Determination of anthocyanins and anthocyanidins in the wild grape (Vitis sylvestris Gmelin) by high-performance liquid chromatography-diode array detection (HPLC-DAD)

The determination of anthocyanins and anthocyanidins in wild grapes (Vitis sylvestris Gmelin) from Tunceli-Turkey are reported together with the influence of the extraction conditions on the results. Representative sets of anthocyanins (delphinidin-3-o-glucoside, cyanidin-3-o-glucoside, pelargonidin-3-o-glucoside, malvidin-3-o-glucoside) and anthocyanidins (delphinidin, cyanidin, pelargonidin, malvidin) were simultaneously determined by high performance liquid chromatography with diode array detection (HPLC-DAD). The extraction conditions including the solvent type, extraction time and liquid:solid ratio (v:w) were optimized. The anthocyanin and anthocyanidin contents changed significantly depending on the extraction conditions. Malvidin-3-o-glucoside was identified to be the primary compound.

Anion exchange polymeric sorbent coupled to high-performance liquid chromatography with UV diode array detection for the determination of ten N-nitrosamines in meat products: a validated approach

Among mechanisms and molecules presumably involved in the carcinogenicity induced by meat consumption, the N-nitrosamines (N-NAs) are a class of compounds characteristic of processed meats. In this work, an analytical method for the determination of ten N-nitrosamines (N-nitrosomorpholine, N-nitrosomethylethylamine, N-nitrosopyrrolidine, N-nitrosodiethylamine, N-nitrosopiperidine, N-nitrosodipropylamine, N-nitrosodimethylaniline, N-nitrosodibutylamine, N-nitrosodiphenylamine and N-nitrosodibenzylamine) in fresh meats and meat products was developed, optimised and validated. The method is based on optimised sample purification by solid-phase extraction (anion exchange polymeric sorbent) and separation/detection by high-performance liquid chromatography coupled to UV diode array detection. The validation procedure allowed to ascertaining good analytical performances in terms of sensitivity, selectivity towards interfering compounds, accuracy and robustness. The values obtained for precision (range: 4.3-14.4%) and recovery percentages (range: 80.8-95.1%) were compared to reference values indicated in the Decision No. 657/2002/EC, resulting as compliant. The measurement uncertainty (lower than 14.6%) was satisfactory for each N-NA as well.

Determination of major phenolic compounds in apples: Part I-Optimization of high-performance liquid chromatography separation with diode array detection

The separation of seven phenolic compounds including gallic acid, chlorogenic acid, epicatechin, quercitrin, rutin, phloridzin, and phloretin present in apple peel and pulp and differing in elution properties has been optimized using high-performance liquid chromatography with diode array detection. Several stationary phases were tested to achieve the efficient separation of phenolic compounds in fruit extracts and C18 was found to be the most efficient. Core-shell and fully porous C18 packings were assessed with respect to the complex composition of the fruit extracts. The developed high-performance liquid chromatography method comprised gradient elution in which mobile phase A was water at pH 2.8 adjusted with acetic acid and B was acetonitrile. The gradient shape was the following: 0 min 95% A/5% B, 2.5 min 85% A/15% B, 12 min 50% A/50% B, 15 min 95% A/5% B. The flow rate was 1 mL/min, injection volume 10 mu L, and UV detection at 255, 280, 320, and 365 nm was applied. Our method was validated for both C18 core-shell and fully porous packings. The resolution 6.2-14.8, symmetry 0.99-1.34, peak capacity 18-60, peak area repeatability 0.45-1.00% relative standard deviation, calibration range 0.125-5 mg/mL (0.2510 mg/mL for chlorogenic acid and rutin), correlation coefficients of calibration curve 0.9976-0.9997, and accuracy evaluated as recovery 95.56-107.54% were determined for the core-shell column.

Phytochemical Profiling and Evaluation of the Hepatoprotective Effect of Cuscuta Campestris by High-Performance Liquid Chromatography with diode array detection

