Which knowledge counts? Contested meanings of water quality, responsibility and involvement in river restoration discourse in Indonesia

Since 2001, the Indonesian government has formulated and implemented several restoration programs to improve the Citarum’s water quality. However, these programs were often contested, particularly concerning the meanings of ‘water quality’ and how those informed approaches to responsibility and involvement. This paper problematises river restoration in view of these controversies towards improving people’s lives and their environment. Our investigation found (1) a selective use of scientific knowledge of water quality and related responsibilities; (2) a tension between broader inclusion and military involvement in river restoration; and (3) a diverse host of informal restoration practices that largely remain unnoticed in view of the government programs. The findings indicate that river governance can benefit from recognising and tuning into below-the-radar restoration practices to tackle river pollution.

Insect diversity on toxic candlenut (Reutealis trisperma) plantation in Bajawa, East Nusa Tenggara

Toxic candlenut (Reutealis trisperma) is an important plantation commodity that serves as barrier to prevent erosion, as main material for bio-pesticides and as renewable source of biofuel production. It has been studied that toxic candlenut development in post-mining field can restore soil structure while also influencing environment surrounding, including insects. The study examined insect diversity in the R. trisperma plantation aged ±7 years in Bajawa field, East Nusa Tenggara from July 2018 until February 2019. Insect were collected by installed malaise traps, yellow traps, and pitfall traps in 5 different plots A, B, C, D, and E for 24 hours. Collected insects were then identified in the laboratory using identification book. The results showed there were 13 orders of insects were collected by traps. The diversity index (H') of insect in the area were 4.053; 3.305; 3.521; 2.973 and 2.611 while the Evenness index (E) of insect were 0.872; 0.836; 0.780; 0.665 and 0.614 respectively. Insect diversity in the Bajawa plantation was high and the dominan individual was Hymenopteran and Dipteran. Several insects were in R. trisperma plant, as herbivores, predators, parasitoids, and pollinators. Further research is needed to determine each family of insect's role in the R. trisperma plantation.

Evaluation of chili accessions for resistance against Thrips sp. (Thysanoptera: Thripidae)

Thrips sp. (Thysanoptera: Thripidae) is one of chili pests that can cause significant crop losses by 23% and be even more harmful due to its ability to transmit virus. The use of resistant variety is one sensible strategy to control this pest attack. Thirty chili accessions were evaluated for resistance by counting of the number of pest, recording leaf damage and observing accessions morphological characters. The evaluation was conducted in the research Field Station of Indonesian Vegetable research Institute, Lembang, west java from March to July 2020. Ten of the 30 accessions had low leaf damage and were considered to be used as female parents in breeding program of resistant chili against Thrips sp.

Cultivation of teak continues to be developed. The decision to harvest teak trees depends on the economic value. The economic value of the teak tree is the money value contained in the teak trunk which is divided into parts which are then called nodes. The determination of the part is based on the diameter size. Perhutani recognizes three classes of nodes namely A1 (10 - 19 cm), A2 (20 - 29 cm) and A3 (> 30 cm) and are expressed in the log volume table (TVL). Perhutani's TVL stated the relationship between tree circumference and log volume. This relationship is expressed in the estimator model and classified by nodes. To enter these numbers using constants that are difficult to account for. For this reason, this study aims to provide a volume estimator model for teak logs. The method used is a destructive method and regression analysis. The results showed that TVL in FMU can be explained based on the class of nodes but ignores the place of growth. The shape of the model is compound. This model works well at tree circumference intervals of 50 - 250 cm. research like this needs to be propagated with wider tree circumference intervals.

Study on Ferrisia virgata and Planococcus minor as vectors of mottle disease in black pepper

Two species of mealybug, Ferrisia virgata and Planococcus minor were previously reported as vectors of Piper yellow mottle virus (PYMoV), the causal agent of mottle disease in black pepper (Piper nigrum). Several activities were performed to study the PYMoV transmission through F. virgata and Pl. minor, involving transmission experiment in ISMCRI greenhouse (Bogor, west java), field observation of mealybugs, and mottle disease incidence at ISMCRI field station (Sukabumi, west java). The result showed that the transmission rate of PYMoV through F. virgata and Pl. minor in black pepper reached 40.00 and 46.67%, respectively. Field temperature >33 °C, light intensity >700.00 lux, wind speed <1.10 m/s, and rainfall <80 mm/month were recorded during the occurrence of a peak population of mealybugs. Mealybugs population in the field was affected by insecticide application. Its population was lower in a block with insecticide spraying than in a block without insecticide spraying. Furthermore, mottle disease incidence was correlated with the population level of mealybugs. Based on PCR detection, the frequency of viruliferous mealybugs did not differ in a block with or without insecticide spraying, i.e., 6/39 and 6/37, respectively. This indicated that other factors than insect vectors might play an important role in disease spread.

Potential of Lactococcus lactis strain LAC3 isolated from Indonesian traditional fermented buffalo milk (Dadih) as antioxidant and antidiabetic agents

Dadih is one of Indonesia indigenous traditional fermented foods made from buffalo milk originating from west Sumatera. Dadih is traditionally made by keeping buffalo milk in a covered bamboo tube for two hours. The goal of this study as to discover secondary metabolites generated by Lactococcus lactis subsp. Lactis strain LAC3 (isolated from Dadih) proceed with bioactivity assay as an antidiabetic and antioxidant agent. The secondary metabolites were isolated then partially purified by ammonium sulfate precipitation followed by gel filtration chromatography. The DPPH technique was used to test antioxidant activity, whereas the α-glucosidase inhibition assay was used to determine antidiabetic activity. The potential fraction was then examined by LC-MS to identify structural information of secondary metabolites. Collected protein fraction (no. 2-12) showed antioxidant activity (39-90%) and exhibited antidiabetic activity around 82-95%. LC-MS results revealed several compounds related to antioxidant and antidiabetic activity such as hydrazine carboxamide derivatives and metformin hydrochloride. Our results indicated that secondary metabolites generated by lactic acid bacteria isolated from dadih have activity as antioxidant and antidiabetic agents and potential for further development as antioxidant and antidiabetic sources.

