The increasing prevalence of multidrug-resistant Acinetobacter baumannii as an opportunistic pathogen in wound infections raises significant concerns due to its antibiotic resistance and biofilm-mediated antibiotic tolerance. This underscores the urgent need to explore an alternative approach to effectively managing wound infections caused by MDR A. baumannii. In this study, combining subinhibitory concentrations of curcumin (25 µg/ml) or chitosan (156 µg/ml) with photodynamic therapy-PDT (10 J/cm²) resulted in a significant reduction of planktonic viabilities and biofilm biovolume of MDR A. baumannii clinical isolates in vitro. In murine A. baumannii-associated wound infection, a combination of curcumin (25 µg/ml) or chitosan (156 µg/ml) with PDT (10 J/cm²) resulted in complete bacterial eradication with wound healing at Day 15 of post-treatments with these combination therapies. Our study demonstrated that combining curcumin or chitosan with PDT at their subinhibitory concentrations represents a promising alternative strategy as localized therapy for effectively managing A. baumannii-associated wound infection. © 2025 The Authors

Artificial Neural Networks (ANNs) implement a specific form of multi-variate extrapolation and will generate an output for any input pattern, even when there is no similar training pattern. Extrapolations are not necessarily to be trusted, and in order to support safety critical systems, we require such systems to give an indication of the training sample related uncertainty associated with their output. Some readers may think that this is a well known issue which is already covered by the basic principles of pattern recognition. We will explain below how this is not the case and how the conventional (Likelihood estimate of) conditional probability of classification does not correctly assess this uncertainty. We provide a discussion of the standard interpretations of this problem and show how a quantitative approach based upon long standing methods can be practically applied. Driving in urban environments often presents difficult situations that require expert maneuvering of a vehicle. These situations become even more challenging when considering large vehicles, such as buses. We present a path planning framework that addresses the demanding driving task of buses in urban areas. The approach is formulated as an optimization problem using the road-aligned vehicle model. The road-aligned frame introduces a distortion on the vehicle body and obstacles, motivating the development of novel approximations that capture this distortion. These approximations allow for the formulation of safe and nonconservative collision avoidance constraints. Unlike other path planning approaches, our method exploits curbs and other sweepable regions, which a bus must often sweep over in order to manage certain maneuvers. Furthermore, it takes full advantage of the particular characteristics of buses, namely the overhangs, an elevated part of the vehicle chassis, that can sweep over curbs. The paper presents the control design approaches for the European research project FLEXOP. The ultimate goal is to develop and apply active flutter suppression and load alleviation techniques on an unmanned flying aircraft demonstrator. Due to the flexible wing of the aircraft new challenges rise for the control design: the traditional rigid body (baseline) control loops have to be augmented with flutter control laws. In our approach, the controllers are designed based on a dynamical model, which is briefly discussed first. Details of the baseline control design, as well as the two different flutter suppression algorithms are discussed in the paper. Computational models of acoustic wave propagation are frequently used in transcranial ultrasound therapy, for example, to calculate the intracranial pressure field or to calculate phase delays to correct for skull distortions. To allow intercomparison between the different modeling tools and techniques used by the community, an international working group was convened to formulate a set of numerical benchmarks. Here, these benchmarks are presented, along with intercomparison results. Nine different benchmarks of increasing geometric complexity are defined. These include a single-layer planar bone immersed in water, a multi-layer bone, and a whole skull. Two transducer configurations are considered (a focused bowl and a plane piston), giving a total of 18 permutations of the benchmarks. Eleven different modeling tools are used to compute the benchmark results. The models span a wide range of numerical techniques, including the finite-difference time-domain method, angular-spectrum method, pseudospectral method, boundary-element method, and spectral-element method. Good agreement is found between the models, particularly for the position, size, and magnitude of the acoustic focus within the skull. When comparing results for each model with every other model in a cross comparison, the median values for each benchmark for the difference in focal pressure and position are less than 10% and 1 mm, respectively. The expansive production of data in materials science, their widespread sharing and repurposing requires educated support and stewardship. In order to ensure that this need helps rather than hinders scientific work, the implementation of the FAIR-data principles (Findable, Accessible, Interoperable, and Reusable) must not be too narrow. Besides, the wider materials-science community ought to agree on the strategies to tackle the challenges that are specific to its data, both from computations and experiments. In this paper, we present the result of the discussions held at the workshop on "Shared Metadata and Data Formats for Big-Data Driven Materials science". We start from an operative definition of metadata, and what features a FAIR-compliant metadata schema should have. We will mainly focus on computational materials-science data and propose a constructive approach for the FAIRification of the (meta)data related to ground-state and excited-states calculations, potential-energy sampling, and generalized workflows. Finally, challenges with the FAIRification of experimental (meta)data and materials-science ontologies are presented together with an outlook of how to meet them. © 2022, CC BY.