Plant-derived antioxidant compounds have the potential to prevent cell damage caused by free radicals. As a holoparasitic plant, Cuscuta campestris Yuncker is being valorized for treatment of liver injury and cancer prevention in traditional medicine. The main purpose of this present study is to elucidate the antioxidant- and anticancer-associated contents of C. campestris by spectroscopic and chromatographic methods. Diethyl ether, ethyl acetate, methanol, n-butanol, and water were used as extraction solvents to reach a wide range of secondary metabolites synthesized by this plant. Antioxidant potentials of these extracts were characterized by (2,2-diphenyl-1-picrylhydrazyl)-free radical scavenging activity. Their anticancer activities were evaluated on SNU-398 hepatocellular carcinoma cells and controlled on the normal adult human dermal fibroblasts (hDFs) cells. Their phenolic compounds were analyzed by high-performance liquid chromatography with diode array detector to illuminate the responsible anticancer agent(s). The ethyl acetate extract revealed the most significant antioxidant effect. Methanol and ethyl acetate extracts were found to be cytotoxic on the SNU-398 cell lines with CC50 values of 18.7 and 19.6 mu gmL(-1), respectively, whereas these extracts were not cytotoxic to the adult hDFs cells. Due to their abundance in the extracts, isorhamnetin, kaempferol, and quercetin may have caused this anticancer activity. Methanol extract has the highest concentration of phenolic compounds. Based on chromatographic analyses, we propose that these anticancer effects were positively correlated with plant phenolic compounds. The results showed that this plant is worth further study for its therapeutic uses.

To control the quality of different forms of Suanzaoren decoction, an effective and reliable method for the simultaneous determination of 13 major components (neomangiferin, mangiferin, spinosin, liquiritin apioside, liquiritin, 6'''-feruloylspinosin, senkyunolide I, timosaponin BII, isoliquiritoside, timosaponin C, jujuboside A, jujuboside B, and timosaponin AIII) was developed and validated for the first time in this study using high-performance liquid chromatography with diode array detection and evaporative light scattering detection. The chromatographic separation was performed on a Venusil MP C-18 column (250 mm x 4.6 mm, 5 mu m) at 30 degrees C with a gradient of acetonitrile/redistilled water as the mobile phase. diode array detection was carried out at a wavelength of 275 nm. The drift tube temperature and the nitrogen gas flow rate of the evaporative light scattering detection were set at 50 degrees C and 1.6 L/min, respectively. The newly developed method was successfully applied to the determination of 13 components in lab-prepared Suanzaoren oral liquid, Suanzaoren mixture, and clinical Suanzaoren granules, and this study showed that this was a useful way to comprehensively evaluate the quality of Suanzaoren decoction in different forms of the preparation.

We introduce a gas chromatography detection approach that uses diode array detection operated in series with flame ionisation detection and demonstrate the utility of the detection approach for determination of volatile organic compounds. diode array detection brings ultraviolet - visible spectroscopy (over a range of 190-640 nm) onto the capillary gas chromatography time-scale, where average peak widths of analytes are on the order of 3-5 s. The non-destructive nature of the diode array detector affords serially coupled flame ionisation detection. This arrangement delivers near-simultaneous selective and universal detection without incurring additional analytical time, and without recourse to column flow splitting. The hyphenated technique is shown to be effective for chromatographic applications spanning an equivalent volatility range from C-1 to C-7 n-paraffin hydrocarbons. The approach introduced herein provides increased sensitivity and selectivity for classes of compounds amenable to electronic spectroscopy such as alkenes, dienes, sulfurs, and aromatic compounds. The approach is demonstrated for direct measurement of carbon disulfide in work place atmospheres with a detection limit of 93 pg on column and for the direct measurement of 1,3-butadiene in hydrocarbon matrices and ambient air with a detection of 73 pg on column, each in less than 5 min. This work presents a simple, sensitive and generic high-performance liquid chromatography with diode array detection method for the simultaneous determination of seven drugs prescribed for the treatment of erectile dysfunction and premature ejaculation. Investigated drugs include the phosphodiesterase-5 inhibitors: sildenafil, tadalafil, and vardenafil, in addition to the selective serotonin reuptake inhibitors: dapoxetine, duloxetine, fluoxetine, and paroxetine. The drugs were separated using a Waters C8 column (4.6 x 250 mm, 5 mu m) with the mobile phase consisting of phosphate buffer pH 3, acetonitrile and methanol in the ratio 60:33:7. The flow rate was 1.2 mL/min, and quantification was based on measuring peak areas at 225 nm. Peaks were perfectly resolved with retention times 3.3, 3.9, 6.4, 7.5, 9.5, 10.7, and 13.4 min for vardenafil, sildenafil, paroxetine, duloxetine, dapoxetine, fluoxetine, and tadalafil, respectively. The developed method was validated with respect to system suitability, linearity, ranges, accuracy, precision, robustness, and limits of detection and quantification. The proposed method showed good linearity in the ranges 5-500, 2-200, 2-200, 3-300, 1.5-150, 2-200, and 2-200 mu g/mL for sildenafil, tadalafil, vardenafil, dapoxetine, duloxetine fluoxetine, and paroxetine, respectively. The limits of detection were 0.18-0.38 mu g/mL for the analyzed compounds. The applicability of the proposed method to real life situations was assessed through the analysis of commercial tablets, and satisfactory results were obtained.

