The impact of COVID-19 pandemic in pregnant patients is an important emerging topic. Evidence of transplacental transmission and typical histopathological alterations in the placenta are controversial in the literature. This was a prospective observational cohort multicenter study in which we selected 23 placentas of 21 patients that showed detection of SARS-CoV-2 RNA by RT-PCR in the placenta tissue and described both morphological and clinical characteristics. Immunohistochemistry was performed to localize the virus in the specimens. Most of the patients were asymptomatic (61.9%) and preterm delivery was observed in 8 patients (34.7%). In relation to histopathological features, all the placentas showed evidence of maternal vascular malperfusion, as well as some degree of villitis with a high frequency of high grade placentitis (73.9%) and chronic and/or acute intervillositis (82.6%). We observed immunopositivity for SARS-CoV-2 antibody in 69.5% of the cases, notably in the syncytiotrophoblast. We reported histopathological features of placentas with viral detection in the tissue, thus providing evidence that SARS-CoV-2 can affect the placenta, although maternal and neonatal clinical outcome is usually mild.

The optimal vaginal microbiota is dominated by Lactobacillus spp., particularly Lactobacillus crispatus or Lactobacillus gasseri. Lactic acid production by Lactobacillus inhibits the growth of other bacterial species. Bacterial vaginosis (BV) is characterized by depletion of vaginal Lactobacillus and treatment with current choice antibiotics often fails urging novel alternatives including probiotics or prebiotics. Testing new components for BV treatment often utilize in vitro quantification of Lactobacillus. Thus, this study aimed to compare methods for L. crispatus and L. gasseri quantification. Type strains were cultured in Yeast extract/peptone/tryptone/Tween 80/glucose (LAPTg) and New York City III (NYCIII) broth media, and their glucose-restricted preparations (LAPTgr, NYCIIIgr). Bacterial growth was assessed by measurements of optical density at 540 nm (OD) and plate counting (PC) (CFU/mL) at 0 h, 8 h, 12 h, and 24 h. Measurements of bacterial growth at all timepoints were compared with ANOVA at 5 % significance level. Higher OD reads were observed in LAPTg (p < 0.05) and NYCIII (p < 0.05) with L. crispatus and L. gasseri when compared to LAPTgr and NYCIIIgr, respectively. However, PC did not differ between LAPTg and LAPTgr (p = 0.628 for L. crispatus; p = 0.161 for L. gasseri). In NYCIII, only L. crispatus exhibited higher PC in NYCIII when compared to NYCIIIgr (p = 0.016). These findings suggest that PC does not reflect Lactobacillus spp. counts under the tested conditions. Future studies exploring BV treatments should combine OD measurements with methods for Lactobacillus quantification other than PC, e. g. molecular methods.

The lack of an effective vaccine against all Plasmodium species, combined with the emergence and spread of resistance to traditional antimalarials, has, together with other factors, contributed to the failure of malaria eradication campaigns around the world. Therefore, the search for drugs with antiplasmodial activity is one of the priorities of the global scientific community and is strongly supported by the WHO. In this study, 6 synthetic compounds derived from 4-aminoquinolines were synthesized. In silico analyzes of their physicochemical and pharmacokinetic properties suggest that all compounds have good oral bioavailability, while they were not cytotoxic in vitro assays. Although compounds 7 and 9 showed the best IC values when evaluated in vitro against P. falciparum (chloroquine-resistant W2 strain), compound 9 showed the best effectiveness in vivo, with rates of inhibition of parasite multiplication varying from 72 to 50% between days 5 and 9 post-infection. Docking-based inverse virtual screening indicated that compound 9 exhibited potential interaction against 1O5X (Triose-phosphate Isomerase), 2OK8 (Ferredoxin-NADP(+) Reductase), 4C81 (2C-methyl-D-erythritol-2,4-cyclodiphosphate synthase (IspF)), and 4N0Z (glutaredoxin 1) targets, while UV-Vis analysis suggested action of compound in the digestive vacuole of the parasite. Together these results suggest that compound 9 is promising as an antimalarial molecule.

