目的:观察维生素C、e对五周龄生长期染铅SD大鼠血铅及海马超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、NOS活性及丙二醛(MDA)、NO含量的影响。方法:大鼠自由饮用0.615mmol/L的醋酸铅4周造成铅中毒模型。然后以维生素C 100 mg/kg、维生素e 100mg/kg单独或联合灌胃1周。测量血铅水平,大鼠海马组织SOD、GSH-Px、NOS活性及MDA和NO含量。结果:与铅模型组比较,给予维生素C,维生素e后,大鼠血铅浓度显著性降低(P<0.05);MDA含量下降,且其联合治疗组与单独维生素C治疗组差异有显著性(P<0.05);SOD,GSH-Px、NO、NOS水平显著高于铅模型组,差异有显著性(P<0.05)。结论:补充维生素C、维生素e可以降低血铅含量,减轻铅中毒引起的海马的脂质过氧化损伤,提高NOS、NO活性。

目的:探讨硫酸镁、维生素e单独应用和联合应用对雄性小鼠实验性肝损伤的保护作用。方法:3月龄雄性小鼠40只随机分设正常对照组、病理模型组、镁离子保护组;在保护组小鼠的饮水中分别加入硫酸镁、硫酸镁和维生素e、维生素e,饲养10d,腹腔注射0.15%CCl4致小鼠肝损伤,观察硫酸镁、维生素e单独应用和联合应用对小鼠实验性肝损伤肝中丙二醛(MDA)的含量,血清中ALT和AST以及肝细胞的形态变化。结果:镁离子和维生素e能显著降低血清中ALT和AST以及肝细胞中脂质过氧化物MDA的含量,减轻肝细胞气球样变性及坏死及炎性细胞浸润,硫酸镁和维生素e合用有协同效应。结论:硫酸镁和维生素e对雄性小鼠实验性肝损伤有保护作用。

Alzheimer's disease is one of the most challenging brain disorders and has profound medical and social consequences. It affects approximately 15 million persons worldwide, and many more family members and care givers are touched by the disease. The initiating molecular event(s) is not known, and its pathophysiology is highly complex. However, free radical injury appears to be a fundamental process contributing to the neuronal death seen in the disorder, and this hypothesis is supported by many (although not all) studies using surrogate markers of oxidative damage. In vitro and animal studies suggest that various compounds with antioxidant ability can attenuate the oxidative stress induced by beta-amyloid. Recently, clinical trials have demonstrated potential benefits from treatment with the antioxidants, vitamin e, selegiline, extract of Gingko biloba, and idebenone. Further studies are warranted to confirm these findings and explore the optimum timing and antioxidant combination of such treatments in this therapeutically frustrating disease. Background Patients with peripheral arterial disease (PAD) have high rates of cardiovascular morbidity and mortality, including that caused by associated coronary heart disease and cerebrovascular disease. Previous studies have shown that coagulation parameters are altered in PAD and that altered coagulation may play a critical role in the susceptibility to cardiovascular complications in PAD. It is therefore important to assess the effect of secondary prevention measures on coagulation in patients with PAD. The Arterial Disease Multiple intervention Trial (ADMIT), a multicenter, randomized, placebo-controlled trial, was conducted to determine the feasibility of a combined lipid-modifying, antioxidant, and antithrombotic treatment regimen in patients with PAD. The objective of this study was to assess the effect of the ADMIT interventions on coagulation. Methods ADMIT participants were randomly assigned to low-dose warfarin, niacin, and antioxidant vitamin cocktail or corresponding placebos in a 2 x 2 x 2 factorial design. Specialized coagulation studies were performed in a subset of 80 ADMIT participants at baseline and after 12 months of treatment. Results Low-dose warfarin (1 to 4 mg/d) resulted in a significant decrease in factor Vile (P< .001) and in plasma F1.2 (P = .001). Unexpectedly, niacin treatment also resulted in significant decrease in both fibrinogen (48 mg/dl;P< .001) and F1.2 (P= .04). von Willebrand factor increased after antioxidant vitamin treatment (P= .04). Conclusions A regimen of low-dose warfarin effectively modifies coagulation in patients with PAD. Niacin also favorably modifies Fibrinogen and plasma F1.2. Niacin, in addition to its lipid effects, modifies abnormal coagulation factors that accompany PAD.

