Background Dysregulated humoral immunity is a hallmark of primary Sjögren's Syndrome (pSS). This dysregulation involves aberrant B-lymphocyte activity resulting in abnormally high immunoglobulin G (IgG) levels and the production of autoantibodies, particularly those reactive with Ro/La ribonuclear complexes. pSS can affect almost any organ system, and current evidence-based therapies only offer some symptom relief, but no licensed therapy has been shown to alter the disease course. Nipocalimab is a high-affinity, fully human monoclonal antibody that reduces circulating IgG levels by selectively blocking the interactions of IgG, including pSS autoantibodies, with the neonatal Fc receptor (FcRn). Nipocalimab has previously induced rapid, safe, and durable serum IgG reductions in healthy volunteers ( NCT02828046 ) [1] and in autoantibody-driven generalized myasthenia gravis (gMG) in adults (Vivacity-MG; NCT03896295 ) [2] , suggesting that nipocalimab may treat a broad range of autoimmune disorders associated with autoantibodies, immune complexes and B-lymphocyte hyperactivity, including pSS. Objectives Here, we describe the key data from Vivacity-MG that illustrate the therapeutic potential of nipocalimab in IgG autoantibody-driven conditions and review the design of a phase 2 study evaluating the efficacy and safety of nipocalimab in patients with pSS ( NCT04968912 ). Methods Study feasibility assessments involved evaluating results from the phase 2 placebo-controlled trial Vivacity-MG. Of the 68 patients enrolled, 54 patients were randomized 1:1:1:1:1 to 4 treatment groups or a placebo group. Results In Vivacity-MG, there were no discontinuations due to treatment-emergent adverse events (TEAEs), severe AEs, or related serious AEs with nipocalimab. The incidence of infections and headaches with nipocalimab were comparable to placebo ( Table 1 ). The energetic and toxicological properties of N-propylnitroguanidine (PrNQ) are underassessment as a replacement for energetics in current use, such as hexahydro-1,3,5-trinitro1,3,5-triazine (RDX) and trinitrotoluene (TNT). This study evaluated the toxicity of PrNQ in several in vitro test systems, including the Ames mutagenicity assay, the Microtox Acute Toxicity Test System, and skin sensitization via the human cell line activation test (h-CLAT) and the direct peptide reactivity assay (DPRA). Data from the h-CLAT were also used to predict a Hazard Category (United Nations Economic Commission for Europe (UNECE), 2015) for acuteoral toxicity. Data from this study are used to assist in making environment and health-based decisions regarding the design and selection of formulas and materials for further development of new munition compounds.

Objectives: The purpose of this paper is to highlight the impact of a pandemic on the lives of vulnerable members of the community who have experienced or are ‘at risk’ of experiencing intimate family violence and child abuse in Saudi Arabia. By reviewing the experience in Saudi Arabia in the context of the international literature, the study explores similarities and differences in the impact of the pandemic on family violence. Methods: The study investigated the impact of the COVID-19 pandemic on family violence and child abuse in Saudi Arabia by conducting a comparative analysis of the prevalence of cases, types of abuse, and geographical location of those experiencing violence between the years 2019 and 2020. Data were obtained from the Family Violence Reporting Center 1919 in Saudi Arabia. Results: The study found that the risk of family violence and child abuse increased during the COVID-19 pandemic in Saudi Arabia, similar to findings in other countries. Conversely, there was a decrease in the number of reports due to factors associated with COVID-19. Conclusions: The study highlighted the importance of ensuring that existing policies and services are sufficient to ensure the risks arising from the impact of COVID-19 responses and minimize the negative consequences. © 2023 King Faisal Specialist Hospital & research Centre (General Organization), Saudi Arabia

The upper extent of a channel is a transition zone from the hillslope to the beginning of the stream channel. Accurately and consistently identifying the upper extent of a channel in the field and locating where hillslope processes transition to stream-channel processes can be a difficult task. Physical characteristics located at the beginning of a channel (i.e., channel head), including geomorphic, sediment, and vegetation indicators, can vary significantly across different landscapes in the United States. Remote tools are useful for examining the upper extent of channels, but these re-mote tools have limitations for identifying the beginning of channels. Even as the resolution of remote data continues to increase, field observations are necessary to validate the remote data on the ground and to accurately and consistently identify and locate the transition from the hillslope to the stream channel. Use of a combination of remote and field evidence is likely the most successful strategy for identifying channel heads. This report presents a case study that demonstrates how a weight-of-evidence approach can combine field and remote evidence to locate the different parts of the transition and ultimately to identify the channel-head location.

