Robustness and stability of spin-glass ground states to perturbed interactions

Across many problems in science and engineering, it is important to consider how much the output of a given system changes due to perturbations of the input. Here, we investigate the glassy phase of ±J spin glasses at zero temperature by calculating the robustness of the ground states to flips in the sign of single interactions. For random graphs and the Sherrington-Kirkpatrick model, we find relatively large sets of bond configurations that generate the same ground state. These sets can themselves be analyzed as subgraphs of the interaction domain, and we compute many of their topological properties. In particular, we find that the robustness, equivalent to the average degree, of these subgraphs is much higher than one would expect from a random model. Most notably, it scales in the same logarithmic way with the size of the subgraph as has been found in genotype-phenotype maps for RNA secondary structure folding, protein quaternary structure, gene regulatory networks, as well as for models for genetic programming. The similarity between these disparate systems suggests that this scaling may have a more universal origin.

Recent studies of genotype-phenotype maps have reported universally enhanced phenotypic robustness to genotype mutations, a feature essential to evolution. Virtually all of these studies make a simplifying assumption that each genotype—represented as a sequence—maps deterministically to a single phenotype, such as a discrete structure. Here we introduce probabilistic genotype-phenotype (PrGP) maps, where each genotype maps to a vector of phenotype probabilities, as a more realistic and universal language for investigating robustness in a variety of physical, biological, and computational systems. We study three model systems to show that PrGP maps offer a generalized framework which can handle uncertainty emerging from various physical sources: (1) thermal fluctuation in RNA folding, (2) external field disorder in the spin-glass ground state search problem, and (3) superposition and entanglement in quantum circuits, which are realized experimentally on IBM quantum computers. In all three cases, we observe a biphasic robustness scaling which is enhanced relative to random expectation for more frequent phenotypes and approaches random expectation for less frequent phenotypes. We derive an analytical theory for the behavior of PrGP robustness, and we demonstrate that the theory is highly predictive of empirical robustness.

Exposure to ambient air pollution component particulate matter (PM) in urban environments has been associated with adverse health effects to the exposed population. This study aimed to investigate the cytotoxic and inflammatory responses of lung A549 cells to ambient particulate matter (PM) extracts (EE) collected from a geographical site in south of city limits of Jackson, Mississippi. PM was collected from the site, extracted, and characterized for chemical composition and microscopic analysis using SEM. Following characterization, the EE was used to treat A549 cells, and various cellular responses were assessed. Our SEM analysis of EE indicated the presence of micrometer-sized particles, and elemental analysis indicated the presence of elements such as C, Si, Fe, S, N, P, and Ca. Cell viability decreased in a dose- and timedependent manner following exposure to EE. Inflammatory gene IL-1β, chemokine CCL-18 and MIP-1, and transcription factor NF-кB were upregulated by EE. EE also increased the level of ROS and mitochondria. In conclusion, our data indicates that EE is cytotoxic, inflammatory, and an oxidant to lung cells Also, the findings of the present study highlight the potential health risks associated with exposure to air pollution and inflammatory lung disease. © 2024 IAE All rights reserved.

Background and Aim: Among low viral load (DNA of hepatitis B virus (HBV) was < 2000 IU/mL), the factor of the loss of hepatitis B surface antigen (HBsAg) remained elusive. Methods: The retrospective study recruited patients with chronic hepatitis B (CHB) who were negative low for hepatitis B e-antigen (HBeAg), had a low viral load, and experienced HBsAg loss during follow-up. CHB patients with low-viral load but without consequent HBsAg loss were also enrolled at the ratio of 1:4. The factors contributing to HBsAg loss were analyzed. Results: A total of 80 patients were recruited for the current study, with a mean age of 63.9 years and 61.3% being male. Among them, 62.5% patients (50/80) were treated with potent nucleoside/nucleotide analogues (NAs) during the follow-up period. Additionally, 12.5% patients (10/80) had a prior history of NAs treatment before enrolment. During the follow-up, HBsAg loss occurred in 17 patients (21.3%). Compared with patients without HBsAg loss, those with HBsAg loss were younger (57.9 years vs 65.5 years;P = 0.01), had lower HBV DNA levels (1.3 log₁₀ IU/mL vs 2.3 log₁₀ IU/mL;P = 0.003), and higher proportion of prior NAs-treated history. Logistic regression analysis revealed that the factors associated with factors associated with HBsAg loss were age < 60 years (OR/CI: 3.95/1.15–13.60, P = 0.03), prior NAs-treated history (OR/CI: 7.59/1.42–40.51, P = 0.01) and current NAs-treated (OR/CI: 0.19/0.05–0.71, P = 0.01). Conclusions: In the study, older age and prior NAs were positively associated with HBsAg loss, and current NAs was negatively associated with HBsAg loss. MiR-1 Is Regulated by Hydrogen Peroxide via MAPK and Limits Cell Migration and Invasion

