BACKGROUND:The rapid emergence of highly transmissible and immune-evasive SARS-CoV-2 variants has required the reformulation of COVID-19 vaccines to target these evolving threats. Although previous infections and booster vaccinations can boost variant neutralisation, it remains uncertain whether monovalent vaccines-delivered via mRNA or protein-based platforms-can trigger novel B-cell responses specific to omicron XBB.1.5 variants. We sought to address this uncertainty by characterising the antibody repertoire of individuals receiving a monovalent booster vaccine. METHODS:In this observational study, we analysed the genetic antibody repertoire of 603 individual plasmablasts from five individuals (recruited at the Icahn School of Medicine at Mount Sinai, New York, NY, USA, from STUDY-16-01215/IRB-16-00971 and STUDY-20-00442/IRB-20-03374) vaccinated with a monovalent XBB.1.5 vaccine, either through mRNA (Moderna or Pfizer-BioNTech; participants 1, 2, and 3) or adjuvanted protein (Novavax; participants 4 and 5) platforms. Before XBB.1.5 booster vaccination, all participants received mRNA-based priming and booster vaccine with ancestral SARS-CoV-2 and four of the five participants had a breakthrough omicron variant infection. We expressed 100 human monoclonal antibodies (mAbs; 50 from participants 1, 2, and 3, and 50 from participants 4 and 5) and evaluated their binding and neutralisation against various SARS-CoV-2 variants, including JN.1. We then selected four mAbs for in-vivo protection experiments by passive immunisation and viral challenge, and cryo-electron microscopy with two selected mAbs complexed with the XBB.1.5 spike (S) protein to determine their structures and binding interactions. FINDINGS:Between October and November, 2023, we enrolled three male and two female participants (mean age 46 years) all of whom were White. The Large-Sized Telescopes (LSTs) of Cherenkov Telescope Array (CTA) are designed for gamma-ray studies focusing on low energy threshold, high flux sensitivity, rapid telescope repositioning speed and a large field of view. Once the CTA array is complete, the LSTs will be dominating the CTA performance between 20 GeV and 150 GeV. During most of the CTA Observatory construction phase, however, the LSTs will be dominating the array performance until several TeVs. In this presentation we will report on the status of the LST-1 telescope inaugurated in La Palma, Canary islands, Spain in 2018. We will show the progress of the telescope commissioning, compare the expectations with the achieved performance, and give a glance of the first physics results. © Copyright owned by the author(s) under the terms of the Creative Commons.

With the increasing prevalence of age-related chronic diseases burdening healthcare systems, there is a pressing need for innovative management strategies. Our study focuses on the gut microbiota, essential for metabolic, nutritional, and immune functions, which undergoes significant changes with aging. These changes can impair intestinal function, leading to altered microbial diversity and composition that potentially influence health outcomes and disease progression. Using advanced metagenomic sequencing, we explore the potential of personalized probiotic supplements in 297 older adults by analyzing their gut microbiota. We identified distinctive Lactobacillus and Bifidobacterium signatures in the gut microbiota of older adults, revealing probiotic patterns associated with various population characteristics, microbial compositions, cognitive functions, and neuroimaging results. These insights suggest that tailored probiotic supplements, designed to match individual probiotic profile, could offer an innovative method for addressing age-related diseases and functional declines. Our findings enhance the existing evidence base for probiotic use among older adults, highlighting the opportunity to create more targeted and effective probiotic strategies. However, additional research is required to validate our results and further assess the impact of precision probiotics on aging populations. Future studies should employ longitudinal designs and larger cohorts to conclusively demonstrate the benefits of tailored probiotic treatments.

Flood has caused significant damage to the agricultural sector, which results in low productivity and causes famine in Kaba-kama community of Basse in the Upper river region of the Gambia. This study analyses the potential role of the community's mitigation efforts to reduce the flood impact in the Gambia. The objective is to determine the influence of community-based intervention activities in reducing the flood impact at the community level. The primary data are obtained in this study from own previous experience by participating in the community work ('Te-setto') to mitigate the impact of flood, and interviews in a form of questionnaires to the members of the VDC in Kaba-kama community. Literature reviews from journals, books, and the final report about the disaster in 2018 by NDMA has served as a secondary source of data for the study. Notifiable, the community-based organization (Village Development Committee -VDC), is the key pioneer of the initiative. This study exemplifies the successful collective efforts by the communities and government in different regions and the country at large in practicing flood adaptive and mitigation strategies, which has potential to be implemented elsewhere in other famine-prone countries in the world.

Recently, several gamma-ray bursts (GRBs) have been detected in the very-high-energy (VHE) gamma-ray energy range by ground-based gamma-ray experiments such as MAGIC, H.E.S.S., and LHASSO. For some GRBs, the VHE emission is consistent with synchrotron self-Compton (SSC) emission from high-energy electrons accelerated in the forward shock of the relativistic jet. However, more statistics are needed to further constrain the emission models. GRB 201216C is a long bright GRB detected in a broad energy range from radio to VHE. The redshift is estimated to be 1.1, making the GRB the most distant source detected in the VHE energy range. MAGIC started the observation 56 seconds after the GRB was triggered by the Swift-BAT telescope. We performed a detailed analysis and detected the signal with about 6 sigma in the first 20 minutes. MAGIC continued the observation for 2.2 hours on the same night and 4 hours on the next night. No signal was detected later than 40 minutes after the GRB trigger. We have performed modelling of the multi-wavelength emission using the MAGIC data. We analysed simultaneous optical data from Liverpool Telescope with MAGIC and included the results in the modelling. The sub-TeV emission is consistent with the single-zone SSC model in the forward shock. In this presentation, we show the final results of the MAGIC data analysis of GRB 201216C and discuss the emission mechanism of the multi-wavelength data. © Copyright owned by the author(s) under the terms of the Creative Commons.

