This research showed the difference of thermocline layer depth during La Niña and El Niño events in the Makassar Strait, the main passage connecting the Pacific and Indian Oceans. La Niña event represented by 2011 and El Niño event represented by 2015. Three-dimensional numerical modeling using Regional Ocean Modeling System (ROMS) is used to determine ocean dynamic characteristics. The tides and atmospheric factor used as generating forces for coupled Ocean-Atmosphere-Wave-Sediment simulation model. Tidal elevation verification results from January to February 2017 showed a high confidence with RMSE = 0.148 m and MAPE = 13.51 %. Ocean circulation showed that water flows from north to south of the Makassar Strait in both conditions, El Niño and La Niña, indicating a transport from the Pacific to the Indian Oceans during those time. Two-dimensional seasonal sea surface temperature (SST) was used to determine the variability of thermocline layer depth. In general, based on simulation results, the depth of thermocline during La Niña was deeper than that during El Niño. The upper threshold of thermocline layer during El Niño phenomenon was shallower (average 27.29 m) compare to the threshold during La Niña (average 39.25 m). So do the lower threshold, during El Niño was shallower (average 160.47 m) compare to the threshold during La Niña (average 165.70 m). The thermocline thickness during El Niño (133.19 m) was found to be thicker compared to La Niña (126.43 m). © Published under licence by IOP Publishing Ltd.

A problem-based mathematic learning model with high validity, practicality, and effectiveness is produced to improve students' competence. The stages on developing it consist of 1) preliminary stage, 2) proto-type stage including planning, evaluation, and revision, 3) product assessment stage. The mathematic learning model based on problem was tested to the college students of PGSD Department of FKIP, Bung Hatta University involving 30 students and 3 mathematic lecturers. The instrument used in this study consisted of 1) instrument of validity appraisal, component of learning model and toolkit, 2) instrument of practicality appraisal from students and lecturers, 3) effectiveness instrument covering test result and students' appreciation appraisal toward problem-based mathematic learning model. The result showed that problem-based mathematic learning model including syntax/learning steps, along with learning tools in term of semester learning plan and students' worksheet was considered valid, practical, and effective. © 2020 Published under licence by IOP Publishing Ltd.

This research has a purpose to develop Bernoulli Mixture model for Bidikmisi data modelling using Bayesian approach. Model development is done by considering the specificity in the data acceptance of Bidikmisi scholarship prototype in East Java Province. Bidikmisi acceptance status having a binary type (0 and 1) coupled with the main criteria factor of parent income and the number of dependents family produces a structure of Bernoulli mixture distribution with two components. The characteristics of each component can be identified through the Bernoulli Mixture modelling by involving the covariates of Bidikmisi scholarship recipients. The estimating parameter was performed using Bayesian Markov Chain Monte Carlo (MCMC) couple with the Gibbs Sampling algorithm. This model is applied to data registrants Bidikmisi districts/cities in the province of East Java as many as 44,489 students. This model shows the smallest value of Deviance Information Criteria (DIC) compared with Bayesian binary logistic regression. © Published under licence by IOP Publishing Ltd.

The purpose of this correlation research was to determine the correlation between student metacognitive skills and student achievement. The sample consisted of 180 students grade 10 from all students of class X Senior High School 1 Kasihan. Saturated sampling technique was used to determine the sample used. Metacognitive Awareness Inventory (MAI) quissionaire was used to measure the level of student metacognitive skills. MAI quissionaire consisted of 18 statements with a 5 point Likert scales that ranging from "strongly agree" to "strongly disagree". The student achievement was measured using the achievement test instruments on ionic bonds and covalent bonds learning. The data were analyzed by using product moment correlation. Result of research indicated that there was very low correlation between metacognitive skills and student achievement. The correlation between student metacognitive skills and student achievement was positive and significant. In other words, if the metacognitive skill is high, then the student achievement are also high. © Published under licence by IOP Publishing Ltd.

Sugarcane bagasse is a biomass with very potential, especially in agricultural countries. In this research, a study related to pretreatment and the ratio of C/N to biogas production from sugarcane bagasse was conducted. The study was carried out on a laboratory scale with physical pretreatment by grinding with a size of 6 mesh, then biological pretreatment with the microbial consortium 5 % g/V, and variations in the C/N ratio at 25 and 30. Cow rumen fluid was used in this study as a source of anaerobic bacteria and technical urea is used to adjust the C/N ratio. The solid total is set at 7 %. Based on research that has been carried out for 60 days, it was found that physical pretreatment by grinding with a size of 6 mesh can increase biogas production. Biogas production increased both at C/N ratio conditions of 25 and 30. Furthermore, biological pretreatment with the microbial consortium 5 % g/V was proven to increase biogas production at both C/N ratios of 25 and 30. Further studies on the C/N ratio, it was found that the C/N ratio of 25 was better than the C/N ratio of 30. This is evident in several variables, including raw and fine substrate conditions, as well as on variables with or without a microbial consortium, with the amount of biogas productivity at a C/N 25 ratio of 53-90.5 % higher than the C/N 30 ratio. © 2023 The Authors. Cloning and Sequencing of Haloacid Dehalogenase Gene from Klebsiella Pneumoniae ITB1

