In vitro and in vivo infection of snakehead rhabdovirus (SHRV) and recombinant chimeric SHRVs in Penaeus vannamei to evaluate availability as a vector for heterologous protein expression

The development of viral vectors for heterologous protein expression in shrimp has been limited by the absence of stable shrimp cell lines capable of supporting consistent viral replication. This study addresses this critical challenge by exploring snakehead rhabdovirus (SHRV) as a potential vector that can propagate in fish-derived cell lines and simultaneously infect shrimp. Recombinant SHRVs (rSHRVs) expressing foreign proteins, including chimeric variants with glycoprotein substitutions from vesicular stomatitis virus (VSV) or spring viremia of carp virus (SVCV), were rescued through reverse genetic technology. In vitro experiments demonstrated that both native and chimeric rSHRVs effectively infected primary cultured lymphoid organ cells from Penaeus vannamei, resulting in the successful expression of reporter proteins. In vivo bioluminescence imaging (BLI) confirmed systemic expression of the foreign protein, with rSHRV-ALuc2 showing efficient infection and spread in shrimp tissues. Among the chimeric variants, rSHRV-ALuc2-GVSV exhibited rapid initial infectivity but did not significantly outperform native rSHRV in prolonged replication. The use of SHRV, which propagates robustly in fish-derived EPC cells while maintaining infectivity in shrimp, represents a significant step towards overcoming the lack of shrimp cell lines for the development of heterologous protein expression tools using recombinant viruses. In the non-invasive route, while oral administration was unsuccessful partially due to viral instability in feed, reverse gavage demonstrated partial success with detectable viral loads but no bioluminescence signals. This study highlights the potential of SHRV as a versatile viral heterologous protein expression vector for shrimp. Further research is required to enhance infection and expression efficiencies and develop practical, non-invasive methods for broader application in aquaculture.

Viral hemorrhagic septicemia virus (VHSV) is a significant pathogen causing mass mortalities in marine and freshwater fish worldwide. Accurate diagnosis through quantitative reverse transcription PCR (RT-qPCR) is essential to prevent its spread, but false negatives can compromise results. In this study, we evaluate the use of heat-inactivated snakehead rhabdovirus (SHRV) as an internal positive control (IPC) for VHSV diagnosis. SHRV's similarity to VHSV in viral structure and genome makes it an ideal IPC. The introduction of SHRV IPC into the RT-qPCR workflow can improve the reliability of diagnostic results by enabling the detection of technical failures during the RNA extraction or amplification process. Our results show that heat-inactivated SHRV preserved RNA integrity for IPC use, and SHRV IPC can provide a useful tool for detecting and interpreting false negatives without affecting assay sensitivity.

Effect of CXCL12-expressing viral hemorrhagic septicemia virus replicon particles on leukocytes migration and vaccine efficacy in olive flounder (Paralichthys olivaceus)

Viral replicon particles are single-cycle viruses defective for function(s) needed for viral replication, which allow them to be recognized as a safer form for the vaccination of animals compared to attenuated live viruses. However, deletion of genes that are critical for the induction of protective immunity can diminish the vaccine potential of viral replicon particles. Therefore, the manipulation of viral replicon particles to produce a molecular adjuvant can be a way to increase immunogenicity of vaccines based on viral replicon particles. Chemokines are a class of chemotactic cytokines that control the migration of diverse cells of vertebrates. CXC chemokine ligand 12 (CXCL12) binds to a receptor CXCR4, and CXCL12-CXCR4 signaling plays an important role in the migration of hematopoietic cells during embryogenesis and the attraction of leukocytes. In the present study, to evaluate the possible use of CXCL12 as a molecular adjuvant for an rVHSV-Delta G vaccine and to know differences between CXCL12a and CXCL12b in the adjuvant ability, we rescued VHSV replicon particles that are expressing olive flounder CXCL12a, CXCL12b, or eGFP (rVHSV-Delta G-CXCL12a, rVHSV-Delta G-CXCL12b, or rVHSVAG-eGFP), and compared the ability to attract olive flounder leucocytes and to induce protection against a VHSV challenge. In the leukocytes migration assay, supernatants collected from cells infected with rVHSV-Delta G-CXCL12a and rVHSV-Delta G-CXCL12b showed significantly higher ability to attract olive flounder leukocytes than the supernatant of cells infected with rVHSV-Delta G-eGFP. Moreover, the significantly higher number of leukocytes were attracted to rVHSV-CXCL12a supernatant compared to rVHSV-CXCL12b supernatant, suggesting that CXCL12a would be more appropriate for the induction of immunity than CXCL12b in olive flounder. In the immunization experiment, olive flounder immunized with rVHSV-iG-CXCL12a showed significantly higher survival rate than fish immunized with rVHSV-D

