Metabolomic analysis can indeed enhance the prime variable dataset for the monitoring of perfusion cultures by providing a higher resolution view of the metabolic state. Metabolic profiles can capture physiological state shifts over the course of the perfusion cultures and indicate a metabolic “signature” of the phase transitions, which is not observable from prime variable data. Notably, metabolomics provides orthogonal (to prime variables) evidence that all cultures follow this same metabolic state shift with cell age, independently of bioreactor scale. Additionally, this analysis can increase the information content of process development experiments by helping better understand the impact of changes in bioreactor operating conditions on cell physiology. In this context, metabolic profiling could be integrated into the monitoring of cell physiology in perfusion cultures.

Background: Antidepressants are among the most commonly prescribed psychotropic agents for older patients. Little is known about the best source of pharmacotherapy information to consult about key factors necessary to safely prescribe these medications to older patients. Objective: The objective of this study was to synthesize and contrast information in the package insert (PI) with information found in the scientific literature about age-related changes of antidepressants in systemic clearance and potential pharmacokinetic drug-drug interactions (DDIs). Methods: A comprehensive search of two databases (MEDLINE and EMBASE from January 1, 1975 to September 30, 2011) with the use of a combination of search terms (antidepressants, pharmacokinetics, and drug interactions) was conducted to identify relevant English language articles. This information was independently reviewed by two researchers and synthesized into tables. These same two researchers examined the most up-to-date PIs for the 26 agents available at the time of the study to abstract quantitative information about age-related decline in systemic clearance and potential DDIs. The agreement between the two information sources was tested with κ statistics. Results: The literature reported age-related clearance changes for 13 antidepressants, whereas the PIs only had evidence about 4 antidepressants (κ < 0.4). Similarly, the literature identified 45 medications that could potentially interact with a specific antidepressant, whereas the PIs only provided evidence about 12 potential medication-antidepressant DDIs (κ < 0.4). Conclusion: The evidence-based literature compared with PIs is the most complete pharmacotherapy information source about both age-related clearance changes and pharmacokinetic DDIs with antidepressants. Future rigorously designed observational studies are needed to examine the combined risk of antidepressants with age-related decline in clearance and potential DDIs on important health outcomes

Cardiorespiratory fitness level correlates inversely with excess post-exercise oxygen consumption after aerobic-type interval training

Background: The purpose of this study was to reveal any association between cardiorespiratory fitness level and excess post-exercise oxygen consumption (EPOC) using three cycling protocols with varying degrees of exercise intensity, i.e., sprint interval training (SIT), high-intensity interval aerobic training (HIAT), and continuous aerobic training (CAT). Findings. Ten healthy men, aged 20 to 31 years, attended a cross-over experiment and completed three exercise sessions: SIT consisting of 7 sets of 30-s cycling at 120% VO₂ with a 15-s rest between sets;HIAT consisting of 3 sets of 3-min cycling at 80∼90% VO₂ with a 2-min active rest at 50% VO₂ between sets;and CAT consisting of 40 min of cycling at 60∼65% VO₂. During each session, resting VO₂, exercise VO₂, and a 180-min post-exercise VO₂ were measured. The net exercise VO ₂ during the SIT, HIAT, and CAT averaged 14.7 ± 1.5, 31.8 ± 4.1, and 71.1 ± 10.0 L, and the EPOCs averaged 6.8 ± 4.0, 4.5 ± 3.3, and 2.9 ± 2.8 L, respectively. The EPOC with SIT was greater than with CAT (P < 0.01) and HIAT (P = 0.12). Correlation coefficients obtained between subjects' VO₂ and the ratio of EPOC to net exercise VO₂ for SIT, HIAT, and CAT were -0.61 (P = 0.06), -0.79 (P < 0.01), and -0.42 (P = 0.23), respectively. Conclusions: Our data suggest that cardiorespiratory fitness level correlates negatively with the magnitude of EPOC, especially when performing aerobic-type interval training. © 2012 Matsuo et al.;licensee BioMed Central Ltd.

Background: Understanding how to successfully deal with human and technical aspects involved in Software Process Improvement (SPI) programs is a challenging issue. Technical aspects are not enough to guarantee the success of activities. Human factors (e.g., experiences, opinions and perceptions) impact the effectiveness of SPI programs. Aim: this paper aims to improve our current understanding on how human aspects can influence SPI programs from the point of view of the professionals involved in such efforts. We compare our findings with the results of previous studies in order to analyze the different contexts in which human aspects influence SPI. Method: We conducted a qualitative study in the context of small companies involved in SPI programs in Amazonas State in Brazil based on the Brazilian Maturity Model - ***. We used semi-structural interviews with software engineers. Results: We identified findings about eleven of the fourteen investigated human aspects. We identified new characteristics that motivate actors to get involved in SPI programs. Finally, while previous research indicated that the process of individual decision making as a negative aspect, we identified this as a positive aspect. Conclusions: Despite the differences in companies' size and maturity models, our findings corroborate many of the important results of previous case studies. The consistency of the presented results with previous research in different organizations, teams, and countries suggest that these results are true for software engineering in general. However, there were differences related to the motivating factors and the perception of the individual decision making.
