Relationships between Secreted Aspartyl Proteinase 2 and General Control Nonderepressible 4 gene in the Candida albicans resistant to itraconazole under planktonic and biofilm conditions

This study aimed to explore the roles of SAP2 and GCN4 in itraconazole (ITR) resistance of C. albicans under different conditions, and their correlations. A total of 20 clinical strains of C. albicans, including 10 ITR resistant strains and 10 sensitive strains, were used. Then, SAP2 sequencing and GCN4 sequencing were performed, and the biofilm formation ability of different C. albicans strains was determined. Finally, real-time quantitative PCR was used to measure the expression of SAP2 and GCN4 in C. albicans under planktonic and biofilm conditions, as well as their correlation was also analyzed. No missense mutations and three synonymous mutation sites, including T276A, G543A, and A675C, were found in SAP2 sequencing. GCN4 sequencing showed one missense mutation site (A106T (T36S)) and six synonymous mutation sites (A147C, C426T, T513C, T576A, G624A and C732T). The biofilm formation ability of drug-resistant C. albicans strains was significantly higher than that of sensitive strains (P < 0.05). Additionally, SAP2 and GCN4 were up-regulated in the ITR-resistant strains, and were both significantly higher in C. albicans under biofilm condition. The mRNA expression levels of SAP2 and GCN4 had significantly positive correlation. The higher expression levels of SAP2 and GCN4 were observed in the ITR-resistant strains of C. albicans under planktonic and biofilm conditions, as well as there was a positive correlation between SAP2 and GCN4 mRNA expression.

Reprogramming adipose mesenchymal stem cells into islet β-cells for the treatment of canine diabetes mellitus

Background Islet transplantation is an excellent method for the treatment of type I diabetes mellitus. However, due to the limited number of donors, cumbersome isolation and purification procedures, and immune rejection, the clinical application is greatly limited. The development of a simple and efficient new method to obtain islet beta-cells is a key problem that urgently requires a solution for the treatment of type I diabetes mellitus. Methods In this study, Pbx1, Rfx3, Pdx1, Ngn3, Pax4 and MafA were used to form a six-gene combination to efficiently reprogram aMSCs (adipose mesenchymal stem cells) into ra-beta Cs (reprogrammed aMSCs-derived islet beta-cells), and the characteristics and immunogenicity of ra-beta Cs were detected. Feasibility of ra-beta Cs transplantation for the treatment of diabetes mellitus in model dogs and clinical dogs was detected. Results In this study, aMSCs were efficiently reprogrammed into ra-beta Cs using a six-gene combination. The ra-beta Cs showed islet beta-cell characteristics. The immunogenicity of ra-beta Cs was detected and remained low in vitro and increased after transplantation. The cotransplantation of ra-beta Cs and aMSCs in the treatment of a model and clinical cases of canine diabetes mellitus achieved ideal therapeutic effects. Conclusions The aMSCs were efficiently reprogrammed into ra-beta Cs using a six-gene combination. The cotransplantation of ra-beta Cs and aMSCs as a treatment for canine diabetes is feasible, which provides a theoretical basis and therapeutic method for the treatment of canine diabetes.

BackgroundThe colorectal cancer mortality rate in China has exceeded that in many developing countries and is expected to further increase owing to multiple factors, including the aging population. However, the optimal policy for colorectal cancer screening is *** synthesized the most up-to-date data using a 12-state Markov model populated with a cohort of Chinese men and women born during 1949-1988, and evaluated 16 conventional and 40 risk-tailored schemes for colorectal cancer screening, considering possible combinations of age (starting at 40 + years and ending at 75 years), frequency, and strategy (standard colonoscopy, fecal immunochemical testing with colonoscopy if positive, or risk-tailored). We projected the incidence and mortality of CRC, cost, and quality-adjusted life years for 2023-2038;and performed incremental cost-effectiveness, probability acceptability, and sensitivity analyses to identify the optimal scheme and the factors affecting this *** 2038, all standard colonoscopy, colonoscopy following fecal immunochemical testing, and risk-tailored schemes were effective in reshaping China's colorectal cancer trajectory, with relative reductions in colorectal cancer incidence and mortality rates of up to 34% and 33.7%, respectively, versus no screening. Two standard colonoscopy, one colonoscopy following fecal immunochemical testing, and four risk-tailored schemes were efficient using a starting age of 40 years. Among these options, a risk-tailored scheme (standard colonoscopy every 5 years for high-risk and annual fecal immunochemical testing screening for moderate-to-low-risk) had a high probability (31.1%) of being optimal (with >= 40% uptake for a high-risk population, in particular), given China's present per capita gross domestic product, and would yield the highest gain in quality-adjusted life years in 17 of 31 *** findings suggest the commencement of colorectal cancer screening at 40 years of age

Foley sound in movies and TV episodes is of great importance to bring a more realistic feeling to the audience. Traditionally, foley artists need to create the foley sound synchronous with the content occurring in the video using their expertise. However, it is quite laborious and time consuming. In this paper, we present FastFoley, a Transformer based non-autoregressive deep-learning method that can be used to synthesize a foley audio track from the silent video clip. Existing cross-model generation methods are still based on autoregressive models such as long short-term memory (LSTM) recurrent neural network. Our FastFoley offers a new non-autoregressive framework on the audio-visual task. Upon videos provided, FastFoley can synthesize associated audio files, which outperforms the LSTM based methods in time synchronization, sound quality, and sense of reality. Particularly, we have also created a dataset called Audio-Visual Foley Dataset(AVFD) for related foley work and make it open-source, which can be downloaded at https:// ***/thuhcsi/icassp2022-FastFoley.