The potency of culturable fungi from tidal and non-tidal swamplands in Indonesia

Tidal and non tidal swamp land in Indonesia is a relatively large sub-optimal land resource and has not been used optimally for agricultural land due to various problems. Technological innovation for swamp land, especially for fertilization (biofertilizer) must pay attention to the indigenous microbial community, so that the application of biofertilizer can be effective. The aim of this research was to culturable fungal diversity and their potential of the tidal swamp soil in South Kalimantan and non tidal swamp soil in South Sumatra. The research was conducted in the Microbiology laboratory – ICABIOGRAD. Two samples were isolated from tidal and non tidal swamp soils in South Kalimantan and South Sumatra. Serial dilution methods and molecular identification were used to assess the diversity of fungal species. Production of IAA, siderophores, and ACC Deaminase was evaluated. Results showed there are 11 species consisting of Trichoderma asperellum KS.26.1, Penicillium sp. KS.26.2, *** KS.26.3, *** SS.27.1, *** SS.27.2, Talaromyces calidicanius SS.27.3, *** SS.27.4, *** SS.27.5, *** SS.27.1 PDA, *** SS.27.2 PDA, *** SS.27.3 PDA were identified from both tidal and non-tidal swamplands. Potential analysis showed that all fungal isolates were able to produce IAA, siderophores, and ACC Deaminase. The fungal diversity was higher in the non-tidal swampland than the tidal swampland as revealed by the differences in percentage occurrence of fungal species. There are potential indigenous fungal in the swampland to be used to improve productivity of the soils.

Drought tolerance screening of selected rice varieties by limited semi controlled field system

Drought tolerance is important trait for rice. Developing simple system for drought tolerance screening will increase efficiency and effectiveness of breeding for the certain purpose. This research aimed to screen selected variety for drought under limited semi controlled space system. The trial was conducted at the limited open space with controlled irrigation and drainage system in the area of Indonesian Centre of Rice research green houses in Subang, west java, from July to November 2019. Twenty varieties selected from previous drought screenings using concrete box method were transplanted at 21 days after sowing with planting space of 20 x 20 cm 1 – 3 seedling/hill in a single row of 10 plants followed randomized complete block design with two replications. The materials were watered for 4 weeks since transplanting. Re-watering was given until harvesting since IR 20 (drought sensitive check) was completely dry. The results showed variation level of drought tolerance among the tested genotypes. Based on leaf drying score at five weeks after treatment, Inpago 9 and Cisaat along with Salumpikit (drought tolerant check) were considered relatively tolerant. Inpago 9 (41.8 g) followed by Inpari 42 (34.7 g), and Inpago 8 (30.1 g) had the highest yield.

Rural development based on local resources utilization has been promoted as a strategy to achieve people prosperity in Indonesia. Although many studies have found that resource-based strategy is no longer relevant nowadays, for many areas this is still considered an appropriate approach. In order to examine the significance, we conducted a study in Sumba Barat Daya District of East Nusa Tenggara, focussing on coffee as one of the most abundant products in the region. Utilizing a socio-ecological approach, we collected data and information to depict the dynamics of coffee-based activities. A detailed study in Kadi Roma Village of Sumba Barat Daya on coffee ecosystem contribution to local community revealed that despite traditionally managed, coffee plays a significant role socially and economically. As a source of income to almost 17,000 smallholder farmers in Sumba Barat Daya, it can be concluded that coffee can become a gateway to attain successful rural development program. It also provides various opportunities for the community to participate which requires supportive innovation ecosystem to be established systematically by relevant local institutions, including include local government. © 2019 IOP Publishing Ltd. Induction of resistance against sugarcane mosaic virus by pathogen-derived resistance and RNA interference methods in transgenic sugarcane

Mosaic disease is the most major devastating pathogen in sugarcane caused by sugarcane mosaic virus (SCMV), otherwise SCMV was also causing mosaic disease in another crops, such as maize and sorghum. It has been reported that sugarcane productivity loss due to SCMV is up to 45% in susceptible sugarcane varieties. Therefore, a new variety for SCMV-resistant sugarcane trait is important to be developed since this strategy will support combating the emerging SCMV in sugarcane field. The coat protein (CP) virus is the most common target for genetic engineering strategies to induce defense mechanism against virus infection. In our studies, the SCMV-resistant transgenic sugarcanes were generated by two different approaches, pathogen-derived resistance (PDR) and RNA interference technologies, and the gene encoding for CP was successfully cloned from the susceptible variety of PS-881 cultivar (MH393888.1). Induction of the resistance using the PDR method that also called protein-based resistance by overexpression of the CP gene resulted in the resistance of transgenic sugarcane against SCMV infection. The full sequence of CP gene produced a highest resistance compared to N-terminally truncated protein. On the other hand, RNA interference (RNAi) approach was also conducted to investigate the efficiency of defense mechanism against SCMV in sugarcane. The transgenic sugarcane expressing intron-hairpin (RNAi) constructs produced a high number of resistances against SCMV infection. In addition, the agronomic performance of transgenic sugarcane under PDR and RNAi approaches targeting the CP gene has also been compared. Based on the observation, the RNAi approach transgenic sugarcane is more resistant against the SCMV infection compared to the PDR approach. This review is to summarize the current promising strategies for generating a new variety of sugarcane that has resistance to SCMV by genetic engineering approach.
