Potent in vitro chondrogenesis of CD105 enriched human adipose-derived stem cells

Adipose-derived stem cells (ASCs) are considered as a promising cell source for cartilage regeneration. However, the heterogeneity of this cell source may affect their ability in cartilage formation. It is therefore necessary to establish an efficient method for isolating the cells that have chondrogenic potential. To date, no specific markers have been reported to be able to isolate such a cell population from human adipose tissue. In recent studies, endoglin (CD105) has been known as a relatively specific marker for identifying mesenchymal stem cells, but no studies show it is related to chondrogenic potential of human ASCs. In this study, human cells from adipose tissue were isolated, cultured, and sorted according to CD105 expression. The sorted cells were then subjected to adipogenic, osteogenic, and chondrogenic induction to confirm their multi-potentiality. In adipogenic conditions, CD105 cells showed stronger Oil Red staining and higher expression of adipose-specific genes compared to CD105+ cells. By contrast, CD105+ cells exhibited better osteogenic potential with stronger Alizarin Red staining and higher expression of osteogenic specific genes than CD105 cells. Noticeably, CD105+ cells also exhibited a much stronger chondrogenic potential than CD105 cells, with stronger collagen II staining and higher gene expression of collagen II and aggrecan. Most importantly, CD105+ cells could form a homogeneous cartilage-like tissue when seeded into a biodegradable scaffold and cultured in chondrogenic media for 8 weeks. These results indicate that sorting of ASC subpopulation with CD105 as a marker may allow better in vitro chondrogenesis and thus provide an important implications for cartilage regeneration and reconstruction using autologous cells from adipose tissue. Establishment and maintenance of three Chinese human embryonic stem cell lines

To establish a potential resource for cell therapy and a developmental model for human diseases, we had isolated three Chinese human embryonic stem cell lines from the inner cell mass of human blastocysts in 2002. All the three cell lines were grown on mouse embryonic fibroblasts as feeder cells;one of these cell lines, chHES-3, has maintained its normal karyotype even after being cultured in vitro for more than 100 passages, after the standardization of mouse feeder preparation. Each hES cell line has been completely characterized. All the three cell lines expressed hES-specific markers and pluripotency-related genes. These cells maintained their normal karyotype during long-term culture and displayed a high telomerase activity. When differentiated in vivo and in vitro, the derivatives representing the three germ layers could be observed. Human leukocyte antigen, ABO blood type, and DNA fingerprinting were also performed to provide a unique identity to each cell line. By establishing these hES cell lines, we provide an appropriate in vitro model to study human development and regeneration. A Ranging Method for OFDMA Uplink System

In orthogonal frequency division multiple access (OFDMA) uplink system, estimations of time delay and carrier frequency offset (CFO) are both particularly challenging. In IEEE 802.16-based networks, such synchronization problem can be resolved in initial ranging process. In this paper, exploiting the unique scheme of interleaved OFDMA, a special ranging symbol which has repetitive scheme in time-domain is designed. Furthermore, a novel ranging method for OFDMA system is proposed by utilizing such repetitive characteristic. Compared with the existing methods, this algorithm just need one ranging symbol and can get more correct estimation of time delay, at the same time, it can estimate the integral CFO in OFDMA uplink. Simulations illustrate efficiency of the proposed algorithm over the existing method(1).

Tracking control of pneumatic artificial muscle actuators based on sliding mode and non-linear disturbance observer

The dynamic properties and non-linear control of the pneumatic muscle actuator (PMA) were investigated in this study for use in a specially designed hand rehabilitation device. The phenomenological model of PMA was established in the lower pressure range applicable for hand rehabilitation. The experimental results show that PMA's characteristics can be approximated by piecewise functions. In order to improve the performance and robustness of control for accurate trajectory tracking, a sliding mode control based on non-linear disturbance observer (SMCBNDO) was designed. The simulation and experimental results demonstrated that the model and the sliding mode control achieved the desired performance in tracking a desired trajectory within guaranteed accuracy. The work indicates that the model and the non-linear control proposed in this study can be applied in PMA-driven hand function rehabilitation devices requiring lower pressures.

This article puts forward a novel routing architecture for complex optical network,which core component is path calculation element （PCE）.As is well known,the PCE-based distributed path computation structure is making the routing control and computation loosely coupled from traditional control *** the study,the resource allocation and routing algorithm are the critical part of PCE *** compare the performance of new architecture and the traditional one,the user uses the flooding suppression,routing delay,resource utilization and traffic blocking probability as performance simulation parameters and taking the verification simulation on objective modular network testbed （OMNeT） platform against to source-node routing *** numerical analysis,computer simulation and experiment work indicate that the operation of PCE-based routing architecture can reduce the flooding information of path calculation request as well as the routing hops significantly and improve the quality of service （QoS） by decreasing the blocking probability when failure happened.