Intramolecular Double Activation by Biligands Sharing a Single Metal Atom for Preferred Two-Electron Oxygen Reduction

It is challenging to selectively promote the two-electron oxygen reduction reaction (2e-ORR) since highly ORR-active electrocatalysts are not satisfied with 2e-ORR and are most likely to go all the way to 4e-ORR, completely reducing dioxygen to water. Recently, however, the possibility of a 2e-ORR preference over 4e-ORR was raised by extensively considering multiple ORR mechanisms and employing a potential-dependent activity measure for constructing volcano plots. Here, we realized the preferred 2e-ORR via an intramolecular double activation of the peroxide intermediate (*OOH) by allowing the intermediate to be easily desorbed before proceeding to 4e-ORR. Dioxygen was transformed to *OOH on a carbon atom of the imidazole ligand of zeolitic imidazolate framework-8 (ZIF-8). When an amine group was introduced via ligand exchange, the selectivity of 2e-ORR was enhanced by 11%. The added amine attracted the oxygen atom of *OOH via a hydrogen bond to weaken the binding strength of *OOH to the carbon active site (double activation). The amine-decorated ZIF-8 exhibited H2O2 faradaic efficiency at 98.5% at ultrahigh-rate production at 625 mg cm-2 h-1 by 1 A cm-2 in a flow cell.

A Chemical Switch System to Modulate Chimeric Antigen Receptor T Cell Activity through Proteolysis-Targeting Chimaera Technology

Despite the excellent efficacy of chimeric antigen receptor (CAR T) cell therapy, concerns about its safety have been constantly raised. The side effects of CAR T cells result from an aberrantly upregulation of CAR T cell activity. Therefore, it is crucial to control the CART cell activity whenever the patient is at risk. For this purpose, the iCas9 system, which induces apoptosis in CAR T cell through caspase-9 dimerization by compound, has been invented and is currently going under clinical trial. However, the iCas9 system is irreversible, as the entire CART cell population is removed from the patient. Thus, CAR T cells, which are very expensive, should be reinfused to the patients after they recovered from the side-effect. Here, we propose a new CAR T cell safety strategy, which targets CAR "protein", not CAR "T cell". In this system, the CAR construct is modified to bear a bromodomain (BD). The addition of a BD in the CAR protein did not interfere with the original CAR functions, such as cytokine secretion and target cell lysis. Our data showed that the use of a proteolysis-targeting chimaera (PROTAC) compound against BD successfully degraded the BD-containing CAR protein Moreover, the CAR expression is recovered when the PROTAC compound is removed from the cell, demonstrating that our system is reversible. In a target cell lysis assay, the PROTAC compound successfully suppressed the lytic activity of CAR T cells by degrading the CAR protein. In conclusion, we developed a new safety system in which CAR T cells can be "reversibly" controlled by a compound.

This study aims at identifying the structural dynamic characteristics using an ambient vibration, and developing a health monitoring system which adopts damage detecting algorithms. One of the main problems for this system design is to measure long-time dynamic response signals and simultaneously estimate the structural dynamic properties, In order to be suitable for a long-time monitoring, we conduct an ambient dynamic testing for a 3-floor moment resistance steel structure and analyze structural dynamic characteristics using time domain estimation techniques. Also, a damage detecting test is performed to evaluate damage state by various detecting algorithms (modal correlation method (MAC & COMAC), eigen-parameter change method). Finally, this paper suggests the optimal algorithms for the identification of the structural damage locations and damage quantities with all such comparisons. The algorithms presented in this paper prove to be applicable in structural health monitoring of structures.

