Theabrownin remodels the circadian rhythm disorder of intestinal microbiota induced by a high-fat diet to alleviate obesity in mice

The intestinal microbiota undergoes diurnal compositional and functional oscillations within a day, which affect the metabolic homeostasis of the host and exacerbate the occurrence of obesity. TB has the effect of reducing body weight and lipid accumulation, but the mechanism of improving obesity caused by a high-fat diet based on the circadian rhythm of intestinal microorganisms has not been clarified. In this study, we used multi-omics and imaging approaches to investigate the mechanism of TB in alleviating obesity in mice based on the circadian rhythm of gut microbiota. The results showed that TB could significantly regulate the levels and rhythmic expression of serum lipid indicators (TG, TC, LDL) and serum hormones (MT, FT3, LEP, CORT). The number of intestinal microbiota colonizing the colonic epithelium underwent daily fluctuations. TB remodeled the rhythmic oscillation of gut microbes (i.e., Lachnospiraceae_NK4A136_group, Alistipes, etc.), including the number, composition, abundance and rhythmic expression of the biogeographic localization of microbes. TB notably reduced the levels of 16 bile acids (TCA, THDCA, TCDA, GHDCA, T-alpha-MCA, etc.) and restored the balance of bile acid metabolism. It was found that TB may mitigate high-fat diet-induced obesity in mice by reshaping the circadian rhythm of the gut microbiome and regulating bile acid metabolism.

CircZfp644-205 inhibits osteoblast differentiation and induces apoptosis of pre-osteoblasts via sponging miR-455-3p and promoting SMAD2 expression

Background Circular RNAs (circRNAs) are involved in the progression of osteoporosis;however, their impact on osteogenic differentiation has yet to be fully elucidated. In this study, we identified a novel circRNA known as circZfp644-205 and investigated its effect on osteogenic differentiation and apoptosis in *** CircZfp644-205, miR-445-3p, and SMAD2 levels were measured using quantitative real-time polymerase chain reaction (qRT-PCR). MC3T3-E1 cells were subjected to microgravity (MG) to establish a cell model. Osteogenic differentiation was assessed using qRT-PCR, Alizarin Red S staining, alkaline phosphatase staining, and western blot. The apoptosis was evaluated using flow cytometry. The relationship between miR-445-3p and circZfp644-205 or SMAD2 was determined using bioinformatics, RNA pull-down, and luciferase reporter assay. Moreover, a hindlimb unloading mouse model was generated to investigate the role of circZfp644-205 in vivo using *** CircZfp644-205 expression was up-regulated significantly in HG-treated MC3T3-E1 cells. Further in vitro studies confirmed that circZfp644-205 knockdown inhibited the osteogenic differentiation and induced apoptosis of pre-osteoblasts. CircZfp644-205 acted as a sponge for miR-455-3p, which reversed the effects of circZfp644-205 on pre-osteoblasts. Moreover, miR-455-3p directly targeted SMAD2, thus inhibiting the expression of SMAD2 to regulate cellular behaviors. Tailoring Co-crystallization over Microphase Separation in Conjugated Block Copolymers via Rational Film Processing for Field- Effect Transistors

