So far, cell culture engineering has used the high-throughput transcriptional and more recently protein profiling techniques attempting to further understand and resolve significant issues regarding the cell culture performance and its optimization. At the metabolic level, flux analysis has been used to monitor changes in the metabolic state that could affect the successful outcome of cell cultures. Metabolomics, referring to the simultaneous quantification of the (relative) concentration of the free small metabolites, is the most recently introduced high-throughput method for the measurement of the metabolic fingerprint of a biological system. Considering the role of metabolism in the context of the overall cellular function, it is easily understandable why quantifying a complete and accurate metabolomic profile is foreseen to have a major positive impact in cell culture engineering research. We will present application of Gas Chromatorgaphy-Mass Spectrometry metabolomics to monitor and analyze mammalian cell culture systems. Specifically, two bioreactor processes will be discussed in which baby hamster kidney cells were cultivated in high-cell density perfusion reactors at both laboratory and manufacturing scales and samples were analyzed over the course of the cultivation. Metabolomics enabled the differentiation between cell cultures based on bioreactor type, and stage of cell growth. Multivariate statistical analysis enabled the identification of the metabolites whose change in concentration was characteristic of the modification in any of these macroscopic parameters. The acquired results support the usefulness of metabolomics as a molecular analysis tool in cell culture engineering. © 2010 IFAC.

The Web Services Agreement specification defines a normative language to formulate Service Level Agreements and a basic protocol to expose service-level descriptions, validate service-level requests, and come to an agreement. This protocol, often called “take-it-or-leave-it”, allows a service provider and a service consumer to decide whether to accept or reject a service offer. Although this approach is sufficient for a number of use cases, others exist with requirements for multi-step negotiation or the adaptation of an existing agreement. In this paper, we describe the Web Services Agreement Negotiation protocol, a proposal by the Open Grid Forum to extend the existing specification. This proposal is the result of combining various research activities that have been conducted to define protocols for negotiating service levels or to supersede the existing “take-it-or-leave-it” protocol. The main characteristics of this proposal are the multi-round negotiation capability, renegotiation capability, and compliance with the original specification.

Structural disjoining potential for grain-boundary premelting and grain coalescence from molecular-dynamics simulations

We describe a molecular-dynamics framework for the direct calculation of the short-ranged structural forces underlying grain-boundary premelting and grain coalescence in solidification. The method is applied in a comparative study of (i) a Σ9⟨115⟩120° twist and (ii) a Σ9⟨110⟩{411} symmetric tilt boundary in a classical embedded-atom model of elemental Ni. Although both boundaries feature highly disordered structures near the melting point, the nature of the temperature dependence of the width of the disordered regions in these boundaries is qualitatively different. The former boundary displays behavior consistent with a logarithmically diverging premelted layer thickness as the melting temperature is approached from below, while the latter displays behavior featuring a finite grain-boundary width at the melting point. It is demonstrated that both types of behavior can be quantitatively described within a sharp-interface thermodynamic formalism involving a width-dependent interfacial free energy, referred to as the disjoining potential. The disjoining potential for boundary (i) is calculated to display a monotonic exponential dependence on width, while that of boundary (ii) features a weak attractive minimum. The results of this work are discussed in relation to recent simulation and theoretical studies of the thermodynamic forces underlying grain-boundary premelting.

Rotating three-dimensional solitons in Bose-Einstein condensates with gravitylike attractive nonlocal interaction

We study formation of rotating three-dimensional high-order solitons (azimuthons) in Bose Einstein condensate with attractive nonlocal nonlinear interaction. In particular, we demonstrate formation of toroidal rotating solitons and investigate their stability. We show that variational methods allow a very good approximation of such solutions and predict accurately the soliton rotation frequency. We also find that these rotating localized structures are very robust and persist even if the initial condensate conditions are rather far from the exact soliton solutions. Furthermore, the presence of repulsive contact interaction does not prevent the existence of those solutions, but allows one to control their rotation. We conjecture that self-trapped azimuthons are generic for condensates with attractive nonlocal interaction.

PatternLab for proteomics is a one-stop shop computational environment for analyzing shotgun proteomic data. Its modules provide means to pinpoint proteins/peptides that are differentially expressed and those that are unique to a state. It can also cluster the ones that share similar expression profiles in time-course experiments, as well as help in interpreting results according to Gene Ontology. PatternLab is user-friendly, simple, and provides a graphical user interface. Copyright © 2010 John Wiley & Sons, Inc.

A new method for compensating phase-shifting error in phase measuring profilometry

A new phase-shifting error compensating algorithm for phase-measuring profilometry is proposed. In this error detecting algorithm, it is supposed that sinusoidal fringe is projected to the plate, thus the phase differences between adjacent pixels are equal. Three adjacent phase points are taken as the research object. Combining the principle of phase compensation measurement, each phase of the point is presented as an equation containing the phase-shifting error. According to the characteristics that the phase differences between adjacent pixels are equal, an equation to calculate the phase-shifting error is constructed. Then the phase shifting error is taken into account in the original algorithm for error compensation. Simulation results show the inhibitory effect of the error compensation by using the novel algorithm. The proposed algorithm is also used for three-dimensional reconstruction. The final result shows that the new compensation algorithm is better than the original one obviously. This error compensating algorithm can effectively improve the accuracy of three-dimensional measurement.

Nanosized magnetite particles, with mean physical diameter of about 7 nm, obtained by chemical coprecipitation procedure were dispersed in water carrier by applying sterical stabilization of particles in order to prevent their aggregation and to ensure colloidal stability of the systems. Different chain length (C12, C14, C18) carboxylic acids (lauric (LA), myristic (MA) and oleic (OA)) were used for double layer coating of magnetite nanoparticles. Structural and magnetic properties were investigated by electron microscopy (TEM), dynamical and static light scattering (DLS, SLS) and magnetometry (VSM) to evaluate the role of chain length and of the saturated/unsaturated nature of surfactant layers. Also investigated were two water based magnetic nanocomposites obtained by encapsulating the magnetic nanoparticles in polymers with different functional properties.

The main knowledge management challenges are to capture, store and reuse contextual knowledge generated during interactions that occur daily in an organization. In this paper, we propose an activity context-aware architecture to support knowledge management in working processes. The required features for this architecture are processing, reasoning and sharing contextual knowledge involving information about activities performed. We also present results from evaluation of our proposal — A-CoBrA — for a specific domain.

"Virtual Coach" refers to a coaching program or device aiming to guide users through tasks for the purpose of prompting positive behavior or assisting with learning new skills. This article reviews virtual coach interventions with the purpose of guiding rehabilitation professionals to comprehend more effectively the essential components of such interventions, the underlying technologies and their integration, and example applications. A design space of virtual coach interventions including self-monitoring, context awareness, interface modality, and coaching strategies were identified and discussed to address when, how, and what coaching messages to deliver in an automated and intelligent way. Example applications that address various health-related issues also are provided to illustrate how a virtual coach intervention is developed and evaluated. Finally, the article provides some insight into addressing key challenges and opportunities in designing and implementing virtual coach interventions. It is expected that more virtual coach interventions will be developed in the field of rehabilitation to support self-care and prevent secondary conditions in individuals with disabilities. © 2010 Elsevier Inc.