Since the beginning of the COVID-19 pandemic, catastrophic mortality and morbidity rates and unknown long-term ramifications were caused, leading to focused efforts on implementing effective vaccination. Nucleic acid-based vaccines (NBVs), an emergent advance in vaccine development, presented high efficacy rates but controversy regarding this approach soon spread. This study assesses healthcare workers (HCWs) knowledge and attitude towards NBVs’ effectiveness. This cross-sectional study was conducted in Jordan from November 2020 to January 2021 through a web-based questionnaire. 320 participants were split into educated and uneducated groups, with the former being exposed to educational material on NBVs before survey completion. The educated group expressed increased concern towards NBVs’ side effects. Specifically, the risk of developing cancer due to genetic modification(40.8%), fever(69.4%), skin rash(58.0%), and amyloid deposition(40.8%) showed a significant difference. The educated group preferred RNA-based NBVs over DNA-based ones(43.9%), had double the response rate, provided more scientifically accurate answers, and showed a more positive attitude towards the ability of NBVs to induce cellular immunity (57.3%) and prevent chronic forms of COVID-19(57.3%) (p < 0.05). Because HCWs are the most integral players in shaping public opinion about NBVs, thorough educational material on NBVs needs to be integrated within the curricula of health-related programs to bridge the gap present. This could prove vital in ensuring the success of the current and future vaccination campaigns.

Background The acquisition of reusable medical devices through loaner system is a worldwide phenomenon. Single-use implants, such as orthopaedic screws, that remain in the surgical tray are subjected to multiple handling and reprocessing until they are implanted. Exposure to physical, chemical and biological agents may compromise their quality/safety and favor biofilm formation. The aim of this study was to assess the surface integrity and microbiological conditions of patient-ready orthopaedic surgical implants (least used cortical screws), provided through loaner system.

IntroductionChronic liver disease is an important cause of morbidity and mortality among people living with human immunodeficiency virus (HIV) and is frequently related to non-alcoholic fatty liver disease (NAFLD). ObjectiveThe objective is to estimate the prevalence and risk factors of hepatic steatosis among consecutive patients with stable HIV infection on antiretroviral therapy (ART). Also, the use of transient elastography (TE) as a mean to identify a subgroup at risk for non-alcoholic steatohepatitis (NASH) and/or liver fibrosis. MethodsHIV infected patients were enrolled between August2016 and February2017. Inclusion criteria: ≥18 years with undetectable HIV viral load. Exclusion criteria: pregnancy; alcohol intake ≥20 g/day and co-infection B or C viruses. Patients underwent ultrasound (US) to diagnose liver steatosis. Significant fibrosis (≥F2) was estimated if at least one of the following were present: APRI > 1.0, FIB4 > 3 and/or liver stiffness ≥7.1kPa. Subjects with TE ≥ 7.1kPa were proposed a liver biopsy and NAFLD Scoring System (NAS) ≥ 3 was considered as diagnosis of NASH. ResultsA total of 98 patients were included. Liver steatosis was diagnosed in 31 patients (31.6%) and was independently associated with male gender, BMI, ALT and total bilirubin levels. The prevalence of significant fibrosis assessed by TE, APRI and FIB4 was 26.9%, 6.4% and 3.2%, respectively. Seven patients had a TE result ≥7.1kPa. NASH was found in 5 (83.3%). ConclusionAmong HIV infected patients undergoing ART, almost one third have NAFLD. Neither TE, APRI or FIB4 were able to act as surrogates for significant liver fibrosis. Nevertheless, TE ≥ 7.1kPa was able to accurately select a subgroup of patients at risk for NASH.

The aim of this study was to evaluate the influence of seasonality on the chemical composition of the essential oil from Hyptis dilatata leaves, to perform biological activity assays such as antimicrobial, inhibition of acetylcholinesterase enzyme and to evaluate the toxicity of the essential oil using Artemia salina as indicator on the test. Hyptis dilatata leaves were collected in rainy and dry seasons, in the morning, in the afternoon and in the evening. It was extracted by hydrodistillation using the extractor of Clevenger condenser double Spell model. The essential oil analysis resulted in 22 chemical components. The major constituents of the dry and rainy periods were a-pinene (18.7% in the night and 12.9% in the afternoon), 3-carene (26.5% and 19.9% in the night), fenchone (43% and 33.7% in the morning) and ß-caryophyllene 9.1% in the afternoon and 6.1% in the morning. The essential oil in vitro inhibited the acetylcholinesterase enzyme in 99.9% in the afternoon (rainy period) and 96.4% in the morning (dry period). Between the dry and rainy seasons, the lowest LC50 microbial activity in vitro was obtained for leaves collected during rainy season, in the morning period tested against the bacterium Staphylococcus aureus (LC50 78.1 mg mL-1). The cytotoxic activity of the essential oil of H. dilatata on Artemia salina showed LC50 results below 100 µg mL-1 and in the afternoon during the rainy period and at night in the dry period the results are above this value. Therefore, the chemical characterization and biological activities tests of essential oils showed promising results in the search for new active substances and the development of bioproducts of vegetable origin.
