Robust production of N-acetyl-glucosamine in engineered Escherichia coli from glycerol-glucose mixture

Achieving a balanced cell growth and biosynthesis of target products is a pivotal challenge confronted in the realm of green biomanufacturing of chemicals, and it holds the promise of significantly enhancing the titer/yield/productivity of the target products. The co-utilization of carbon sources has emerged as a proven approach for the precise regulation of cell growth and biosynthesis processes, thereby serving as a potent strategy to expedite the production of chemicals. In our previous study, we successfully demonstrated the efficient production of N-acetyl-glucosamine in engineered Escherichia coli by leveraging an appropriate catabolic division of labor, utilizing a mixture of glycerol and glucose as the carbon source. In this study, we further refined the division of labor between these two carbon sources by meticulously regulating the expression of the zwf gene, which encodes glucose-6-phosphate dehydrogenase and is crucial in diverting carbon source to the pentose phosphate pathway (PPP). After comparing three strategies for balancing cell growth and production, the engineered strain NAG-1 with zwf knocked out aided by the optimization of the glycerol-to-glucose ratio and the feeding mode of carbon sources resulted in robust production of N-acetyl-glucosamine. Remarkable production in a 5 L bioreactor was achieved, obtaining 249 g/L of N-acetyl-glucosamine with a yield of 0.684 g/g of total carbon sources (specifically, 0.791 g/g of glucose) and a productivity of 3.46 g/L/h. These results establish NAG-1 as the most efficient microbial cell factory reported thus far for the bioproduction of N-acetyl-glucosamine. The robust production of N-acetyl-glucosamine in E. coli using a mixture of glycerol and glucose suggests the immense potential of mixed carbon sources in the industrial green biomanufacturing of chemicals. © 2025 The Authors

Swarm-Based Dynamic Coverage of Multi-ASV Systems in the Presence of Measurement Noises

This paper is concerned with the dynamic coverage of multiple autonomous surface vehicles (ASVs) in the presence of static and dynamic obstacles, limited communication distance, and measurement noises. A modular control method including planning, guidance, and control is proposed. At the planning level, a swarm-based dynamic coverage strategy is developed to achieve the coverage of a given domain. Both the collision avoidance and connectivity preservation issues are solved by using artificial potential fields. At the kinematic level, novel adaptive line-of-sight guidance laws are presented based on cascade extended state observers (CESOs), such that accurate guidance signals are given by compensating for time-varying sideslip angle and suppressing measurement noises. At the kinetic level, two CESOs are developed to estimate the internal model uncertainties and external environmental disturbance, and antidisturbance kinetic control laws are designed subsequently. The proposed CESOs overcome the noises sensitivity problem of classical observers, and thus facilitate the practical implementation under measurement noises. The input-to-state stability of the closed-loop system is confirmed by the Lyapunov theory and cascade stability theory. Simulation and hardware-in-the-loop experiment results are provided to demonstrate the effectiveness of the proposed swarm-based dynamic coverage method. © 1967-2012 IEEE.

The special position of terahertz wave in the electromagnetic spectrum makes it possess the characteristics of orientation, broadband, penetration and low energy, which promotes the extensive research of terahertz wave in the fields of communication, radar, imaging,sensing, security inspection and so on. The solid-state terahertz sources based on semiconductor devices have attracted extensive attention in the field of terahertz information technology due to their characteristics such as being able to work at room temperature, being small in size, being easy to integrate and having good frequency stability. Terahertz planar Schottky diode is a kind of low parasitic semiconductor device. Its high cutoff frequency makes it work well in the terahertz range. The frequency multiplier based on planar Schottky diode is an important part of terahertz solid state source. In this review, the development of Schottky diodes technology in recent years have been introduced, including the structures and preparation of Schottky diodes. In addition, the current situation and performance of different types of terahertz sources based on Schottky diodes are further introduced,and the future development trend is discussed.

Bifidobacterium longum *** and Bifidobacterium adolescentis play important roles in the guts of infants and adolescents,*** this study,using a neonatal rat model,we compared the protective effects of these 2 bifidobacterial species against Salmonella *** results demonstrated that *** *** was more effective than *** in alleviating the severity of infection in newborn rats exposed to Salmonella enterica serovar Typhimurium strain *** *** attenuated intestinal inflammation and mucosal damage induced by Salmonella infection,as well as protecting intestinal nerves and intestinal barrier function through TLR4/My D88 *** *** also displayed the potential to modulate gut metabolites by promoting the biosynthesis of unsaturated fatty acids(arachidonic acid,eicosapentaenoic acid andα-linolenic acid)and purine metabolism(guanine,adenine,inosine and adenosine),thereby regulating metabolic ***,the benefits of *** *** were also observed in the liver,spleen and brain,improving nerve reflexes and suppressing ***,these findings provide novel insights into the prevention and treatment of gutrelated diseases in newborns,highlighting the potentially significant role of *** *** in clinical applications.

Drying processing is an indispensable procedure for the application of *** drying methods usually offer significant influence on the physicochemical properties of *** study investigated the effects of freeze and hot-air drying approaches on krill using lipidomics and *** analysis indicated the dramatic differences between freeze dried and hot-air dried krill.A total of 276 lipids were observed to be changed,including phosphatidylcholines(PC),triacylglycerols(TG)as well as fatty acids(FA)and their ***,several long-chain polyunsaturated phospholipids,such as PC(20:5_22:6),PC(20:5_20:5)and PE(16:0_22:6)displayed higher contents in hot-air dried krill,suggesting that method was beneficial to the high-quality krill oil *** the other hand,the levels of 13 proteins in hot-air dried krill were noted to be higher,whereas other 101 proteins were found to be higher in freeze dried *** results indicated that hot-air drying processing reserved more phospholipids in krill oil,but degraded lots of krill tissue *** freeze drying maintained more proteins in tissue,increasing the risk of deterioration for long-term *** together,this comprehensive analysis provided novel insights into the processing of krill to facilitate the utilization of krill resource.