Investigation on type-II Cu₂S-CdS core/shell nanocrystals: synthesis and characterization

Type-II Cu2S-CdS core/shell (including Cu2S/CdS and CdS/Cu2S) semiconductor nanocrystals (NCs). which call produce efficient spatial separation of electrons and holes between the core and shell, were Successfully synthesized. The results from absorption spectra, photoluminescence (PL) spectra. X-ray photoelectron spectra (XPS). X-ray diffraction (XRD), transmission electron microscopy (TEM), and PL lifetime measurements all confirmed the formation of the type-II core/shell NCs. The emission wavelength of the type-II Cu2S/CdS core/shell NCs can be readily tuned between 515 nm and 760 inn through two methods: core size control and shell thickness control. The highest PL quantum yield that the Cu2S/CdS core/shell NCs reached was similar to 12% by controlling the shell thickness, while the core Cu2S nanocrystals had no PL at all. The PL decay data revealed that Cu2S/CdS NCs had a dual exponential characteristic and the PL emission with the longest lifetime (more than 1400 ns) occupied more than 85%. After overcoating ZnS, a Cu2S/CdS/ZnS core/shell1/shell2 structure was formed, which resulted in a blue-shift of the emission wavelength. Using the synthesis strategy of Cu2S/CdS, CdS/Cu2S type-II NCs also produced tunable emissions in the visible range.

The solution-treated Mg-4Y-4Sm-0.5Zr alloy was extruded at temperatures from 325℃ to 500℃.Dynamic recrystallization(DRX) completely occurs when the alloy is extruded at 350℃and *** grains of the extruded alloy are obviously refined by the occurrence of *** average grain size of the extruded alloy increases with increasing the extrusion temperature,leading to a slight decrease of the ultimate tensile strength(UTS) and the yield strength(YS) .On the contrary,the UTS and YS of the extruded and aged alloy increase with increasing the extrusion *** of UTS of 400 MPa,YS larger than 300 MPa and elongation(EL) of 7%are achieved after extrusion at 400℃ and ageing at 200℃ for 16 *** grain refinement and precipitation are efficient strengthening mechanisms for the Mg-4Y-4Sm-0.5Zr alloy.

Optimum location of surface wells for remote pressure relief coalbed methane drainage in mining areas

Based on engineering tests in the Huainan coal mining area,we studied alternative well location to improve the performance of surface wells for remote pressure relief of coalbed methane in mining *** key factors,affecting location and well gas production were analyzed by simulation tests for similar *** exploitation results indicate that wells located in various positions on panels could achieve relatively better gas production in regions with thin Cenozoic layers,low mining heights and slow rate of longwall advancement,but their periods of gas production lasted less than 230 days,as opposed to wells in regions with thick Cenozoic layers,greater mining heights and fast rates of longwall *** near panel margins achieved relatively better gas production and lasted longer than centerline *** rules of development of mining fractures in strata over panels control gas production of surface *** fractures located in areas determined by lines of compaction and the effect of mining are well developed and can be maintained for long periods of *** the well at the end of panels and on the updip return airway side of panels,determined by lines of compaction and the effect of mining,would result in surface wells for remote pressure relief CBM obtaining their longest gas production periods and highest cumulative gas production.

Calculation of valence band structures of InAs/GaAs quantum ring and quantum dot:using numerical Fourier transform method

This article puts forward a new method in calculating the band structures of low-dimensional semiconductor structures. In this study, the valence band structures of InAs/GaAs quantum ring and lens-shaped quantum dot are calculated with four-band model, in the framework of effective-mass envelope function theory. To determine the Hamiltonian matrix elements, this article develops the numerical Fourier transform method instead of the widely used analytical integral method. The valence band mixing is considered. The hole energy levels change dramatically with the geometrical parameters of the quantum ring and quantum dot. It is demonstrated that numerical Fourier transform method can be adopted in low-dimensional structures with any shape. The results of Fourier transform method are consistent with the ones of analytical integral in literature; and they are helpful for studying and fabricating optoelectronic devices.

Using a cross-sectional case-control study of amnestic-mild cognitive impairment (aMCI), we characterised the relationships among cognitive function, serum levels of angiotensin converting enzyme (ACE), brain activity, and ACE insertion or deletion (VD) polymorphism. Forty-eight patients with aMCI and 36 well-matched normal controls were assessed by a comprehensive battery of standardized neuropsychological tests. In addition, regional homogeneity (ReHo) approaches were used to analyze blood oxygen level-dependent functional magnetic resonance imaging data on the resting state in all subjects, and genotyping of the serum ACE was measured in aMCI patients. The D carriers with aMCI patients were found to have markedly higher serum ACE levels than I homozygote carriers. Importantly, compared with the carried I homozygote group of patients with aMCI, the D carriers of aMCI patients were significantly impaired in the AVLT-delayed recall and had decreased ReHo over the bilateral precuneus, left middle occipital gyrus, right inferior parietal lobe, and right angular gyrus, whilst increased ReHo was found mainly in the left medial frontal gyrus, right paracentral lobe, and right anterior cingulate cortex. The findings indicated that ACE genotype was associated with episodic memory, serum levels of ACE, and resting-state brain activity in aMCI patients, and the findings of cognitive function and brain activity further suggests that the ACED allele may have a specific role in semantic memory dysfunction and brain activity in aMCI.