MicroRNA-1 (miR-1) is a conserved microRNA that is highly expressed in skeletal and cardiac muscle tissues. Moreover, miR-1 regulates genes and has critical roles in cell migration and invasion. Downregulation of miR-1 has been found in many pathologies of numerous organs, including the lungs. What exactly contributes to the downregulation of miR-1 is not fully understood, and in the present study, we investigated whether ROS regulate miR-1 and its role in cell migration and invasion. A549 cells were grown and maintained in DMEM:F12 (1:1) and supplemented with 10% FBS and 1000 U of Penicillin/Streptomycin and maintained as recommended by the manufacturer (ATCC). Cell migration and invasion, IHC, Western blot, qPCR, ROS, miR-1 transfection, and qPCR were used to determine miR-1 regulation and its role in cell migration. Exogenous miR-1 decreased the formation of ROS and inhibited cell migration and invasion, whereas inhibition of miR-1 increased ROS formation and stimulated cell migration and invasion. Inhibition of miR-1 induced the formation of actin filaments contractile structures, whereas exogenous miR-1 limited the formation of these structures. Hydrogen peroxide significantly decreased miR-1 level, whereas inhibition of Nox4 had no effect on miR-1 level. Alpha amanitin did not decrease miR-1 level, whereas inhibition of NF-кB temporally decreased miR-1 level. This study demonstrates that ROS suppress miR-1 and that miR-1 is posttranscriptionally regulated via MAPK. Endogenous Nox4-dependent ROS are not involved in miR-1 regulation, whereas exogenous ROS regulates miR-1. NF-κB plays a key role in miR-1 regulation in both redox and nonredox environments. Moreover, Mir-1 limits cell migration and invasion even in the presence of ROS. TSP-1 is a major regulator of TGFβ and its expression is upregulated by ROS. Mississippi Burning: An Exploration of Community Noise Levels from a Rural Mississippi Community Grappling with Wood Pellet Production Manufacturing

Wood pellets are being touted as a renewable and sustainable form of energy. As a result of this growing popularity, the global wood pellet market is expected to reach $16 billion by 2026. Currently, there are approximately 100 pellet manufacturing plants in the United States, with most of the production occurring in predominantly Black and poor communities in the rural South. At the request of residents concerned about the noise pollution emanating from wood pellet production plants in their community, we carried out a community noise exposure assessment. From August 2023 to March 2024, and working closely with a local community organization we measured a variety of noise metrics in Gloster, a wood pellet impacted city in rural Mississippi, comparing these noise pollution levels to a background community with no wood pellet industrial activity. We observed statistically significant higher levels of A-weighted, C-weighted, and Z-weighted sound in Gloster compared to Mendenhall. These differences persisted across different times of the day and days of the week. When examining the decibel levels for each of the center frequencies of the 1:3 octave band, this pattern persisted. Our preliminary findings suggest that a rural community’s soundscape can be negatively impacted by unchecked wood pellet manufacturing. We argue that the sound level differences in communities impacted by wood pellet manufacturing and those that are not, combined with the strategic placement of wood pellet plants in vulnerable communities in the rural south, may also be an environmental justice issue. © 2024, The Authors. Mammography is known as one of the best forms to screen possible breast cancer in women, and recently deep learning models have been developed to assist the radiologist in the diagnosis. However, their lack of interpretability has become a significant drawback to their extended use in clinical practice. This paper introduces a novel approach for detecting and localising pathological findings in mammography exams through the use of a EfficientNet-based deep learning model. The model is trained using cropped segments of labelled pathological findings from Vindr Mammography Dataset. Achieving an average F1-score of 72.7 %, and reaching on mass and suspicious calcifications an F1-Score of 79.9 % and 84.5 % respectively. Using this classifier we propose a method to visualise from local information the regions of interest where pathological findings could be present on the complete image. Plus, we describe the limitations regarding area coverage of these patches on the model’s capability of generalization and certainty on its predictions, explaining its functionality.