Lung cancer is the major cause of death in industrialized western societies. Its link to tobacco abuse is well established and efforts should be made to eliminate this potent environmental carcinogen. The concept of chemoprevention, the use of agents to inhibit and reverse lung cancer carcinogenesis, has great appeal. The CAReT study, conducted in 18 000 high-risk smokers in the US, found that a combination of beta-carotene and retinyl palmitate resulted in a 28% increase in the incidence of lung cancer. A similar study conducted in Finland, the ATBC trial utilizing alpha tocopherol and beta-carotene, had similar findings for the group taking beta-carotene. These two trials have caused a rethinking of the use of natural compounds as chemoprevention agents. These agents should no longer be regarded as harmless, but as having potential toxicities. A new approach in the chemoprevention of cancer has been the concept of surrogate endpoints, biological changes that are on the pathway to cancer. Trials are underway to determine what are appropriate surrogate endpoints for lung cancer chemoprevention trials. (C) 2000 elsevier Science Ireland Ltd. All rights reserved.

effect of folic acid and antioxidant vitamins on endothelial dysfunction in patients with coronary artery disease

OBJeCTIVeS The purpose of this study was to determine whether lowering homocysteine levels with folic acid, with or without antioxidants, will improve endothelial dysfunction in patients with coronary artery disease (CAD). BACKGROUND elevated plasma homocysteine levels are a risk factor for atherosclerosis. Homocysteine may promote atherogenesis through endothelial dysfunction and oxidative stress. MeTHODS In a double-blind, placebo-controlled, randomized trial, we used vascular ultrasound to assess the effect of folic acid alone or with antioxidants on brachial artery endothelium-dependent flow-mediated dilation (FMD). Seventy-five patients with CAD (screening homocysteine level greater than or equal to 9 mu mol/liter) were randomized equally to one of three groups: placebo, folic acid alone or folic acid plus antioxidant vitamins C and e. Patients were treated for four months. ReSULTS Plasma folate, homocysteine, FMD and nitroglycerin-mediated dilation were measured before and after four months of treatment. Plasma folate, homocysteine and FIL LD were unchanged in the placebo group. Compared with placebo, folic acid alone increased plasma folate by 475% (p < 0.001), reduced plasma homocysteine by 11% (p = 0.23) and significantly improved FMD from 3.2 +/- 3.6% to 5.2 +/- 3.9% (p = 0.04). The improvement in FMD correlated with the reduction in homocysteine (r = 0.5, p = 0.01). Folic acid plus antioxidants increased plasma folate by 438% (p < 0.001), reduced plasma homocysteine by 9% (p = 0.56) and insignificantly improved FR ID from 2.6 +/- 2.4% to 4.0 +/- 3.7% (p = 0.45), as compared with placebo. Nitroglycerin-mediated dilation did not change significantly in any group. CONCLUSIONS Folic acid supplementation significantly improved endothelial dysfunction in patients with coronary atherosclerosis. Further clinical trials are required to determine whether folic acid supplementation may reduce cardiovascular events. (C) 2000 by the American College of Cardiology.

The effects of combined conventional treatment, oral antioxidants and essential fatty acids on sperm biology in subfertile men

We evaluated the effects of combined conventional treatment, oral antioxidants (N-acetyl-cysteine or vitamins A plus e) and essential fatty acids (FA) on sperm biology in an open prospective study including 27 infertile men. The evaluation included sperm characteristics, seminal reactive oxygen species (ROS), FA of sperm membrane phospholipids, sperm oxidized DNA (8-OH-dG), and induced acrosome reaction (AR). Treatment did not improve sperm motility and morphology, nor decrease the concentration of round cells and white blood cells in semen. Sperm concentration increased in oligozoospermic men (7.4 +/- 1.3 to 12.5 +/- 1.9 million/ml). Treatment significantily reduced ROS (mean +/- SeM) (775.3 +/- 372.2 to 150.3 +/- 105.2 x 10(3) counts/10 second) and 8-OH-dG (45.3 +/- 10.4 to 16.8 +/- 3.3 fmol/mug DNA). Treatment increased the AR (551 +/- 2.2 to 71.6 +/- 2.2 %), the proportion of polyunsaturated FA of the phospholipids, and sperm membrane fluidity. The overall pregnancy rate was 4.5% in 134 months. The per month pregnancy rate tended to be higher in partners of (ex)smokers (7.15%, n = 14, 70 months) than in never-smokers (1.6%, n = 13, 64 months) (OR:4.57, 95% Cl:0.55-38.1). In a 2 .times. 2 .times. 2 factorial experiment, protein intake, Se supplementation, and intake of P were related to the incidence of retained placenta in cows fed varying concentrations of these nutrients during the dry period. The population incidence of 38% retained placenta in 26 control cows was independent of P intake which ranged between 39-95 g/day. When a protein supplement was included in the concentrate as soybean meal, the incidence of retained placenta was reduced to 20% for supplemented animals from 50% for the animals that received no soybean meal. This increased protein in the ration was reflective of a mean increase from .02 ppm of Se to .06 ppm of Se in the ration. By increasing the mean daily intake of Se at least 3 wk prepartum from .23 mg to .92 mg daily, overall incidence of retained placenta was reduced from 38% to 0%. A positive prophylactic effect was achieved regardless of whether .alpha.-tocopherol was supplemented as well. These mature dairy cows were deficient in Se, and supplementation of Se reduced the incidence of retained placenta.
