In silico analysis of Phlogacanthus thyrsiflorus Nees. and its protective effects against cardiomyopathic stress induced by alloxan in mice model

This study aims to evaluate the protective role of Phlogacanthus thyrsiflorus (P. thyrsiflorus) on hyperglycemia induced oxidative stress and apoptosis in the hearts of diabetic mice. Acute toxicity testing and preliminary phytochemical screens were performed on methanolic flower extract (MFE) after it had been prepared. Thereafter, molecular docking was performed. The results identified active compounds which demonstrated strong binding affinity with the selected target proteins involved in oxidative stress under diabetic conditions. Moreover, the oxidative damage in the tissues of the Normal control mice (NCM), Diabetic control mice (DCM), Ascorbic acid-treated Diabetic mice (D + AA), and MFE-treated Diabetic mice (D + MFE) groups were assessed using lipid peroxidation and the protein carbonyl test. The diabetic mice treated with MFE showed a significant reduction in protein carbonyl levels and malondialdehyde (MDA). The effect of MFE on apoptosis was demonstrated by TUNEL enzyme where a few apoptotic cells were shown by the terminal deoxynucleotidyl transferase (TdT)-mediated dUTP nick-end labeling (TUNEL) assay. Therefore, in diabetic mice, the MFE of P. thyrsiflorus has cardiomyopathy benefits via controlling hyperglycemia-induced oxidative stress and apoptosis. © The Author(s) under exclusive licence to Society for Plant research 2025.

The aim of this study was to analyze the level of income, knowledge, and impact of climate change on fishing household. This research was conducted in Pekon Tegineneng, Limau Subdistrict, Tanggamus Regency. Limau Subdistrict is the center of marine capture fishery in Tanggamus Regency. Data collection was carried out from July to August 2020 with total respondent reached 80 fishing households. Respondents were determined through the approach of simple random sampling. The data collected consisted of both qualitative and quantitative data. Data collection was performed through the method of survey, Focus Group Discussion (FGD), and interview with key informants. Data were analyzed by the qualitative descriptive approach. The level of monthly income and expenditure per fishing household amounted to Rp. 4,110,000 and Rp. 3,971,200, respectively. Knowledge level of fisherman about climate change was considered excellent as most fishermen have already known the phenomena of climate and environmental changes. The impact of climate change on fishing household included: difficulty to predict fishing season, change in fishing location (away from the shore), reducing and unpredictable fishing frequency and fishing time, also declining of fish catch. © Published under licence by IOP Publishing Ltd.

Argumentative Segmentation Enhancement for Legal Summarization

We use the combination of argumentative zoning [1] and a legal argumentative scheme to create legal argumentative segments. Based on the argumentative segmentation, we propose a novel task of classifying argumentative segments of legal case decisions. GPT-3.5 is used to generate summaries based on argumentative segments. In terms of automatic evaluation metrics, our method generates higher quality argumentative summaries while leaving out less relevant context as compared to GPT-4 and non-GPT models. © 2023, CC BY-NC-ND.

In recent times, there has been growing attention towards exploring the nutritional and functional aspects of potato protein, along with its diverse applications. In the present study, we examined the anti-osteoclast properties of potato protein hydrolysate (PP902) in vitro. Murine macrophages (RAW264.7) were differentiated into osteoclasts by receptor activator of nuclear factor-κB ligand (RANKL), and PP902 was examined for its inhibitory effect. Initially, treatment with PP902 was found to significantly prevent RANKL-induced morphological changes in macrophage cells, as determined by tartrate-resistant acid phosphatase (TRAP) staining analysis. This notion was further supported by F-actin analysis using a confocal microscope. Furthermore, PP902 treatment effectively and dose-dependently down-regulated the expression of RANKL-induced osteoclastogenic marker genes, including TRAP, CTR, RANK, NFATc1, OC-STAMP, and c-Fos. These inhibitory effects were associated with suppressing NF-κB transcriptional activation and subsequent reduced nuclear translocation. The decrease in NF-κB activity resulted from reduced activation of its upstream kinases, including I-κBα and IKKα. Moreover, PP902 significantly inhibited RANKL-induced p38MAPK and ERK1/2 activities. Nevertheless, PP902 treatment prevents RANKL-induced intracellular reactive oxygen species generation via increased HO-1 activity. The combined antioxidant and anti-inflammatory effects of PP902 resulted in significant suppression of osteoclastogenesis, suggesting its potential as an adjuvant therapy for osteoclast-related diseases. © 2024 Wiley Periodicals LLC.