Gallium nitrate ameliorates type II collagen-induced arthritis in mice

Rheumatoid arthritis (RA) is a chronic autoimmune inflammatory disease. Gallium nitrate has been reported to reserve immunosuppressive activities. Therefore, we assessed the therapeutic effects of gallium nitrate in the mouse model of developed type II collagen-induced arthritis (CIA). CIA was induced by bovine type II collagen with Complete Freund's adjuvant. CIA mice were intraperitoneally treated from day 36 to day 49 after immunization with 3.5 mg/kg/day, 7 mg/kg/day gallium nitrate or vehicle. Gallium nitrate ameliorated the progression of mice with CIA. The clinical symptoms of collagen-induced arthritis did not progress after treatment with gallium nitrate. Gallium nitrate inhibited the increase of CD4+ T cell populations (p < 0.05) and also inhibited the type II collagen-specific IgG2a-isotype autoantibodies (p < 0.05). Gallium nitrate reduced the serum levels of TNF-alpha, IL-6 and IFN-gamma (p < 0.05) and the mRNA expression levels of these cytokine and MMPs (MMP2 and MMP9) in joint tissues. Western blotting of members of the NF-kappa B signaling pathway revealed that gallium nitrate inhibits the activation of NF-kappa B by blocking I kappa B degradation. These data suggest that gallium nitrate is a potential therapeutic agent for autoimmune inflammatory arthritis through its inhibition of the NF-kappa B pathway, and these results may help to elucidate gallium nitrate-mediated mechanisms of immunosuppression in patients with RA. AMP-activated protein kinase (AMPK) plays a central role in controlling hepatic lipid metabolism through modulating the downstream acetyl CoA carboxylase (ACC) and sterol regulatory element-binding protein-1c (SREBP-1c) pathway. Saponins, particularly platycodin D, from the roots of Platycodon grandiflorum (Changkil saponins, CKS) have a variety of pharmacological properties, including antioxidant and hepatoprotective properties. The aim of this study was to investigate the effects of CKS on hepatic lipogenesis and on the expression of genes involved in lipogenesis, and the mechanisms involved. CKS attenuated fat accumulation and the induction of the lipogenic genes encoding SREBP-1c and fatty add synthase in the livers of HFD-fed rats and in steatotic HepG2 cells. Blood biochemical analyses and histopathological examinations showed that CKS prevented liver injury. CKS and platycodin D each increased the phosphorylation of AMPK and acetyl-CoA carboxylase in HFD-fed rats and HepG2 cells. The use of specific inhibitors showed that platycodin D activated AMPK via SIRT1/CaMKK beta in HepG2 cells. This study demonstrates that CKS or platycodin D alone can regulate hepatic lipogenesis via an AMPK-dependent signalling pathway. (C) 2013 Elsevier Inc. All rights reserved.

We have developed a nitrogen-incorporated silicon oxide (SiOxNy) deposition process using plasma enhanced atomic layer deposition (PEALD) for the gate insulator of recessed-gate Al-GaN/GaN metal-insulator-semiconductor heterojunction field-effect transistors. The SiOxNy film deposited on a recessed GaN surface exhibited a breakdown field of 13.2 MV/cm and a conduction band offset of 3.37 eV, which are the highest values reported for GaN MIS structures to the best of our knowledge. The fabricated normally-off transistor exhibited very promising characteristics such as a threshold voltage of 2.2 V, a maximum drain current density of 428 mA/mm, and a breakdown voltage of 928 V.

Comparison of elemental contents of Korean space foods using instrumental neutron activation analysis

The analysis of mineral contents in space foods is needed to obtain an information on a comprehensive elemental composition as well as the investigation on the effects of human nutrition and health based on the dietary intake of mineral elements. Recently, six items of new Korean space foods (KSFs) such as kimchi, bibimbap, bulgogi, a ramen, a mulberry beverage and a fruit punch which was developed by the KAERI, and the contents of more than 15 elements in the samples were examined by using instrumental neutron activation analysis (INAA). Five biological certified reference materials, NIST SRM were used for analytical quality control. The results were compared with those of common Korean foods reported, and these results will be applied toward the identification of irradiated foods.
