In eukaryotes,microtubule polymers are essential for cellular plasticity and fate ***-binding(EB)proteins serve as scaffolds for orchestrating microtubule polymer dynamics and are essential for cellular dynamics and chromosome segregation in ***,we show that EB1 forms molecular condensates with TIP150 and MCAK through liquid–liquid phase separation to compartmentalize the kinetochore–microtubule plus-end machinery,ensuring accurate kinetochore–microtubule interactions during chromosome segregation in *** of EB1–TIP150 polymer formation by a competing peptide prevents phase separation of the EB1-mediated complex and chromosome alignment at the metaphase equator in both cultured cells and Drosophila ***220 of EB1 is dynamically acetylated by p300/CBP-associated factor in early mitosis,and persistent acetylation at Lys220 attenuates phase separation of the EB1-mediated complex,dissolves droplets in vitro,and harnesses accurate chromosome *** data suggest a novel framework for understanding the organization and regulation of eukaryotic spindle for accurate chromosome segregation in mitosis.

Transient liquid phase(TLP)bonding is a promising process for the joining and repairing of nickel-base *** of the most important parameters in TLP bonding is the bonding time required for suf-ficient isothermal solidification which prevents the formation of undesirable precipitated *** the present work,the effect of bonding time on the microstructure,type,and evolution of precipitates in the non-isothermal solidified zone(NSZ)and their effect on micro-mechanical properties were systemat-ically investigated using multi-scale tests in TLP bonded Mar-M247 superalloy joints with Ni-15.2Cr-3.74B interlayer at 1230℃.For a bonding time of 5 min,dual-phase M_(23)(C,B)_(6)-γ/γ’(where M is a mix-ture of Hf,Ta,Cr,and Ni)with eutectic configuration was formed in *** the increase in bonding time,the evolution of NSZ microstructure can be summed up as eutectic M_(23)(C,B)_(6)-γ/γ’,semi-striping dual-phase M_(23)(C,B)_(6)-γ/γ’,discontinuously striping M_(23)(C,B)_(6)-γ/γ’,followed by the disintegration of *** the NSZ counterpart,the isothermal solidified zone(ISZ)is mainly composed ofγ/γ’.Ac-companied by the dissolution of M_(23)(C,B)_(6) in the centerline,the proportion of the ISZ increases greatly until the joints are completely occupied by ***,a bamboo-like structure with domain size of-100μm was formed in the joint centerline,along withγ’reorganized themselves all into cubic shapes and distributed *** property tests demonstrated that in comparison to samples with longer bonding time,the NSZ of the shortest bonding time(5 min)has the highest strength and a subsequent decrease in strength was observed with prolonging the bonding time and post-bond heat ***,possible solidification/transformation path,segregation behavior,and formation mechanism of NSZ/ISZ evolution were discussed.

Selenopeptides may be a valuable bioactive compound to promote gut microbiota-targeted therapeutic methods for intestinal disease and ***,limited information is available on the utilization of selenopeptides by gut microbiota,especially Selenium(Se)*** this purpose,the present study aimed to investigate the protective effect of selenopeptide(RYNA(Se)MNDYT,Se-P2,purity of≥95%)and its original peptide(RYNAMNDYT,P2,purity of≥95%)in vivo by the microbiota-metabolite axis and further analyze the potential contribution of Se biofortification to Se-P2 *** results showed that Se-P2 exhibits a higher protective effect on lipopolysaccharide(LPS)-induced inflammation than P2,including pathology of the colon and liver,which suggested that the bioactivity of P2 was promoted by the organic combination of ***,gut microbiota composition tended to be a healthy structure by Se-P2 pretreatment in LPS-injured mice,which had a positive effect on LPS-induced gut microbiota ***,only Se-P2 promoted an increase in the relative abundance of Lactobacillus,Alistipes,and Roseburia and a decrease in the relative abundance of Akkermansia,Erysipelatoclostridium,and Bacteroides in LPS-injured *** changes in gut microbiota were obviously correlated with the changes in metabolites and affected the metabolic pathways of valine,leucine,isoleucine,phenylalanine,tyrosine,and tryptophan biosynthesis and phenylalanine *** may be one of the key reasons for Se-P2 to exert bioactivity through the microbiota-metabolite ***,Se-biofortification in Se-enriched Cordyceps militaris affected the parental proteins of Se-P2 to modulate mitogen-activated protein kinase,GPI anchored protein,and carbohydrate metabolism,translation,folding,sorting and degradation,which may contribute to the bioactivity of *** study provides information on the effect of Se on selenopeptides in vivo,which further promotes the prospective

Dietary parboiled rice(PR)has a low risk of disease,but little is known about the contribution of PR to the prevention of *** potential role and underlying mechanisms of PR in hyperlipidemia were evaluated in this *** C57BL/6J mice were fed with a normal diet,high-fat diet(HFD)containing refined rice(HFDRR)or PR(HFDPR).It was found that PR intervention improved lipid accumulation in *** data analysis revealed that 27 genes were up-regulated(mostly involved in lipid breakdown)and 86 genes were down-regulated(mostly involved in inflammatory responses)in the HFDPR group compared to the HFDRR *** 15 differentially expressed genes(DEGs)were validated by quantitative real-time PCR(RT-qPCR),while protein interaction network showed that protein tyrosine phosphatase receptor type C(PTPRC)has a central *** gut microbiota of mice was also altered after different dietary treatments,with higher ratio of Firmicutes and Bacteroidetes,increased abundances of Ruminococcaceae,Lachnospiraceae,Christensenellaceae,Porphyromonadaceae,Rikenellaceae and Prevotellaceae,and decreased abundances of Lactobacillaceae,Peptostreptococcaceae,Erysipelotrichaceae and Actinobacteria in the HFDRR *** addition,it was observed that PPAR signaling pathway may act as a bridge between DEGs and differential gut *** results suggested that PR can prevent hyperlipidemia by modulating liver genes and gut microbiota.