Simultaneous determination of atorvastatin and valsartan in human plasma by solid-based disperser liquid-liquid microextraction followed by high-performance liquid chromatography-diode array detection

A simple, sensitive, and efficient method has been developed for simultaneous estimation of valsartan and atorvastatin in human plasma by combination of solid-based dispersive liquid-liquid microextraction and high performance liquid chromatography-diode array detection. In the proposed method, 1,2-dibromoethane (extraction solvent) is added on a sugar cube (as a solid disperser) and it is introduced into plasma sample containing the analytes. After manual shaking and centrifugation, the resultant sedimented phase is subjected to back extraction into a small volume of sodium hydrogen carbonate solution using air-assisted liquid-liquid microextraction. Then the cloudy solution is centrifuged and the obtained aqueous phase is transferred into a microtube and analyzed by the separation system. Under the optimal conditions, extraction recoveries are obtained in the range of 81-90%. Calibration curves plotted in drug-free plasma sample are linear in the ranges of 5-5000 mu g L-1 for valsartan and 10-5000 mu g L-1 for atorvastatin with the coefficients of determination higher than 0.997. Limits of detection and quantification of the studied analytes in plasma sample are 0.30-2.6 and 1.0-8.2 mu g L-1, respectively. Intra-day (n = 6) and inter-days (n = 4) precisions of the method are satisfactory with relative standard deviations less than 7.4% (at three levels of 10, 500, and 2000 mu g L-1, each analyte). These data suggest that the method can be successfully applied to determine trace amounts of valsartan and atorvastatin in human plasma samples. 2016 Elsevier B.V. All rights reserved.

Determination of Tyrian purple by high performance liquid chromatography with diode array detection

Indigotin, indirubin, 6-bromoindigotin, 6'-bromoindirubin, 6-bromoindirubin, 6,6'-dibromoindigotin and 6,6'-dibromoindirubin, the colouring components of Tyrian purple, are quantified by an efficient HPLC method coupled to a diode array detector. The compounds were separated using gradient elution, on a RP-column (Alltima C18, 250 mm x 3.0 mm i.d., 5 mu m), thermostated at 35 degrees C, with a mobile phase consisting of solvents (A) H2O + 0.1% (v/v) trifluoroacetic acid and (B) acetonitrile + 0.1% (v/v) trifluoroacetic acid, at a flow rate of 0.5 mL min(-1). The method was validated in terms of linearity, detection and quantification limits, precision, accuracy, ruggedness and robustness, the latter with respect to small changes in column temperature and in flow-rate, pH- and solvent composition of the mobile phase. Under optimal conditions, the developed analytical scheme offers limits of detection in the range 0.02-0.05 mu g mL(-1) and satisfactory linearity up to 2.5 mu g mL(-1) for all analytes. Four samples produced from the hypobranchial glands of Hexaplex (Murex) trunculus molluscs, collected in the coastlines of Tunisia and Croatia, were treated with hot DMSO and analysed by the established HPLC method, using the standards addition approach. To evaluate the matrix effect, a comparison of the slopes of the standards in solvent regression curves with those of the standard addition method's calibration curves, using the Student's t-test was carried out. The accuracy was evaluated by recovery experiments. Amounts of indigotin, indirubin, and their mono- and dibrominated derivatives ranging between 0.01 to 12.2 mu g mg(-1) were found in the DMSO extracts of the four molluscan samples. Effective cleanup for the determination of six quinolone residues in shrimp before HPLC with diode array detection in compliance with the European Union Decision 2002/657/EC

A high-performance liquid chromatographic method was developed for the determination of six quinolone residues (ciprofloxacin, enrofloxacin, sarafloxacin, oxolinic acid, nalidixic acid, and flumequine) in shrimp tissue samples. Separation was carried out by a LiChrospher (R) 100 RP-8e column, running at a 22 min gradient elution program, and the mobile phase consisted of citric acid (0.4 mol/L), acetonitrile and methanol. detection was achieved by a diode array detector, monitoring at 255 and 275 nm. Sample preparation included initial extraction with citric acid solution and further clean-up by solid-phase extraction, employing Lichrolut RP-18 cartridges. Validation was performed according to the European Union Decision 2002/657/EC. The detection capability was 127.2 mu g/kg for ciprofloxacin, 115.2 mu g/kg for enrofloxacin, 126.2 mu g/kg for sarafloxacin, 113.1 mu g/kg for oxolinic acid, 125.2 mu g/kg for nalidixic acid, and 239.0 mu g/kg for flumequine. Recoveries ranged between 83.0 and 121.6%. The Youden test was applied to study the method ruggedness.