An inadequate amino acid(AA)supply in animals under protein-restricted conditions can slow skeletal muscle *** translation can be activated by short-term leucine(Leu)stimulation;however,whether muscle mass increases under long-term Leu supplementation and how the gut and muscle respond to Leu supplementation are largely *** this study,we investigated if muscle mass increases with long-term Leu supplementation under protein-restricted *** identified changes in the link between the gut and muscles under different amino acid supply conditions,using goats as the study object.A total of 27 Xiangdong black male goats with average initial body weight(BW)of 10.88±1.22 kg were randomly divided into three dietary treatments:a normal protein diet(NP,14.24%crude protein[CP]);a low protein diet(LP,8.27%CP with supplemental 1.66%rumen-protected lysine[RPLys]and 0.09%rumen-protected methionine[RPMet]);and LP diet with rumen-protected Leu(RPLeu)(LP t RPLeu,8.75%CP with supplemental 1.66%RPLys,0.09%RPMet and 1.46%RPLeu).The animal trial lasted for 110 d,consisting of 20 d of adaptation and a 90 d of experimental *** results showed that long-term protein restriction increased gut tryptophan hydroxylase 1(TPH1)activity(P<0.001),tryptophan(Trp)catabolism(P<0.001),and 5-hydroxytryptamine(5-HT)synthesis(P<0.001),which all subsequently reduced goat ***-term Leu supplementation inhibited 5-HT synthesis(P<0.001),decreased Trp catabolism in the gut,and increased appetite in ***-term protein restriction enhanced jejunal and ileal branched-chain amino acid transferase(BCAT)(P<0.001)and branched-chain a-Keto acid dehydrogenase(BCKD)(P=0.048)activities,which increased branchedchain amino acid(BCAA)*** results showed that protein restriction decreased the intestinal mucosal expression of solute carrier family 1 member 5(SLC1A5)(P=0.032)and solute carrier family 7 member 5(SLC7A5)(P<0.001),reduced BCAA transport from the

The long-standing challenge in designing far-infrared transparent conductors(FIRTC)is the combination of high plasma absorption edge(λ_(p))and high conductivity(σ).These competing requirements are commonly met by tuning carrier concentration or/and effective carrier mass in a metal oxide/oxonate with low optical dielectric constant(ε_(opt)=2-7).However,despite the highσ,the transparent band is limited to mid-infrared(λ_(p)<5μm).In this paper,we break the trade-off between highσandλ_(p)by increasing the“so-called constant”ε_(opt)that has been neglected,and successfully develop the material family of FIRTC withε_(opt)>15 andλ_(p)>15μ*** FIRTC crystals are mainly octahedrally-coordinated heavy-metal chalcogenides and their solid solutions with shallow-level *** highε_(opt)relies on the formation of electron-deficiency multicenter bonds resulting in the great electron-polarization *** new FIRTC enables us to develop the first“continuous film”type far-infrared electromagnetic shielder that is unattainable using traditional ***,this study may inaugurate a new era in far-infrared optoelectronics.

Macleaya cordata extract(MCE)is a potential replacement for *** the current study,effects of MCE on the gastrointestinal health and humoral responses of host animals were explored.A total of 30 weanling goats with similar body weight of 9.15±1.36 kg were randomly allocated into three groups(n=10 per group):control group(CON group,fed with a basal diet),antibiotic group(Abx group,fed with the basal diet supplemented with 0.18 g/d vancomycin and 0.36 g/d neomycin),and MCE group(fed with the basal diet supplemented with 5 g/d MCE),for three *** showed that antibiotic addition decreased the height and area of rumen papillae,ruminal mucosa Toll-like receptor 8(TLR8),interleukin-8(IL-8)and interleukin-1β(IL-1β)gene relative expression levels and microbial diversity,altered the volatile fatty acid(VFA)profile in the rumen,and increased monocytes amount and CD4^(+)T cells percentage in the peripheral blood(P<0.05)compared to CON *** addition increased the average daily gain,ileal villus height,villus height/crypt depth,and immunoglobulin M(IgM)content in the peripheral blood(P<0.05)compared to the ***,MCE addition decreased the proportion of isobutyric acid in the chyme of the ileum(P=0.005)compared to the CON *** results suggest that antibiotic supplementation may suppress the epithelial state and microbial diversity and fermentation in goats,but stimulate cellular response to maintain the growth performance of *** administration improved the epithelial state and humoral response to promote the growth performance in goats.

Research on Cooperative Path Planning and Formation control for Multiple UAVs 1

Considering the background of Multi-UAVs formation cooperation, a Cooperative Multi-Swarm Particle Swarm Optimization (CMS-PSO) scheme as well as a prescribed performance control (PPC) method is introduced. Firstly, the cost function is developed and a novel CMS-PSO approach is applied to deal with three dimensional (3D) path planning problem. Then, the nonlinear mathematical formation model is given. So as to confine the location differences of the formation system, a PPC algorithm integrated with the backstepping methodology is constructed in details.