In sharp contrast to widely studied coil-coil block copolymers (BCPs), investigation into conjugated rod-rod BCPs is relatively few and limited in scope. Moreover, the ability to exquisitely tailor microphase separation (i.e., forming two dissimilar crystals from two blocks, respectively) and co-crystallization (i.e., co-crystals of two blocks) in conjugated rod-rod BCPs offers a robust route to scrutinize their processing-structure-property relationship. Herein, we report the tailoring of co-crystalline and microphase-separated structures in a family of poly(3-butylthiophene)-block-poly(3-hexylselenophene) (denoted P3BT-b-P3HS) with different molecular weights via three processing strategies (i.e., drop-casting, meniscus-assisted solution shearing (MASS), and post solvent annealing) and the subsequent exploration of the correlation between their different structures (i.e., microphase separation and co-crystallization) and charge transport properties for use in organic field-effect transistors (OFETs). In the case of drop-casting, a larger molecular weight of P3BT-b-P3HS and a faster rate of solvent evaporation are found to favor their co-crystallization over the microphase separation of P3BT and P3HS. Interestingly, the MASS process effectively enables a phase transition from initial microphase-separated P3BT-b-P3HS to their co-crystallization. By contrast, the co-crystalline structure of P3BT-b-P3HS in a certain molecular weight range originally attained from drop-casting could be transformed into a microphaseseparated structure upon post solvent annealing. Afterward, the correlation between the crystalline structures yielded from the three processing strategies and charge mobilities of P3BT-b-P3HS is established. BackgroundCancer is still a leading cause of mortality. Over the years, cancer therapy has undergone significant advances driven by advancements in science and technology. A promising area of drug discovery in this field involves the development of therapeutic targets for cancer treatment. The urgent need to identify new pharmacological targets arises from the impact of tumor resistance on the effectiveness of current medications. Specifically, the RPS6KB2 gene on chromosome 11 has been implicated in cell cycle regulation and exhibits higher expression levels in tumor tissue. Given this association, there is a potential for this gene to serve as a target for cancer *** conducted an analysis using the GTEx, TCGA, and CCLE databases to explore the relationship between RPS6KB2 and immune infiltration, the tumor microenvironment (TME), microsatellite instability (MSI), and more. Cell proliferation was assessed using EDU detection, while cell invasion and migration were evaluated via wound healing and Transwell assays. Additionally, western blot analysis was employed to measure expression of Bax, Bcl-2, MMP2, MMP9, PCNA, and proinflammatory *** data analysis and molecular biology methods, our study carefully examined the potential role of RPS6KB2 in cancer therapy. The data revealed that RPS6KB2 is aberrantly expressed in most cancers and is associated with poor prognosis. Further analysis indicated its involvement in cancer cell apoptosis and migration, as well as its role in cancer immune processes. We validated the significance of RPS6KB2 in hepatocellular carcinoma (HCC), highlighting its capacity to upregulate proinflammatory *** research indicates that RPS6KB2 is a prognostic biomarker associated with immune infiltration in cancer that can affect antitumor immunity by increasing secretion of proinflammatory factors, providing a potential drug target for cancer treatment.

Efficient and Ultrafast Oxyorthosilicate Scintillator by Activator Valence State Manipulation

There is an urgent need for faster, brighter, and more controllable scintillation materials in advanced nuclear medicine, high-energy physical experiments, and dark matter particle detection. Nevertheless, the trade-off between high emission efficiency and fast timing characteristics remains a common challenge in the entire optical field. To address this issue, we develop a composition engineering strategy that involves multisite selective doping. This strategy aims to transform nearly all Ce3+ into fast-emitting Ce4+ while synergistically suppressing the electron traps. Even at very low doping concentrations, the designed Ca2+, Al3+, and Ce3+ tridoped oxyorthosilicate exhibits a scintillation decay (tau(d)) acceleration of 20%, accompanied by a 25% increase in light yield (LY). The ratio of emission efficiency and timing characteristics (LY/tau(d)) can be enhanced by 56%, which realizes the perfect balance of high LY and fast tau(d). Our work provides a method for designing efficient, ultrafast, and controllable scintillators in multicomponent systems, thus paving the way for high-resolution radiation detection and imaging applications.

Substrate-induced hybridization of plasmon modes in the composite nanostructure of nanodisk array/thin film for spectrum modulation

Hybridization coupling among plasmon modes is an effective approach to manipulate near-field properties thus optical spectral shapes of plasmonic nanostructures. Generally, mode hybridization coupling is achieved by modifying the topography and dimensions of nanostructures themselves, with few concerns about substrate-induced manipulation. Herein, we propose a composite nanostructure consisting of a gold (Au) nanodisk array and a thin Au film supported by a dielectric substrate. In this configuration, both the refractive index of the dielectric substrate and thin gold film's thickness mediate the interaction of plasmon modes supported by upper and lower interfaces of the composite nanostructure, resulting in two hybridized plasmon modes. We systematically investigate the relationship between optical fields at the top surface of plasmon modes before and after the hybridization coupling. Specifically, the near-field amplitude at the top surface of the unhybridized modes is stronger than that of individual hybridized mode, and lower than the near-field summation of these two hybridized modes. This work not only provides a straightforward strategy for generating two plasmon modes in a nanostructure but also elucidates the variation of the optical field during the hybridization process, which is of crucial significance for applications, such as upconversion enhancement and multi-resonance sensing.