Analysis of sperm quality is still one of the most important tools for evaluating a man's fertility. Sperm quality includes several aspects including motility and viability. Mice (Mus musculus) are usually used as experimental animals because the reproductive system is almost the same as human. The purpose of this study is to determine the motility, viability, and type of charge of sperm mice (Mus muculus) through the study of exposure electric fields in an ex vivo condition. Observations were made in the time At for sperm without influence and influenced by an electric field. Mice sperm is diluted with a solution of Phosphate Buffered Saline (PBS). The liquid is inserted into the hemocytometer and then placed between two iron plates connected with a DC voltage source. The distance between the two plates d = 7.0 cm. The magnitude of the voltage used is Δv = 0 (control) and Δv = 12.5 V. Sperm movement was observed using a fluorescence microscope then recorded using NIS Elements software. Then the movement of sperm is analyzed using Tracker software. Sperm without an electric field (Δv = 0) has a viability of 60 minutes longer than sperm affected by an electric field (ΔV = 12.5 V) which has a viability of 30 minutes. The sperms move almost randomly in all directions without the influence of the electric field but tend to move to a positive plate when under the influence of the electric field. Based on the direction of the movement of mice sperm, the sperm is thought to be negatively charged. From the results of this study, it was found that the fast zig-zag motion had a velocity v > 37 μm/s. © Published under licence by IOP Publishing Ltd.

This paper presents a method which detects faults in the measurement required for navigation of an unmanned surface vehicle. The measurement includes acceleration and angular rate by an inertial measurement unit (IMU), and the location by global navigation satellite system (GNSS). The method uses Kalman filter to estimate the state variables which represent the amount of faulty measurements. The method is tested by the experiment in which a boat equipped by commercial IMU and GNSS navigates in a sea trial. The result verifies that faults in acceleration and angular rate measurements are detected robustly and the GNSS position fault is detected when there is considerable location error.

Diabetes mellitus is one of the most pressing health concerns because so many people are afflicted by its disabling symptoms. Factors such as age, excess body fat, insufficient physical activity, a history of diabetes in one's family, a sedentary lifestyle, an unhealthy diet, hypertension, etc., all increase the likelihood of developing diabetes mellitus. Health complications are more common in people with diabetes, including cardiovascular disease, renal failure, stroke, blindness, and nerve injury. To validate a diagnosis of diabetes, hospitals typically perform a battery of procedures on the patient. Big data analytics has many vital applications in the healthcare sector. Numerous large computer systems are used in the healthcare sector. With the help of big data analytics, researchers can sift through mountains of data in search of previously unseen patterns and insights. Current techniques have a poor degree of precision in classification and forecast. While previous research has focused on factors such as glucose, body mass index, age, insulin, etc., the proposed model takes these into account and also the other factors that may be more relevant to the development of diabetes. The newer sample is superior to the older one based on categorization accuracy. A workflow algorithm for diabetes prognosis is also required to improve the accuracy. © 2023 IEEE.

A key component of microbial communication, autoinducer-2 (AI-2) signaling, affects several physiological processes, including environmental adaptation and biofilm formation in lactic acid bacteria (LAB). The multifarious contribution of AI-2, synthesized by LuxS, in improving biofilms and tolerance to hostile conditions in LAB has been investigated in this review. The evolutionary conservation and diversity of AI-2 are shown by a phylogenetic analysis of luxS gene among several LAB species. Furthermore, AI-2 signaling in LAB improves resistance to unfavorable environmental factors, including pH fluctuations, temperature extremes, and antimicrobial agents. Lactic acid bacteria could set off defenses against harmful impacts from environmental stresses. © 2025 Institut Pasteur
