In-silico structural and functional analysis of nonsynonymous single nucleotide polymorphisms in human FOLH1 gene

Non-synonymous single nucleotide polymorphisms (nsSNPs), also known as missense SNPs, can seriously affect an individual’s vulnerability to numerous diseases, including cancer. In this study, we conducted a comprehensive in-silico analysis to examine the structural and functional implications of nsSNPs within the Folate Hydrolase 1(FOLH1) gene, which encodes the Prostate-Specific Membrane Antigen (PSMA). A total of 504 SNPs were retrieved, and after filtering, 15 pathogenic nsSNPs were identified using five different in-silico tools. Three of these SNPs—R255H (rs375565491), R255C (rs201789325), and G168E (rs267602926)—were consistently predicted to be pathogenic across all in-silico tools. MutPred2 was used to predict the structural and functional consequences of the identified mutations. The analysis revealed multiple alterations in the PSMA protein, including changes in helical conformations, glycosylation patterns, transmembrane properties, and solvent accessibility. Furthermore, I-Mutant 2.0 analysis demonstrated a decrease in protein stability for most nsSNPs, except for rs267602926 (G168E), which was predicted to increase stability. Conservation analysis using ConSurf revealed varying degrees of amino acid conservation, with R255H and R255C identified as highly conserved residues, indicating their potential functional and structural significance. Additionally, post-translational modification (PTM) analysis indicated that while phosphorylation and methylation sites remained unchanged, specific glycosylation sites were lost in two pathogenic mutant variants (R255H and R255C), potentially affecting PSMA function and adversely impacting prostate cancer. Our findings highlight the importance of in silico studies to investigate the structural and functional impacts of FOLH1 nsSNPs on the PSMA protein. Such in silico studies can deepen our understanding of the roles of nsSNPs in prostate cancer onset, progression, and drug resistance.

For damage detection, this research article discusses an easy-to-compute damage index derived from the governing dynamic of the structure that has potential practical application in Structural Health Monitoring (SHM). The research uses simplified structural models to explore the sensitivity of the index to damages, to compare the index performance with a traditional but popular damage detection method, and to understand the local/global predictive capability of the index. The research uses two simple models, namely, single- and two-degree-of-freedom systems. The results suggest that the damage index is local, that can only monitor damages occurring near the points of measurements, but it is sensitive to damages, unlike the natural frequency, which is global but less sensitive.

Understanding the reliability of engineering methods is crucial for its adoption and deployment. This research focuses on the reliability of the Power Spectral Density (PSD) method via the use of the F statistic for damage detection. To the author best knowledge, the method is rather classic but its realibility has not been discussed in the context of a large data size. Priory, the research anticipates that the accuracy is a function of the damage level. In this study, we evaluate 3500 cases with five levels of structural integrity, namely, healthy condition and damaged conditions with 1%, 5%, 10%, and 20% damage levels. The dataset is established via a numerical analysis of a seven degree-of-freedom system loaded with a concentrated dynamic force with random magnitude. A spring on the system is reduced in its stiffness to simulate damages. Our significant findings are the following: it is challenging for the PSD-based method to differentiate the healthy condition from the damaged conditions when the damage level is small. However, the reliability is high at 95% probability when the structural integrity has dropped by five percent.

Dynamic laplace: Efficient centrality measure for weighted or unweighted evolving networks

