Cortical spreading depression (CSD) is a slowly propagating wave of depolarization of brain cells, followed by temporary silenced electrical brain activity. Major structural changes during CSD are linked to neuronal and possibly glial swelling. However, basic questions still remain unanswered. In particular, there are open questions regarding whether neurons or glial cells swell more, and how the cellular swelling affects the CSD wave *** this study, we computationally explore how different parameters affect the swelling of neurons and astrocytes (star-shaped glial cells) during CSD and how the cell swelling alters the CSD wave spatial distribution. We apply a homogenized mathematical model that describes electrodiffusion in the intraand extracellular space, and discretize the equations using a finite element method. The simulations are run with a two-compartment (extracellular space and neurons) and a three-compartment version of the model with astrocytes added. We consider cell swelling during CSD in four scenarios: (A) incorporating aquaporin-4 channels in the astrocytic membrane, (B) increasing the neuron/astrocyte ratio to 2:1, (C) blocking and increasing the Na + /K + -ATPase rate in the astrocytic compartment, and (D) blocking the Cl - channels in astrocytes. Our results show that increasing the water permeability in the astrocytes results in a higher astrocytic swelling and a lower neuronal swelling than in the default case. Further, elevated neuronal density increases the swelling in both neurons and astrocytes. Blocking the Na + /K + -ATPase in the astrocytes leads to an increased wave width and swelling in both compartments, which instead decreases when the pump rate is raised. Blocking the Cl - channels in the astrocytes results in neuronal swelling, and a shrinkage in the astrocytes. Poly-(carbonate-co-ether) was synthesized via catalytic polymerization between epoxidized linseed oil (ELO), propylene oxide (PO) and carbon dioxide (CO2) using heterogeneous catalyst namely Co-Zn double metal cyanide (Co-Zn DMC). The influence of polymerization time was studied. It was revealed that the properties of the resultant polymers were affected by the polymerization time. Within the range of polymerization time studied, the maximum average molecular weight (Mn) of the resultant polymer obtained was 6.21 105 g⋅mol-1 with polydispersity index (PDI) of 1.05 at reaction time of 24 hours. © Published under licence by IOP Publishing Ltd.

Aptamers are oligonucleotides and peptides with short and medium length around 15-100 nucleotides or amino acids and a molecule of interests due to its specific binding affinities to the vast array of target molecules such as ions, complex proteins and antigens up to cellular surface and cell organelles. Aptamers shows high potential of application in therapeutics and diagnostics. Aptamers are usually obtained through rigorous in-vitro screening procedures known as Systematic Evolution of Ligands by EXponential enrichment (SELEX). There are growing interests in the aptamer screening approach through computational methods such as using molecular docking and molecular modelling (MD) simulations. The approach had been increasingly popular due to minimal use of chemical and reagents, balanced with the reasonable computational costs. In-silico approaches were able to poke the details of aptamer-ligand interactions which is hard to elucidate experimentally. The authors review the successful works in screening and analysing aptamer-target complexes interactions and the effort to design the aptamers through computational simulations. The own personal experience of authors in designing a specific aptamer for Hepatitis B surface antigen through computational screening method was also discussed. In conclusion, the advancement of computational chemistry provides a promising future for the aptamers research.

Evaluation on Quality Attributes of Pectin Coated - Cassava Chips

Deep fried-based food is commonly consumed in Malaysia due to their tasty and good appearance of the food. However, high consumption of the food may lead to adverse effect to consumer’s health beside high usage of oil-palm based cooking oil in deep frying process. Thus, this research aimed to investigate the effect of edible coating using different pectin concentration (1%, 2% or 3%) on cassava chips. Coated-cassava were prepared through dipping method into pectin and calcium chloride solution. Series of analysis such as lipid determination, texture analysis, colour determination, FTIR analysis, sensory evaluation and statistical analysis were conducted. Highest pectin concentration which 3% pectin-coated chips proved to reduce highest lipid uptake up to 11.750 ± 0.076 compared to uncoated chips (52.360 ± 0.762). For colour determination, it showed that the pectin coating did not changed the initial colour of the chips and enhanced the colour brightness after frying. Between all samples, the control or uncoated chips has the lowest result for hardness which was 577.216 ± 95.194 while 3% pectin coated chips had highest values in term of hardness which were 1338.209 ± 246.900. FTIR was able to recognize presence of the (N-O) group in all coated cassava chips compared to uncoated chips. The uncoated and pectin-coated chips had no significance different in overall acceptance in sensory analysis. This research concluded that edible coating from pectin with 3% concentration was able to reduce lipid uptake and enhanced several characteristics in cassava chips. Newly developed value-added coated cassava chips would offer a healthy, tasty and better appearance snack to consumer.

