Predicting human pharmacokinetics of novel compounds is a critical step in drug discovery and clinical study design but continues to be a challenging task for hepatic transporter substrates, particularly in predicting their liver exposures. In this study, using bosentan as an example, we prospectively predicted systemic exposure and the (pseudo) steady-state unbound liver-to-unbound plasma ratio (Kpuu) in healthy subjects using 1) a mechanistic approach solely based on in vitro hepatocyte assays and 2) an approach based on hepatic process rates from monkey in vivo data but Michaelis-Menten constants from in vitro data. Both methods reasonably match the observed human systemic time course data, but the second method leads to better prediction accuracy. In addition, the second method can predict a human Kpuu value that is close to the value deduced using clinical data. We also generated rat and monkey liver Kpuu values in terminal studies. However, these directly measured animal values are different from the deduced human value.

Taking into account the analysis of discretization methods for common differential equations, the paper proves the doubts concerning their suitabilityfor the radical solution to the problem of electric power systems (EPS) modeling. This involves continuous and methodically accurate reproduction of the whole range of EPS processes within real time and at the unlimited interval. The modern concept of EPS hybrid modeling and means of its implementation are suggested in this paper.

The problems of a mechanical control systems description base on struc¬tured representation of their elements are investigated. A set problems of struc¬tured representation of a mechanical control systems, problems of generation of ones mathematical models and control laws, problems of admissible and optimal synthesis, self-organizing and genetic development of the structures of the systems are formulated. Some results for solving of the given problems are produced. Software package VSLIDE for modeling and synthesis of a complicated mechanical controlled systems such as multi-links manipulators, multi-manipulators robots, space apparatus etc., are created.

Several options for modeling high volume conveyors are examined, and the advantages and disadvantages of each option are noted. Several key issues, such as computer memory, run time, and model accuracy, are evaluated for each modeling approach. A high-volume conveyor sorting system at a Federal Express facility is used to illustrate how one modeling option has been successfully implemented. The option selected required that unique conveyor constructs be developed. These constructs provide all of the control of the physical system and make the model efficient from a computer memory and execution speed standpoint.

Computer simulation is often used as an analysis tool during the design of automated guided vehicle (AGV) systems. However, because of the complexities inherent in automated material handling systems, general-purpose simulation languages must be used creatively to capture the desired detail in the model. This paper presents an average-value model of a line-commutated converter-based HVDC system using dynamic phasors. The model represents the low-frequency dynamics of the converter and its ac and dc systems, and has lower computational requirements than a conventional electromagnetic-transient (EMT) switching model. The developed dynamic-phasor model is verified against an EMT model of the CIGRE HVDC Benchmark. Simulation results confirm the validity and accuracy of the average value model in predicting the low-frequency dynamics of the ac- and dc-side quantities. Merits and applicability limitations of the average model are highlighted.