Parabens (PBs) are esters of p-hydroxybenzoic acid, and there are growing concerns due to their potential to disrupt endocrine function and their wide use as preservatives in foodstuffs, including beverages. The consumption of bottled and hand-shaken teas is gradually replacing traditional tea consumption through brewing. However, no study has reported PB concentrations in different types of teas or packaging and their associated health risks. Our aim was to determine the concentration of PBs (methyl- (MetPB), ethyl- (EthPB), propyl- (PropPB), butyl-paraben (ButPB)) in green, black, and oolong teas in two varieties of products (bottled and hand-shaken teas), using UPLC-MS/MS. Additionally, we estimated the health risks associated with tea consumption in the general adult population of Taiwan. A Monte Carlo simulation was applied to estimate the distribution of daily PB intake through bottled (n = 79) and hand-shaken (n = 71) tea consumption. Our findings revealed geometric mean concentrations in bottled green/black/oolong teas were 714.1/631.2/532.1 ng/L for MetPB, 95.2/ 30.5/14.9 ng/L for EthPB, 77.9/28.3/non-detected (ND) ng/L for PropPB, and 69.3/26.6/ND ng/L for ButPB. Hand-shaken green/black/oolong teas exhibited concentrations of 867.5/2258/1307 ng/L for MetPB, 28.5/28.8/14.5 ng/L for EthPB, 25.4/18.3/17.8 ng/L for PropPB, and 30.3/18.0/15.5 ng/L for ButPB. The median MetPB concentrations in hand-shaken black (2333 ng/L) and oolong teas (1215 ng/L) were significantly higher than those in bottled black (595.4 ng/L) and oolong teas (489.3 ng/L). Conversely, median concentrations of EthPB, PropPB, and ButPB in bottled teas were significantly higher than those in hand-shaken teas. MetPB was the predominant PB, constituting 73.2–91.9% in bottled teas and 85–94% in hand-shaken teas. Background:Several countries rely on International medical graduates (IMGs) to fill critical service gaps. IMGs experience a range of challenges related to migration, acculturation, accreditation and career. Purpose:This study aimed to explore the journeys, perceptions and challenges of IMGs based in Australia. Methods:We used a sequential explanatory mixed methods approach for design and analysis. An online survey was distributed Australia-wide to IMGs through various avenues (including snowballing) between 13th October 2023 and 31 December 2023. We later conducted individual interviews between February and April 2024. Quantitative data was descriptively analysed. Qualitative data from survey written responses and oral interviews were thematically analysed. Study data was triangulated and congruence assessed. Full Ethics approval was obtained. Results:The study comprised 286 survey participants and 36 interview participants. Working conditions, location/lifestyle and personal/family opportunities were among the reasons for Australia's popularity as a host country. The most disliked aspects were system processes (eg, bureaucracy, certifications, licencing etc) (87/199; 43.7%), and discrimination, bias, racism, or prejudice (53/199; 26.6%). Two major qualitative themes identified were: (1) IMGs have a rich and diverse set of experiences and (2) IMGs encounter unexpected challenges with widespread impacts on IMGs, family and society. We found high congruency of triangulated data from the quantitative and qualitative studies supporting concepts around IMG diversity of experience and choices, life stage priorities, privilege, mismatched expectations, criticism of bureaucracy, gaps in understanding nuances of the Australian workplace, social challenges, and impact of challenges. Conclusion:IMGs describe numerous permutations of journeys, challenges and general experiences which result from factors both inside and outside the IMG's control. Challenges, particularly those

Cysteine-cysteine chemokine receptor type 5 (CCR5) is thought to play an important role in the trafficking of lymphoid cells but has recently also been associated with AMPK signaling pathways that are implicated in energy metabolism in skeletal muscle. We hypothesized that genetic deletions of CCR5 would alter mitochondria content and exercise performance in mice. CCR5^−/− and wild-type mice with the same genetic background were subjected to endurance exercise and grip strength tests. The soleus muscle was stained with immunofluorescence for myosin heavy chain 7 (MYH7) and succinate dehydrogenase (SDH) analysis as well as the expression of genes associated with muscle atrophy and mitochondrial oxidative phosphorylation were measured using qPCR. Although there were no differences in the weight of the soleus muscle between the CCR5^−/− group and the wild-type mice, the CCR5^−/− mice showed the following muscular dysfunctions: (i) decreased MYH7 percentage and cross-section area, (ii) higher myostatin and atrogin-1 mRNA levels, (iii) dropped expression of mitochondrial DNA-encoded electron respiratory chain genes (cytochrome B, cytochrome c oxidase subunit III, and ATP synthase subunit 6) as well as mitochondrial generation genes (PPARγ and PGC-1α), and (iv) lower SDH activity and exercise performance when compared with wild-type mice. In addition, genes associated with mitochondrial biogenesis (PGC-1α, PPARγ, and MFN2) and mitochondrial complex (ND4 and Cytb) were upregulated when the skeletal muscle cell line C2C12 was exposed to cysteine-cysteine chemokine ligand 4 (a ligand of CCR5) in vitro. These findings suggested that attenuation of endurance exercise performance is related to the loss of mitochondrial content and lower SDH activity of soleus muscle in CCR5 knockout mice. The present study provides evidence indicating that the chemokine receptor CCR5 might modulate the skeletal muscle metabolic energy system during exercise. © 2023

This paper proposes a discrete-time Model Reference Adaptive Controller (MRAC), applied to a Half-Bridge synchronous buck converter for charging batteries. The system uses an LCL filter that interfaces the power converter and the batteries. A virtual plant method is proposed for better transitory response of the adaptive control system. The model reference utilizes a reduced order, to simplify the project of the adaptive controller. Simulation results are presented for validation of the control project.

Designing biomimetic tissue models and developing scaffolds under cell-compatible protocols are a challenge in the field of tissue engineering. Cell encapsulation technique was selected with hydrogel fabrication due to its similarity to the condition within the human body. This study proposed SF-based hydrogels with the aim of application as cell encapsulation matrix. The vital properties, including gelation, diffusion, and cell viability were evaluated. SF/DMPG hydrogels exhibited rapid gelation and sufficient diffusion. L929 cells encapsulated in SF/DMPG survived after the fabrication process, with proliferation occurred at day 1 of culture which was done in static condition. The developed hydrogels showed potential application as a cell encapsulation matrix using tissue engineering, especially bioink.