Lactate levels in the brain and blood of schizophrenia patients: A systematic review and meta-analysis

Background: The pathophysiological mechanisms of schizophrenia are still unclear. Converging evidence suggests that energy metabolism abnormalities are involved in schizophrenia, and support its role in the pathophysiology of this disease. Lactate plays an important role in energy metabolism. Many studies have reported changes in the levels of lactate in the brain and serum of schizophrenia patients;however, the results from these studies are not consistent. To overcome this limitation, the goal of the present meta-analysis is to analyze the changes in lactate levels in the brain and blood of schizophrenia ***: For this systematic review and meta-analysis, we performed a thorough search of relevant literature in the English language, using the MEDLINE, Cochrane, and Embase databases. Results: In the present meta-analysis, 20 studies were scrutinized, including 13 studies on brain lactate levels, which involved 322 schizophrenia patients and 324 healthy individuals as controls. 7 studies on blood lactate levels, involving 234 schizophrenia patients and 238 healthy individuals, were also included. Brain lactate levels were elevated in schizophrenia patients, both in vivo and in post-mortem studies. Nevertheless, blood lactate levels in schizophrenia patients have revealed no statistically significant difference, as compared with control ***: In comparison with healthy individuals, schizophrenia patients had higher lactate levels in the brain, rather than in the blood. These findings suggest independent regulatory mechanisms of lactate levels in the brain and peripheral tissues. Abnormal lactate metabolism in the brain may be an important pathological mechanism in schizophrenia.

State Capacity in Response to COVID-19: A Case Study of China

Countries across the world responded very differently in their prevention and control of coronavirus disease 2019 (COVID-19), and state capacity is likely driving these differences. In this study, we define state capacity in preventing and controlling COVID-19 as the extent to which a state takes rapid intervention measures based on scientific evidence to prevent and control infectious diseases. This case study explores China's experience in terms of pandemic prevention and control, showing that the goal of pandemic prevention and control in the country is a concrete combination of outcome goals and multidimensional process objectives. This research also demonstrates the important role of state capacity in pandemic prevention and control in China by analyzing different ways to quickly achieve accessibility and full coverage of intervention measures for the target population. Finally, we argue that a country's political system is not a decisive factor in pandemic prevention and control. Rather, the historical experience of a country in dealing with similar outbreaks and current state capacity play important roles.

Shooting Intelligent Detection System has important applications in the military industry. Existing target shooting systems mainly face problems such as high cost, complex operation, and poor environmental adaptability. Considering the above limitations, in this paper we propose a fast, low-cost, and easy-to-deploy bullet hole recognition algorithm based on visual detection, which mainly consists of four parts: image preprocessing, extraction of target center, bullet hole extraction and ring value determination. In the process of bullet hole extraction, a method for segmenting effective target surface is proposed to cope with the problem of off-target bullet holes. Our method can not only accurately localize the bullet holes, but also obtain the exact ring value at that location. In addition, due to the robust adaptability of our method, it can even be applied to outdoor scenarios. © 2023 IEEE.

Generative adversarial network (GAN)-based neural vocoders have been widely used in audio synthesis tasks due to their high generation quality, efficient inference, and small computation footprint. However, it is still challenging to train a universal vocoder which can generalize well to out-of-domain (OOD) scenarios, such as unseen speaking styles, non-speech vocalization, singing, and musical pieces. In this work, we propose SnakeGAN, a GAN-based universal vocoder, which can synthesize high-fidelity audio in various OOD scenarios. SnakeGAN takes a coarse-grained signal generated by a differentiable digital signal processing (DDSP) model as prior knowledge, aiming at recovering high-fidelity waveform from a Mel-spectrogram. We introduce periodic nonlinearities through the Snake activation function and anti-aliased representation into the generator, which further brings desired inductive bias for audio synthesis and significantly improves the extrapolation capacity for universal vocoding in unseen scenarios. To validate the effectiveness of our proposed method, we train SnakeGAN with only speech data and evaluate its performance for various OOD distributions with both subjective and objective metrics. Experimental results show that SnakeGAN significantly outperforms the compared approaches and can generate high-fidelity audio samples including unseen speakers with unseen styles, singing voices, instrumental pieces, and nonverbal vocalization.

Genome-wide association studies (GWASs) have reported multiple single nucleotide polymorphisms (SNPs) associated with schizophrenia, yet the underlying molecular mechanisms are largely unknown. In this study, we aimed to identify schizophrenia relevant genes showing alterations in mRNA and protein expression associated with risk SNPs at the 10q24.32-33 GWAS locus. We carried out the quantitative trait loci (QTL) and summary data-based Mendelian randomization (SMR) analyses, using the PsychENCODE dorsolateral prefrontal cortex (DLPFC) expression QTL (eQTL) database, as well as the ROSMAP and Banner DLPFC protein QTL (pQTL) datasets. The gene CNNM2 (encoding a magnesium transporter) at 10q24.32-33 was identified to be a robust schizophrenia risk gene, and was highly expressed in human neurons according to single cell RNA-seq (scRNA-seq) data. We further revealed that reduced Cnnm2 in the mPFC of mice led to impaired cognition and compromised sensorimotor gating function, and decreased Cnnm2 in primary cortical neurons altered dendritic spine morphogenesis, confirming the link between CNNM2 and endophenotypes of schizophrenia. Proteomics analyses showed that reduced Cnnm2 level changed expression of proteins associated with neuronal structure and function. Together, these results identify a robust gene in the pathogenesis of schizophrenia.