Monochloroacetic acid (MCA) production continues to grow every year and it causes environmental problems. Klebsiella pneumoniae ITB1 is identified to be able to grow on minimum medium with MCA as a sole of carbon source. This strain breaks down MCA to produce 2-hydroxyacetic acid and releases chloride ion into the medium, which can be detected by colorimetric method. Enzyme responsible for this activity is haloacid dehalogenase. This experiment amplified the haloacid dehalogenase gene of Klebsiella pneumoniae ITB1 by PCR. The result was 0.69 kb DNA fragment. This fragment was cloned to pGEM-T easy and then transformed to competent E. coli TOP10 cells. Recombinant colonies were selected based on ampicillin resistant and β-galactosidase activity. Recombinant plasmid confirmation was conducted by size screening, re-PCR, and restriction analysis. The restriction analysis showed positive result as the Eco RI digests of pGEM-HAD recombinant produced two DNA fragments of 3.15 kb and 0.71 kb. Nucleotide sequence of hakp1 DNA fragment confirmed that it is a haloacid dehalogenase gene with 99% similarity to Klebsiella pneumonia . Amino acids sequence deduced by in silico analysis of hakp1 showed 100% sequence similarity with haloacid dehalogenase of multispecies Klebsiella ( Accession No . WP_004179015.1). Conserve residues analysis using ClustalW2 of Hadkp1 protein compared to another three haloacid dehalogenases i.e. from Burkholderia cepacia MBA4, Xanthobacter autotrophicus GJ10, and Pseudomonas sp. YL showed 20 conserve residues. Five of the 20 conserve residues were catalytic residues, which are aspartate (D), threonine (T), serine (S), lysine (K) and tyrosine (Y). The 3D structure of Hadkp1 protein predicted by in silico analysis has been conducted by I TESSER. Blastp analysis also showed that Hadkp1 protein of Klebsiella pneumoniae ITB1 has high similarity to enolase-phosphatase of Klebsiella pneumonia . This study aimed to analyze the effect of Bacillus megaterium PTB 1.4 on the population of intestinal microflora, digestive enzyme activity, and the growth of catfish. Gnotobiotic and normal fish were used. Treatment using gnotobiotic was divided into gnoto (with feed and 100 μg/mL rifampicin) and gnotoplus (with feed, 100 μg/mL rifampicin, and 1% probiotic);whereas treatment using normal fish was divided into normalplus (with feed and 1% probiotic) and normal (only feed). The amount of bacteria on gastrointestinal tract was measured 30 days after treatments using the total plate count method. The results indicated no significant difference in bacterial growth between gnotobiotic and normal fish. The total amount of probiotic bacteria with normalplus treatment was significantly different with gnotoplus. The activity of protease and amylase enzymes, and specific growth rate in normalplus treatment were significantly higher (p < 0.05) than other treatments. Bacillus megaterium PTB 1.4 increased the activity of digestive enzymes and the growth of catfish. © 2017 Institut Pertanian Bogor

Many large oil palm plantation in Indonesia are located in lowland and flood prone areas, especially in South Sumatera. This causes the plant cannot produce optimally, do not grow well, and even death. The purpose of this research was to investigate the productivity of oil palm located in lowland and floodprone areas of Kelola Sendang Landscape, South Sumatera through groundtruthing (ground check) for validation of a GIS model. The research was conducted at lowland area of 6 (six) oil palm plantation companies located in the area of Musi Banyuasin District and Banyuasin District, South Sumatera Province. The result showed that the diversity of oil palm plant growth was quite high due to the number of inserted or abnormal plants and plant death. The low productivity of oil palm crops in lowland areas in the companies was caused by inadequate water management, lack of maintenance of plants, especially fertilization and weed control. The level of oil palm productivity in lowland areas were less than 50% of S3 standards land suitability class. © 2018 Published under licence by IOP Publishing Ltd.

The purpose of this research is to obtain the best formulation of corn-cob fermente d by Trichoderma viridae, Trichoderma reesei, Aspergillus oryzae, Rhizopus oligosporus as material feed for java barb to enhance the growth rate of the fish. This research had been conducted in the Laboratory of Nutrition and Feed, Faculty of Husbandry, and Wet Laboratorium, Faculty of Fisheries and Marine Science, all of Padjadjaran University. Research design applied use Completely Randomized Design, 6 treatments and 3 times replication. The conclusion of feeding trial to java barb shown that the highest growth is found at 5% corncob fermentation fish feed, while the highest growth rate and the lowest fish feed conversion were found in more than 5%.
