol TE/g. Extraction of cowpea seeds using 70% acidified acetone produced a similar amount of total phenolic contents to those of 70-80% acetone. Traditional extraction treatments of shaking and maceration extracted a similar amount of the secondary metabolites. These simple extraction methods, therefore, could be suggested to extract flavonoid and phenolic contents as well as antioxidant activity in cowpea where access to modern types of equipment is limited.

We present population structure analysis of two differed Ongole grade cattle strains in Indonesia i.e., common Ongole grade (PO) and Kebumen Ongole grade (PO Kebumen). The aim of this study was to identify the genomic characteristic of both PO and PO Kebumen cattle strains. In this study, genotyping method of 15 PO and 25 PO Kebumen was carried out using an Illumina BovineSNP50 Bead Chip. In order to deeper analyses, we used additional 50K SNP data of 20 PO, 20 Nellore, 20 Brahman, 20 Simmental, and 20 Limousin that freely available in the data repositories. An estimation of the genetic population study was carried out using following approaches: pairwise fixation indices, heterozygosity and Nei's standard genetic distance, and ancestral based admixture. These analyses revealed a B. taurus influence in PO but none in PO Kebumen. Furthermore, we also identified the close admixture of PO Kebumen to Nellore. Since Nellore was originated from Ongole cattle, we presume the PO Kebumen is the maintained Pure Ongole Breed in Indonesia.
