One of the striking features during the stockpile formation is the internal segregation patterns such as stratification developed due to particle size or shape difference. This study employs the discrete element method to investigate the influencing factors and segregation-triggering mechanisms in piling process. The results reveal that the percolation and void-filling mechanism distinguishes the motions of particles with different sizes. This mechanism causes large particles to flow over the pile surface and segregate at the bottom, propelling small particles to fill the voids between large particles, resulting in segregated layers. Consequently, this disparity in particle motion gives rise to stratification patterns. A larger size ratio and a relatively smaller mass ratio can induce stronger segregation. For mixtures of ellipsoids, the segregation weakens due to their poor flowability. The stratification patterns still exist at smaller scales for ellipsoids, and the triggering mechanism is based on the “static avalanche” instead of void-filling.

The Plasmodium falciparum cysteine protease falcipain-2(FP-2) is an attractive antimalarial target. Here, we discovered that the natural compound NP1024 is a nonpeptidic inhibitor of FP-2 with an IC50 value of 0.44 μmol L–1. The most exciting finding is that both in vitro and in vivo, NP1024 directly targets FP-2 in malaria parasite-infected erythrocytes as a natural fluorescent probe, thereby paving the way for an integration of malaria diagnosis and treatment.