With its origin in sociology, Social Network Analysis (SNA), quickly emerged and spread to other areas of research, including anthropology, biology, information science, organizational studies, political science, and computer science. Being it's objective the investigation of social structures through the use of networks and graph theory, Social Network Analysis is, nowadays, an important research area in several domains. Social Network Analysis cope with different problems namely network metrics, models, visualization and information spreading, each one with several approaches, methods and algorithms. One of the critical areas of Social Network Analysis involves the calculation of different centrality measures (i.e.: the most important vertices within a graph). Today, the challenge is how to do this fast and efficiently, as many increasingly larger datasets are available. Recently, the need to apply such centrality algorithms to non static networks (i.e.: networks that evolve over time) is also a new challenge. Incremental and dynamic versions of centrality measures are starting to emerge (betweenness, closeness, etc). Our contribution is the proposal of two incremental versions of the Laplacian Centrality measure, that can be applied not only to large graphs but also to, weighted or unweighted, dynamically changing networks. The experimental evaluation was performed with several tests in different types of evolving networks, incremental or fully dynamic. Results have shown that our incremental versions of the algorithm can calculate node centralities in large networks, faster and efficiently than the corresponding batch version in both incremental and full dynamic network setups. Diffusion-weighted MRI (DWI) is essential for stroke diagnosis, treatment decisions, and prognosis. However, image and disease variability hinder the development of generalizable AI algorithms with clinical value. We address this gap by presenting a novel ensemble algorithm derived from the 2022 Ischemic Stroke Lesion Segmentation (ISLES) challenge. ISLES’22 provided 400 patient scans with ischemic stroke from various medical centers, facilitating the development of a wide range of cutting-edge segmentation algorithms by the research community. By assessing them against a hidden test set, we identified strengths, weaknesses, and potential biases. Through collaboration with leading teams, we combined top-performing algorithms into an ensemble model that overcomes the limitations of individual solutions. Our ensemble model combines the individual algorithms’ strengths and achieved superior ischemic lesion detection and segmentation accuracy (median Dice score: 0.82, median lesion-wise F1 score: 0.86) on our internal test set compared to individual algorithms. This accuracy generalized well across diverse image and disease variables. Furthermore, the model excelled in extracting clinical biomarkers like lesion types and affected vascular territories. Notably, in a Turing-like test, neuroradiologists consistently preferred the algorithm’s segmentations over manual expert efforts, highlighting increased comprehensiveness and precision. Validation using a real-world external dataset (N=1686) confirmed the model’s generalizability (median Dice score: 0.82, median lesion-wise F1 score: 0.86). The algorithm’s outputs also demonstrated strong correlations with clinical scores (admission NIHSS and 90-day mRS) on par with or exceeding expert-derived results, underlining its clinical relevance. Indonesia is one of the countries whose people are most affected by the 2019 corona virus (COVID-19). When the corona virus spreads faster, the Indonesian government imposed large-scale social restrictions in big cities to villages in Indonesia. With the changing situation in carrying out work activities during this pandemic, it will certainly affect the quality of organization in companies and the quality of education in schools and colleges. In order to identify all processes in teaching and learning activities that are running well or not, schools need to monitor and measure employee performance. Key performance indicators (KPIs) are generally used as benchmarks to determine how well a company's performance can be represented by employee performance. In this study, the key performance indicator preparation process was carried out through the stages of designing a questionnaire, distributing questionnaires to employees in educational institutions, conducting factor analysis and then developing a KPI model. The output is in the form of a KPI model that can be used as a reference in the formation of KPIs in educational institutions, namely Y = 5,360 + 1,380 X1-1, 068 X3.

Background: Remote approaches to workplace concussion assessment have demonstrated value to end users. The feasibility of administering physical concussion assessment measures in a remote context has been minimally explored, and there is limited information on important psychometric properties of physical assessment measures used in remote contexts. Objective: The objectives of this feasibility study were to determine recruitment capability for a future larger-scale study aimed at determining sensitivity and reliability of the remote assessment, time required to complete study assessments, and acceptability of remote assessment to people with brain injuries and clinicians;document preliminary results of the sensitivity of the remote assessment when compared to the in-person assessment;and estimate the preliminary interrater and intrarater reliability of the remote assessments to inform procedures of a future larger-scale study that is adequately powered to reliably estimate these parameters of interest. Methods: People living with acquired brain injury attended 2 assessments (1 in-person and 1 remote) in a randomized order. The measures administered in these assessments included the finger-to-nose test;balance testing;and the Vestibular/Ocular Motor Screening (VOMS) tool, including documentation of change in symptoms and distance for near point convergence, saccades, cervical spine range of motion, and evaluation of effort. Both assessments occurred at the Ottawa Hospital Rehabilitation Center. After the assessments, a clinician different from the person who completed the original assessments then viewed and documented findings independently on the recordings of the remote assessment. The same second clinician viewed the recording again approximately 1 month following the initial observation. In this paper strategies to control the speed of a generic DC motor system are discussed. They range from the conventional, like PID and state-feedback, to reference model adaptive algorithms such as MRAC, VS-MRAC and DMARC. The goal of this study is to determine the best control strategy for high-performance applications regarding to stability, robustness and optimal controller effort. In theory and sometimes in practice, all these control strategies proof themselves reliable, however, in heavy-duty set-point references the application requires high performance of the controller under disturbances, unmodeled dynamics and variable speed reference conditions. The use of appropriate control strategies can highly contribute to this purpose. To support the conclusions, the control strategies have been embedded in a microcontroller PIC18F4520 and applied to a test motor in order to obtain experimental results. © 2018 IEEE.

Magnetic Resonance Images of the brain provide detailed anatomical information that allows morphological analysis of the different brain structures. The analysis of the cortical folding patterns variation is inspiring great interest, as this relates to cognitive function. The sulci are a depression in the cerebral cortex and represent ≈ 70% of the cortical surface. The sulcal width, depth and cortical thickness are the most frequent morphological descriptors applied to the sulci. In this work, the 3D tortuosity is proposed as new descriptor to capture information about the twist and turns of the sulci. 1 The 3D tortuosity of the central sulcus of both hemispheres was measured using the Minimal Interval Resonance Imaging in the Alzheimer's Disease (MIRIAD) database, for a set of high-resolution MRI of 66 subjects: 43 patients with Alzheimer Disease (AD) and 23 control subjects. As it is known AD causes significant gray matter loss which cause morphological changes in the cortical structure. It is expected for the tortuosity to capture these changes and serve as a biomarker to differentiate between populations. The result of the Wilcoxon tests show that the tortuosity values of the central sulci are significantly larger for the AD patients (p < 0.05) for the left hemisphere, which is consistent with the hypothesis. The evaluation of the proposed tortuosity measurement of the Central Sulcus as a potential biomarker was performed and the results indicate its effectiveness to extract additional anatomical information and discriminate between AD patients and Controls.