BACKGROUND:Lytic Epstein-Barr virus (EBV) infection plays a major role in the pathogenesis of nasopharyngeal carcinoma (NPC). For patients with recurrent or metastatic NPC and resistant to conventional therapies, adoptive cell therapy using EBV-specific cytotoxic T cells (EBV-CTLs) is a promising option. However, the long production period (around 3 to 4 weeks) and low EBV-CTL purity (approximately 40% of total CD8 T cells) in the cell product limits the application of EBV-CTLs in clinics. Thus, this study aimed to establish a protocol for the rapid production of EBV-CTLs. METHODS:By culturing peripheral blood mononuclear cells (PBMCs) from EBV-seropositive donors with EBV-specific peptides and interleukin (IL)-2, IL-15, and interferon α (IFN-α) for 9 days, we identified that IL-15 can enhance IL-2-mediated CTL activation and significantly increase the yield of CTLs. RESULTS:When IFN-α was used in IL-2/IL-15-mediated CTL production from days 0 to 6, the productivity of EBV-CTLs and EBV-specific cytotoxicity significantly were reinforced relative to EBV-CTLs from IL-2/IL-15 treatment. Additionally, IFN-α-induced production improvement of virus-specific CTLs was not only the case for EBV-CTLs but also for cytomegalovirus-specific CTLs. CONCLUSION:We established a novel protocol to rapidly expand highly pure EBV-CTLs from PBMCs, which can produce EBV-CTLs in 9 days and does not require feeder cells during cultivation.

Machine learning is a modern approach to problem-solving and task automation. In particular, machine learning is concerned with the development and applications of algorithms that can recognize patterns in data and use them for predictive modeling. Artificial neural networks are a particular class of machine learning algorithms and models that evolved into what is now described as deep learning. Given the computational advances made in the last decade, deep learning can now be applied to massive data sets and in innumerable contexts. Therefore, deep learning has become its own subfield of machine learning. In the context of biological research, it has been increasingly used to derive novel insights from high-dimensional biological data. To make the biological applications of deep learning more accessible to scientists who have some experience with machine learning, we solicited input from a community of researchers with varied biological and deep learning interests. These individuals collaboratively contributed to this manuscript's writing using the GitHub version control platform and the Manubot manuscript generation toolset. The goal was to articulate a practical, accessible, and concise set of guidelines and suggestions to follow when using deep learning. In the course of our discussions, several themes became clear: the importance of understanding and applying machine learning fundamentals as a baseline for utilizing deep learning, the necessity for extensive model comparisons with careful evaluation, and the need for critical thought in interpreting results generated by deep learning, among others. © 2021, CC BY.

Background: Pruritus is the most common initial symptom reported by patients with dermatomyositis (DM) and polymyositis (PM). However, there is limited data regarding the impact of pruritus on cancer and mortality in patients with DM and PM. In this study, we aimed to investigate how pruritus is associated with cancer and mortality in patients with DM and PM. Methods: This nationwide, population-based retrospective cohort study included adult DM and PM patients from Taiwan’s National Health Insurance research Database between 2005 and 2022. Sex- and age-matched pruritic patients, identified by over 6 weeks of antipruritic medication use, and nonpruritic patients were analyzed. The primary outcome was cancer occurrence or all-cause mortality. The association between pruritus and these outcomes was estimated using Cox proportional hazards models. Results: Among 919 matched pairs of pruritic and nonpruritic patients, cancer was observed in 19.96% in the long-term pruritic group (LPG), 14.63% in the short-term pruritic group (SPG), and 10.34% in the non-pruritic group (NPG) ( p < 0.0001). All-cause mortality was documented as 30.37% in the LPG, 29.69% in the SPG, and 37.76% in the NPG ( p < 0.0001). After adjusting for sex, age, and other comorbidities, pruritus was associated with an increased risk of cancer (hazard ratio (HR) 1.708, 95% confidence interval (CI) 1.229–2.374) and a lower risk of all-cause mortality (HR 0.483, 95% CI 0.409–0.569). Conclusion: This population-based study revealed that pruritus appeared to be associated with increased risks of cancer and decreased all-cause mortality. Thus, pruritus may serve as a pragmatic factor for risk stratification and tailored treatment strategies in DM and PM. Cancer screening, particularly for nasopharyngeal and breast cancers in East Asian populations, is recommended for patients with DM or PM, especially those presenting with pruritus. The Large-Sized Telescope (LST) prototype of the future Cherenkov Telescope Array (CTA) is located at the Northern site of CTA, on the Canary Island of La Palma. It is designed to provide optimal performance in the lowest part of the energy range covered by CTA, observing gamma rays down to energies of tens of GeV. The LST prototype started performing astronomical observations in November 2019 during the commissioning of the telescope and it has been taking data since then. In this contribution, we will present the tuning of the characteristics of the telescope in the Monte Carlo (MC) simulations to describe the data obtained, the estimation of its angular and energy resolution, and an evaluation of its sensitivity, both with simulations and with observations of the Crab Nebula. © Copyright owned by the author(s) under the terms of the Creative Commons Attribution-NonCommercial-NoDerivatives 4.0 International License (CC BY-NC-ND 4.0)