Analytical Solution of the Stress Intensity Factors of Multiple Closed Collinear Cracks

Purpose According to the interaction between multiple cracks, a method for calculating the stress intensity factors of multiple closed collinear cracks in an infinite plate is proposed. In addition, the effects of the crack angle, crack length, crack spacing, lateral pressure coefficient and crack surface friction coefficient on the stress intensity factors are analyzed. Method The integral equation of the stress intensity factors is derived by the superposition principle, and the approximate solution of the integral equation is derived by the Chebyshev polynomial. The results obtained are compared with the finite element solutions of the same problem and the solutions of other scholars to verify the correctness of the equation. Results The results show that the intensity factor solution obtained by the proposed method is in good agreement with the finite element solution and other scholars' solutions. A larger crack length, smaller crack spacing, smaller friction coefficient and smaller lateral pressure coefficient can produce larger stress intensity factors, and the stress intensity factors are maximized when the crack angle is 45 degrees. Conclusions It is proved that the method based on the initial solution and superposition principle can be effectively applied to the calculation of the stress intensity factors of multiple collinear cracks.

Transforming Polymorphs via Meniscus-Assisted Solution-Shearing Conjugated Polymers for Organic Field-Effect Transistors

The ability to tune polymorphs of conjugated polymers affords a robust platform for investigating the processing-structure-property relationship. However, simple and generalizable routes to polymorphs have yet to be realized. Herein, we report a viable meniscus-assisted solution-shearing (MASS) strategy to effectively modulate polymorphs (i.e., polymorphs I and II) of poly(3-butylthiophene) (P3BT) and scrutinize the correlation between the two different polymorphs and charge transport characteristics. Specifically, polymorph II exists solely in drop-cast P3BT films. Intriguingly, confined shearing of P3BT renders efficient transformation of polymorph II to I. The kinetics of polymorph transformation associated with the changes in molecular packing and thus photophysical properties are elucidated. The resulting organic field-effect transistors reveal a strong correlation of device performance to attained polymorphs and crystal orientations of P3BT. Such polymorph transformation via the convenient MASS technique can be readily extended to other conjugated polymers of interest. This study highlights the robustness of MASS in regulating polymorphs of conjugated polymers to interrogate their interdependence of processing, structure, and property for a wide range of optoelectronic applications.

Hydrolysis of 2D Nanosheets Reverses Rheumatoid Arthritis Through Anti-Inflammation and Osteogenesis

Rheumatoid arthritis (RA) is a kind of inflammation homeostasis disorder that dysfunctions the joints. Clinically, medications against RA focus simply on mitigating the focal inflammation, without considering pro-osteogenesis re-modeling of the bone microenvironment. In the present work, 2D layered calcium disilicide nanoparticles (CSNs) are fabricated by facile aqueous exfoliation. The hydrolysis of CSNs produces anti-oxidative H2, alkaline Ca(OH)2, and silica. These moieties play significant roles in anti-oxidation, anti-inflammation, and pro-osteogenesis resulting in considerably better RA therapeutic consequences than anti-inflammation alone. Hydrogen gas is validated to eliminate excessive hydroxyl radicals and regulate macrophage re-polarization;the generated Ca(OH)2 can neutralize the acidic microenvironment and inhibit the osteoclast activity;and, the dissolved Ca2+ can effectively complex with phosphates to mineralize Ca3(PO4)2, promoting the osteogenesis of the focal joint. The multifunctional performances of CSNs are further confirmed in arthritic mouse and rabbit models, providing an advanced and robust therapeutic strategy against RA with high biocompatibility and clinical transformable promises.

Ursolic acid, a natural endogenous compound, inhibits browning in fresh-cut apples

Enzymatic browning occurs rapidly in fresh-cut apples and can lead to economic losses. Although various methods have been developed to prevent this browning, physical methods are costly, and chemical methods are controversial, suggesting the potential usefulness of an endogenous natural browning inhibitor. In this study, the effects of ursolic acid (UA), a natural endogenous compound, on apple browning were investigated. It was found that UA inhibited the activity of polyphenol oxidase through Cu2+ chelation. Molecular docking indicated that the inhibition was reversible and mixed-type. Moreover, UA treatment promoted the scavenging of reactive oxygen species in fresh-cut apple and reduced the proliferation of microorganisms on the apple surface. Similar effects were observed on the browning of chestnut, eggplant, potatoes, and pears. In conclusion, the findings demonstrate the efficacy of UA on inhibiting browning in fresh-cut apple, together with an analysis of the mechanism involved.
