Posttranslational modifications (PTMs) of histone proteins, such as acetylation, methylation, phosphorylation, and ubiquitylation, play essential roles in regulating chromatin dynamics. Combinations of different modifications on the histone proteins, termed ‘histone code’ in many cases, extend the information potential of the genetic code by regulating DNA at the epigenetic level. Many PTMs occur on non-histone proteins as well as histones, regulating protein–protein interactions, stability, localization, and/or enzymatic activities of proteins involved in diverse cellular processes. Although protein phosphorylation, ubiquitylation, and acetylation have been extensively studied, only a few proteins other than histones have been reported that can be modified by lysine methylation. This review summarizes the current progress on lysine methylation of non-histone proteins, and we propose that lysine methylation, like phosphorylation and acetylation, is a common PTM that regulates proteins in diverse cellular processes.

Rac GTPase signaling in mechanotransduction during embryonic morphogenesis

How cells sense and respond to mechanical forces is attracting considerable attention. We recently demonstrated that mechanical tension originating from one tissue strongly influences the differentiation and morphogenesis of another tissue during C. elegans embryogenesis (Nature 471: 99-103). Specifically, we found that the repeated contractions of muscle cells stimulate a signaling cascade involving the Rac GTPase within the epidermis. This pathway ultimately leads to strengthen hemidesmosome-like junctions and promote embryonic morphogenesis. Our work provides further evidence that mechanical inputs impact on development, much like inputs involving growth factors and morphogens. After briefly outlining the pioneering work that inspired us, I will present the mechanotransduction process underlying the response to tension and the key experiments supporting our conclusions. © 2011 Landes Bioscience.

A Drosophila model for congenital heart disease

Congenital heart disease (CHD) represents a major cause for perinatal death and childhood morbidity. Understanding the molecular-genetic basis of CHD is therefore crucial for improved prenatal diagnosis and optimal therapeutic strategies. The power of Drosophila genetics has been utilized in recent years to identify new factors involved in human disease in general and CHD in particular. This review summarizes current approaches that aim to dissect models of CHD using classical genetics together with a multitude of newly developed methods for cardiac analysis in the fruit fly Drosophila. © 2009.

stem Cell Mobilization and Egress of AML Blasts from the Bone Marrow Are Both Regulated by MT-MMP-Mediated Activation of MMP-2.

The cell-surface proteolytic enzymes membrane-type (MT)-matrix metalloproteinases (MMPs) activate secreted latent forms of MMPs and play a key role in cell migration and tumor cell invasion and metastasis. Previously we reported that mobilized peripheral blood (mPB) CD34 + cells as well as AML blasts secrete inactive (pro)MMP-2, in contrast to normal steady-state bone marrow (BM) CD34 + cells which do not. In this study we hypothesized that the egress from BM of normal hematopoietic stem/progenitor cells during G-CSF-induced mobilization or of AML blasts shares a common mechanism that involves activation of secreted proMMP-2 by MT-MMPs. We evaluated the expression of MT-MMPs (MT1-MMP, MT2-, MT3- MT4-, MT5-, MT6-) in normal and leukemic hematopoietic cells, as well as in BM stromal cells, and determined their role in proMMP-2 activation and migration. We found MT1-MMP expression (mRNA and protein) in mPB CD34 + cells and in the majority of AML samples (from 21 out of 26 patients) and upregulation of MT1-MMP by G-CSF in steady-state BM CD34 + cells. Moreover, we confirmed the secretion of proMMP-2 in media conditioned by hematopoietic (mPB CD34 + , AML) cells and stromal cells. However, we detected the active form of MMP-2 in co-cultures of the hematopoietic (mPB CD34 + , AML) cells with stromal cells, as well as in co-cultures of steady-state BM CD34 + cells stimulated with G-CSF with stroma. In cultures of stromal cells alone G-CSF had no effect on the expression of either MT1-MMP or MMP-2. To evaluate the role of MT-MMPs in proMMP activation, we examined the leukemic KG-1 cell line, which we found not to express any MT-MMPs, and did not detect active MMP-2 in co-cultures with stroma. On the other hand, primary AML samples that did not express MT1-MMP but expressed MT2-, MT4- or MT5-MMPs activated proMMP-2 in co-cultures with stroma. The organization of the chromatin structure is essential for maintaining cell-type-specific gene expression and therefore for cell identity. This structure is highly dynamic and is regulated by a large number of chromatin-associated proteins that are required for normal development and differentiation. Recurrent somatic mutations have been found with high frequency in genes coding for chromatin-associated proteins in cancer, and several of these are required for cancer maintenance. In this review, we discuss recent advances in understanding the role of chromatin-associated proteins in transcription, development, and cancer. Specifically, we focus on selected examples of proteins belonging to the histone methyltransferase, histone demethylase, or bromodomain families, for which specific small molecule inhibitors have been developed and are in either preclinical or clinical trials.

This book focuses on natural products, in particular medicinal plants and their derived products, as an indispensable source of bioactive molecules that serve as either drug candidates or lead compounds for drug design and discovery . There are several advantages for plant-derived therapeutics, including wide availability, diverse pharmacological actions, and a generally good profile of safety and tolerability. Over the recent years, there have been numerous reports from clinical studies testifying the efficacy and safety of medicinal plants and phytochemicals in treating human diseases. A plethora of basic studies has also unraveled molecular mechanisms underlying the health benefits of herbal medicines. The goal is to present the readers a carefully curated collection of plant-derived natural products and their underlying molecular mechanisms.