CD8+T cells are a major prognostic determinant in solid tumors, including colorectal cancer (CRC). However, understanding how the interplay between different immune cells impacts on clinical outcome is still in its infancy. Here, we describe that the interaction of tumor infiltrating neutrophils expressing high levels of CD15 with CD8+T effector memory cells (TEM) correlates with tumor progression. Mechanistically, stromal cell-derived factor-1 (CXCL12/SDF-1) promotes the retention of neutrophils within tumors, increasing the crosstalk with CD8+T cells. As a consequence of the contact-mediated interaction with neutrophils, CD8+T cells are skewed to produce high levels of GZMK, which in turn decreases E-cadherin on the intestinal epithelium and favors tumor progression. Overall, our results highlight the emergence of GZMKhighCD8+TEMin non-metastatic CRC tumors as a hallmark driven by the interaction with neutrophils, which could implement current patient stratification and be targeted by novel therapeutics.

Indicine cattle, also referred to as zebu (Bos taurus indicus), play a central role in pastoral communities across a wide range of agro-ecosystems, from extremely hot semiarid regions to hot humid tropical regions. However, their adaptive genetic changes following their dispersal into East Asia from the Indian subcontinent have remained poorly documented. Here, we characterize their global genetic diversity using high-quality whole-genome sequencing data from 354 indicine cattle of 57 breeds/populations, including major indicine phylogeographic groups worldwide. We reveal their probable migration into East Asia was along a coastal route rather than inland routes and we detected introgression from other bovine species. Genomic regions carrying morphology-, immune-, and heat-tolerance-related genes underwent divergent selection according to Asian agro-ecologies. We identify distinct sets of loci that contain promising candidate variants for adaptation to hot semi-arid and hot humid tropical ecosystems. Our results indicate that the rapid and successful adaptation of East Asian indicine cattle to hot humid environments was promoted by localized introgression from banteng and/or gaur. Our findings provide insights into the history and environmental adaptation of indicine cattle.

With the evolution of the 5G, 6G and beyond, device-to-device (D2D) communication has been developed as an energy-, and spectrum-efficient solution. In cellular network, D2D links need to share the same spectrum resources with the cellular link. A reconfigurable intelligent surface (RIS) can reconfigure the phase shifts of elements and create favorable beam steering, which can mitigate aggravated interference caused by D2D links. In this paper, we study a RIS-assisted single cell uplink communication network scenario, where the cellular link and multiple D2D links utilize direct propagation and reflecting one-hop propagation. The problem of maximizing the total system rate is formulated by jointly optimizing transmission powers of all links and discrete phase shifts of all elements. The formulated problem is an NP-hard mixed integer non-convex non-linear problem. To obtain practical solutions, we capitalize on alternating maximization and the problem is decomposed into two sub-problems. For the power allocation, the problem is a difference of concave functions (DC) problem, which is solved with the gradient descent method. For the phase shift, a local search algorithm with lower complexity is utilized. Simulation results show that deploying RIS and optimizing the phase shifts have a significant effect on mitigating D2D network interference. Copyright © 2020, The Authors. All rights reserved.
