Homozygous human embryonic stemcells (hESCs) are thought to be better cell sources for hESC banking because their human leukocyte antigen (HLA) haplotype would strongly increase the degree of matching for certain populations with relatively smaller cohorts of cell lines. Homozygous hESCs can be generated from parthenogenetic embryos, but only heterozygous hESCs have been established using the current strategy to artificially activate the oocyte without second polar body extrusion. Here we report the first successful derivation of a human homozygous ESC line (chHES-32) from a one-pronuclear oocyte following routine in vitro fertilization treatment. cAHES-32 cells express common markers and genes with normal hESCs. They have been propagated in an undifferentiated state for more than a year (>P50) and have maintained a stable karyotype of 46, XX. When differentiated in vivo and in vitro, c/zHES-32 cells can form derivatives from all three embryonic germ layers. The almost undetectable expression of five paternally expressed imprinted genes and their HLA genotype identical to the oocyte donor indicated their parthenogenetic origin. Using genome-wide single-nucleotide polymorphism analysis and DNA fingerprinting, the homozygosity of c/zHES-32 cells was further confirmed. The results indicated that 'unwanted' one-pronuclear oocytes might be a potential source for human homozygous and parthenogenetic ESCs, and suggested an alternative strategy for obtaining homozygous hESC lines from parthenogenetic haploid oocytes.

Assuming homogenity of the underlying statistics, interspike interval probability density functions recorded from behaving macaque monkeys are determined with a special statistical fitting method. The nature of the problem needed robust statistics approach, the results were validated with the Kolmogorov-Smirnov test. The results suggest that there is no single/universal interspike interval probability density function, but many, and the underlying statistics is definitely not Poissonian.

We have developed a direct conjugation method of ROD (Arg-Gly-Asp) into polylactide (PLA) using co-solvent system, which showed high reaction yield compared to one solvent system presumably due to good solubility of reactants. First, hydroxyl groups at the end of PLA were changed into carboxyl acid by ring opening reaction of succinic anhydride. For a coupling reaction with high solubility, ROD was dissolved in water and then PLA in dioxane was added into ROD solution in the presence of 1-(3-dimenthylaminopropyl)3-ethylcarbodiimide hydrochloride (EDC). The conjugation of ROD was confirmed by 1H-NMR, GPC and elementary analysis. The ROD conjugated PLA (RGD-PLA) on poly(lactide-co-glycolide) (PLGA) film significantly increased the number of attached cells compared to PLGA without RGD-PLA, while it did not change static contact angle of PLGA. This result demonstrated this conjugation method would be a useful tool to increase cell affinity of biomaterials without change in hydrophilic/hydrophobic property for tissue engineering applications.