Municipal wastewater treatment plant generates a large amount of sludge each year with a generation rate of up to 30.8 kg per population equivalent per year. These sewage sludge cakes contain an abundant amount of lipids that can be extracted, and their compound can be utilized in the production of value-added products. The separation of lipids subsequently reduces the volume of sludge to be managed and disposed of. Lipids can be separated using the conventional Soxhlet extractor, and the yield depends on process parameters such as solvent to sludge ratio, temperature, extraction time, and particle size. Among these parameters, particle size was scarcely studied while extraction time shows an insignificant effect on lipids yield. Therefore, this paper focuses on the impact of the particle size as effective extraction size as well as significant extraction parameters; temperature and sludge-to-solvent ratio on the lipids yield of extraction from the sewage sludge cake using methanol as solvent. Four sludge particle size ranges (x) were selected as the effective extraction size (x≤0.425mm, 0.4254.00mm), five solvents to sludge ratio (7, 8, 10, 13, and 20mL/g) and solvent temperature ranging from 60 to 90℃ were studied with the percentage of lipids yield as the response. The results show that the fine treatment region of x≤2.00mm resulted in higher lipids output compared to the coarse treatment region of x>2.00mm. Dermatophytes are responsible for a majority of fungal infections in humans and other vertebrates, causing dermatophytosis. Treatment failures are often associated with biofilm formation, making dermatophytes resistant to antifungals. In this study, effects of a rhamnolipid (RL-SS14) produced by Pseudomonas aeruginosa SS14 on planktonic cells of Trichophyton rubrum and Trichophyton mentagrophytes, their biofilm formation, and disruption of mature biofilms were assessed. The composition of RL-SS14 was analysed using FTIR, HPLC-ESI-MS, and GC–MS. Minimum inhibitory concentrations against the planktonic forms of T. rubrum and T. mentagrophytes were 0.5 mg/mL and 0.125 mg/mL, respectively. Crystal-violet (biofilm biomass) and safranin (extracellular matrix) staining revealed that RL-SS14 significantly inhibited biofilm formation and also reduced preformed biofilms in a dose-dependent manner. Microscopic visualization of treated biofilms via SEM, AFM, and CLSM revealed marked morphological damage, cell death, and reduced extracellular matrix. The results indicate the potential of RL-SS14 as an anti-biofilm agent against dermatophytes. © 2020 The Authors

Malaysia produces a very high amount of oil palm biomass especially the oil palm empty fruit bunch (OPEFB) every year. The OPEFB in reality can be converted into value-added panels such as particleboard. The application may vary such as use for furniture, indoor building materials, door, window, floor, office partition and many more. Normally, the commercial particleboard were made from biomass with addition of formaldehyde based resins and it is harmful to the environment due to the formaldehyde released. In this study, the formaldehyde based resins were excluded and the binderless particleboard made from OPEFB with addition of natural binder such as corn, potato, tapioca and wheat starch were evaluated. These panels were manufactured with a target density of 0.8 g/cm 3 . The hot pressing temperature, hot pressing time, and pressing pressure used were 180 °C, 20 min, and 12 MPa respectively. From the results, the highest modulus of rupture (MOR) and modulus of elasticity (MOE) were found to be the panel with the addition of potato starch as the natural binder (15.33 MPa, 2078 MPa) following by wheat (14.41 MPa, 1823 MPa), tapioca (14.18 MPa, 1771 MPa) and corn starch (12.59 MPa, 1335 MPa) respectively. The highest dimensional stability was also found to be the panel with the addition of potato starch with the lowest water absorption (69%) and thickness swelling (50%) value. Overall results has met the Japanese Industrial Standard (JIS) for type-8 and type-13 except for water absorption and thickness swelling. Therefore, particleboard made from OPEFB without using any synthetic resin was successfully made with the addition of natural binder such as potato, wheat, tapioca and corn starch and probably can be replace as a substitute for conventional particleboard in future.
