Oral squamous cell carcinoma(OSCC)is a prevalent malignant tumor affecting the head and neck region(Leemans et al.,2018).It is often diagnosed at a later stage,leading to a poor prognosis(Muzaffar et al.,2021;Li et al.,2023).Despite advances in OSCC treatment,the overall 5-year survival rate of OSCC patients remains alarmingly low,falling below 50%(Jehn et al.,2019;Johnson et al.,2020).According to statistics,only 50%of patients with oral cancer can be treated with *** discovered,it is more frequently at an advanced *** addition,owing to the aggressively invasive and metastatic characteristics of OSCC,most patients die within one year of ***,the pursuit of novel therapeutic drugs and treatments to improve the response of oral cancer to medication,along with a deeper understanding of their effects.

Endometrial cancer is the most common gynecological malignancy,affecting up to 3%of women at some point during their lifetime(Morice et al.,2016;Li and Wang,2021).based on the pathogenesis and bio‑logical behavioral characteristics,endometrial cancer can be divided into estrogen-dependent(I)and non-estrogen-dependent(II)types(Ulrich,2011).Type I accounts for approximately 80%of cases,of which the majority are endometrioid carcinomas,and the remaining are mucinous adenocarcinomas(Setiawan et al.,2013).

Accumulating evidence has confirmed the links between transfer RNA(tRNA)modifications and tumor *** present study is the first to explore the role of tRNA methyltransferase 5(TRMT5),which catalyzes the m1G37 modification of mitochondrial tRNAs in hepatocellular carcinoma(HCC)***,based on bioinformatics and clinical analyses,we identified that TRMT5 expression was upregulated in HCC,which correlated with poor *** TRMT5 attenuated HCC proliferation and metastasis both in vivo and in vitro,which may be partially explained by declined extracellular acidification rate(ECAR)and oxygen consumption rate(OCR).Mechanistically,we discovered that knockdown of TRMT5 inactivated the hypoxia-inducible factor-1(HIF-1)signaling pathway by preventing HIF-1αstability through the enhancement of cellular oxygen ***,our data indicated that inhibition of TRMT5 sensitized HCC to doxorubicin by adjusting HIF-1α.In conclusion,our study revealed that targeting TRMT5 could inhibit HCC progression and increase the susceptibility of tumor cells to chemotherapy ***,TRMT5 might be a carcinogenesis candidate gene that could serve as a potential target for HCC therapy.

Mesenchymal stromal cells (MSCs) exhibit significant potential in the context of cell therapy because of their capacity to perform a range of interconnected functions in damaged tissues, including immune modulation, hematopoietic support, and tissue regeneration. MSCs are hypoimmunogenic because of their diminished expression of major histocompatibility molecules, absence of costimulatory molecules, and presence of coinhibitory molecules. While autologous MSCs reduce the risk of rejection and infection, variability in cell numbers and proliferation limits their potential applications. Conversely, allogeneic MSCs (allo-MSCs) possess broad clinical applications unconstrained by donor physiology. Nonetheless, preclinical and clinical investigations highlight that transplanted allo-MSCs are subject to immune attack from recipients. These cells exhibit anti-inflammatory and proinflammatory phenotypes contingent on the microenvironment. Notably, the proinflammatory phenotype features enhanced immunogenicity and diminished immunosuppression, potentially triggering allogeneic immune reactions that impede long-term clinical efficacy. Consequently, preserving the low immunogenicity of allo-MSCs in vivo and mitigating immune rejection in diverse microenvironments represent crucial challenges for the widespread clinical application of MSCs. In this review, we elucidate the immune regulation of allo-MSCs, specifically focusing on two distinct subgroups, MSC1 and MSC2, that exhibit varying polarization states and immunogenicity. We discuss the factors and underlying mechanisms that induce MSC immunogenicity and polarization, highlighting the crucial role of major histocompatibility complex class I/II molecules in rejection post-transplantation. Additionally, we summarize the immunogenic regulatory targets and applications of allo-MSCs and outline strategies to address challenges in this promising field, aiming to enhance allo-MSC therapeutic efficacy for patients. © 2024 The Auth

Alterations in the mesenchymal-epithelial transition factor(MET)gene are critical drivers of non-small cell lung cancer(NSCLC).In recent years advances in precision therapies targeting MET alterations have significantly expanded treatment options for NSCLC *** alterations include MET exon 14 skipping mutations(MET exon 14 skipping),MET gene amplifications,MET point mutations(primarily kinase domain mutations),and MET protein *** identification of these alterations and appropriate selection of patient populations and targeted therapies are essential for improving clinical *** East China Lung Cancer Group,Youth Committee(ECLUNG YOUNG,Yangtze River Delta Lung Cancer Cooperation Group)has synthesized insights from China’s innovative drug development landscape and clinical practice to formulate an expert consensus on the diagnosis and treatment of NSCLC patients with MET *** consensus addresses key areas,such as optimal testing timing,testing methods,testing strategies,quality control measures,and treatment *** offering standardized recommendations,this guidance aims to streamline diagnostic and therapeutic processes and enhance clinical decision-making for NSCLC with MET alterations.

TSNAdb: A Database for Tumor-specific Neoantigens from Immunogenomics Data Analysis

Tumor-specific neoantigens have attracted much attention since they can be used as biomarkers to predict therapeutic effects of immune checkpoint blockade therapy and as potential targets for cancer immunotherapy. In this study, we developed a comprehensive tumor-specific neoantigen database （TSNAdb v1.0）, based on pan-cancer immunogenomic analyses of somatic mutation data and human leukocyte antigen （HLA） allele information for 16 tumor types with 7748 tumor samples from The Cancer Genome Atlas （TCGA） and The Cancer Immunome Atlas （TCIA）. We predicted binding affinities between mutant/wild-type peptides and HLA class I molecules by NetMHCpan v2.8/v4.0, and presented detailed information of 3,707,562/1,146,961 potential neoantigens generated by somatic mutations of all tumor samples. Moreover, we employed recurrent mutations in combination with highly frequent HLA alleles to predict potential shared neoantigens across tumor patients,which would facilitate the discovery of putative targets for neoantigen-based cancer *** is freely available at http：//***/tsnadb.