Heart failure with preserved ejection fraction(HFpEF)is a complex clinical syndrome with cardiac dysfunction,fluid retention and reduced exercise tolerance as the main *** treatment of HFpEF is using combined medications of related comorbidities,there is an urgent need for a modest drug to treat ***(GE),an iridoid glycoside extracted from Gardenia Jasminoides,has shown significant efficacy in the treatment of cardiovascular,digestive and central nervous system *** this study we investigated the therapeutic effects of GE on HFpEF experimental models in vivo and in *** was induced in mice by feeding with HFD and L-NAME(0.5 g/L)in drinking water for 8 weeks,meanwhile the mice were treated with GE(25,50 mg/kg)every other *** echocardiography and exhaustive exercise were performed,blood pressure was measured at the end of treatment,and heart tissue specimens were collected after the mice were *** showed that GE administration significantly ameliorated cardiac oxidative stress,inflammation,apoptosis,fibrosis and metabolic disturbances in the hearts of HFpEF *** demonstrated that GE promoted the transcriptional activation of Nrf2 by targeting MMP2 to affect upstream SIRT1 and downstream GSK3β,which in turn alleviated the oxidative stress in the hearts of HFpEF *** H9c2 cells and HL-1 cells,we showed that treatment with GE(1 μM)significantly alleviated H2O2-induced oxidative stress through the MMP2/SIRT1/GSK3β *** summary,GE regulates cardiac oxidative stress via MMP2/SIRT1/GSK3β pathway and reduces cardiac inflammation,apoptosis,fibrosis and metabolic disorders as well as cardiac dysfunction in HFpEF.

The plant cell wall is a dynamic and complex structure,serving as a crucial interface for both host defense and pathogen *** Arabidopsis thaliana leaves,pectin,a major component of primary cell walls,constitutes approximately 50%(w/w)of the cell wall(Zablackis et al.,1995).Pectin includes various polysaccharides,such as homogalacturonan(HG),xylogalacturonan,apiogalacturonan,rhamnogalacturonan I,and rhamnogalacturonan II,with HG being the most abundant and extensively *** research suggests that dynamic alterations in HG are key indicators of plant–microbe interactions at the cell wall interface(Du et al.,2022;Su,2023).Pectin is synthesized in the Golgi apparatus and transported to the cell wall in a methyl-esterified form,which is subsequently demethylated by pectin methylesterase(PME).The resulting unesterified pectin may form cross-linked structures with calcium ions or serve as a substrate for pectin lyase or polygalacturonase(PG),leading to its degradation(Du et al.,2022).

With the application of resins in various fields, numerous waste resins that are difficult to treat have been produced. The industrial wastewater containing Cr(Ⅵ) has severely polluted soil and groundwater environments, thereby endangering human health. Therefore, in this paper, a novel functionalized mesoporous adsorbent PPR-Z was synthesized from waste amidoxime resin for adsorbing Cr(Ⅵ). The waste amidoxime resin was first modified with H3PO4 and ZnCl_(2), and subsequently, it was carbonized through slow thermal decomposition. The static adsorption of PPR-Z conforms to the pseudo-second-order kinetic model and Langmuir isotherm, indicating that the Cr(Ⅵ) adsorption by PPR-Z is mostly chemical adsorption and exhibits single-layer adsorption. The saturated adsorption capacity of the adsorbent for Cr(Ⅵ) could reach 255.86 mg/g. The adsorbent could effectively reduce Cr(Ⅵ) to Cr(Ⅲ) and decrease the toxicity of Cr(Ⅵ) during adsorption. PPR-Z exhibited Cr(Ⅵ) selectivity in electroplating wastewater. The main mechanisms involved in the Cr(Ⅵ) adsorption are the chemical reduction of Cr(Ⅵ) into Cr(Ⅲ) and electrostatic and coordination interactions. Preparation of PPR-Z not only solves the problem of waste resin treatment but also effectively controls Cr(Ⅵ) pollution and realizes the concept of “treating waste with waste”.

Recently,there has been increasing interest in the use of bacteria for cancer therapy due to their ability to selectively target tumor sites and inhibit tumor ***,the complexity of the interaction between bacteria and tumor cells evokes unpredictable therapeutic risk,which induces inflammation,stimulates the up-regulation of cyclooxygenase Ⅱ(COX-2)protein,and stimulates downstream antiapoptotic gene expression in the tumor microenvironment to reduce the antitumor efficacy of chemotherapy and *** this study,we encapsulated celecoxib(CXB),a specific COX-2 inhibitor,in liposomes anchored to the surface of Escherichia coli Nissle 1917(ECN)through electrostatic absorption(C@ECN)to suppress ECN-induced COX-2 up-regulation and enhance the synergistic antitumor effect of doxorubicin(DOX).C@ECN improved the antitumor effect of DOX by restraining COX-2 *** addition,local T lymphocyte infiltration was induced by the ECN to enhance immunotherapy efficacy in the tumor *** the biosafety of C@ECN,a hypoxia-induced lysis circuit,pGEX-Pvhb-Lysis,was introduced into the ECN to limit the number of ECNs in *** results indicate that this system has the potential to enhance the synergistic effect of ECN with chemical drugs to inhibit tumor progression in medical oncology.
